<P>Inspired by the isothermal enzymatic process of rolling circle amplification (RCA) of DNA strands, we have developed a system to achieve more than a 200-fold increase in the synthesis of DNA nanostructures using a single-stranded circular DNA template. <P><B>Abstract</B></P><P>Transformation behavior of Ti–(45.0−<I>x</I>)Ni–5.0Cu–<I>x</I>Fe(at.%) (<I>x</I>=1.0–2.5) alloys were investigated by means of electrical resistivity measurements and X-ray diffraction. The one-stage B2-B19′ transformation occurred in a Ti–44.0Ni–5.0Cu–1.0Fe alloy, while the two-stage B2-B19-B19′ transformation occurred in a Ti–43.5Ni–5.0Cu–1.5Fe alloy. The three-stage B2-R-B19-B19′ transformation behavior was found in Ti–43.3Ni–5.0Cu–2.0Fe and Ti–43.0Ni–5.0Cu–2.5Fe alloys. The Fe content dependence of <I>T</I><SUB>R</SUB>(the B2-R transformation start temperature), <SUB><SUP>M′</SUP>s</SUB> (the R-B19 transformation start temperature) and <I>M</I><SUB>s</SUB> (the B19-B19′ transformation start temperature) were 100, 80 and 20K/at.%Fe, respectively.</P>
Gene Silencing by siRNA Microhydrogels via Polymeric Nanoscale Condensation

<P>The first attempt to prepare biologically active siRNA-based microhydrogels is reported. The self-assembled microhydrogels were fabricated using sense/antisense complementary hybridization between single-stranded linear and Y-shaped trimeric siRNAs. Reductively Dissociable siRNA‐Polymer Hybrid Nanogels for Efficient Targeted Gene Silencing

<P><B>Abstract</B></P><P>A highly efficient approach for target‐specific gene silencing based on a reductively dissociable nanogel incorporating small interfering RNA (siRNA) crosslinked with linear polyethylenimine (LPEI) via disulfide bonds is presented. Thiol‐terminated siRNA at both 3′‐ends is electrostatically complexed with thiol‐grafted LPEI. The prepared siRNA/LPEI complex contains inter‐ and intramolecular linkages, generating a mutually crosslinked siRNA/LPEI nanogel (MCN) that exhibits excellent structural stability against the addition of heparin but is readily disintegrated to biologically active, monomeric siRNA upon exposure to reductive conditions. Accordingly, the highly condensed, stable MCN shows greatly enhanced cellular uptake and gene silencing efficiency compared to the siRNA/LPEI complexes without crosslinks or with only LPEI‐mediated crosslinks.</P>
Isolation and characterization of Bacillus amyloliquefaciens from Allomyrina dichotoma larva feces

A bacterium was isolated from feces of Allomyrina dichotoma larva that feeds on well-rotted sawdust of woods. The isolate was identified as a Gram-positive and spore-forming stain. We named the isolate as Bacillus amyloliquefaciens on the basis of morphological and biochemical properties as well as 16S rRNA gene sequences. The culture of bacteria beneficially increased root and shoot growth of tomato, pepper and cucumber plants compared to the distilled water control. In addition, the bacterial culture strongly inhibited the mycelial growth of several fungal phytopathogens. The drop collapse assay with this culture showed a surfactant activity that is a major indicator for the selection of biocontrol agent. Also, a bacterium has ability of wastewater treatment. These data demonstrate the potential application of B. amyloliquefaciens as a biocontrol agent.

Background/Aims: Hemoglobin is the predominent pigment in the gastrointestinal mucosa, and the development of electronic endoscopy has made it possible to quantitatively measure the mucosal hemoglobin volume, by using a hemoglobin index (IHb). The aims of this study were to make a software program to calculate the IHb and then to investigate whether the mucosal IHb determined from the electronic endoscopic data is a useful marker for evaluating the color of intramucosal gastric carcinoma with regard to its value for discriminating between the histologic types. Methods: We made a software program for calculating the IHb in the endoscopic images. By using this program, the mean values of the IHb for the carcinoma (IHb-C) and those of the IHb for the surrounding non-cancerous mucosa (IHb-N) were calculated in 75 intestinal-type and 34 diffuse-type intramucosal gastric carcinomas. We then analyzed the ratio of IHb-C to the IHb-N (C/N ratio). Results: The C/N ratio in the intestinal-type carcinoma group was higher than that in the diffuse-type carcinoma group (p<0.001). In the diffuse-type carcinoma group, the C/N ratio in the body was lower than that in the antrum (p=0.022). The accuracy rate, sensitivity, specificity, and the positive and negative predictive values for the differential diagnosis of the diffuse-type carcinoma from the intestinal-type carcinoma were 94.5%, 94.1%, 94.7%, 88.9% and 97.3%, respectively. Conclusions: IHb is useful for making quantitative measurement of the endoscopic color in the intramucosal gastric carcinoma, and the C/N ratio by using the IHb would be helpful for distinguishing the diffuse-type carcinoma from the intestinal-type carcinoma.
