      • COMMUNITY PARTICIPATION IN AND BENEFITS OF TOURISM DEVELOPMENT IN THE TURPAN VALLEY, XINJIANG AUTONOMOUS REGION OF CHINA

        While western scholars such as Jamal and Gertz (1995), Tosun (2005), Fallon and Kriwoken (2003), usually presume that community participation is critical to sustainable tourism development, Chinese scholar Wenjun Li (2006) boldly states that it is possible for a community to achieve benefits of tourism development despite weak involvement in the planning and management of a tourist destination. Our survey of government officials, tourist business operators and residents in seven locations in the World Heritage-listed Turpan Valley in the Xinjiang province found that top-down, government-led participation is necessary and desirable, and does not detract from stakeholders' perception that they are sharing in the benefits of tourism development in the region.

        The contents and layout of the units on thermodynamics, which appear in seven kind of high school physics textbook, are compared. Characteristics and problems of each textbook are discussed, and suggestions are made on for the future textbook revision. As we analyzed each textbook, firstly, the other of content is edited differently in each textbook, so problems of curriculum, which is being carried out now, appeared. Secondly, the content of thermodynamics unit is edited differently in each textbook, and it is expressed by using macroscopic quantity rather than microscopic quantity. Thirdly, the number of experiments is differ in each textbook, so, it gives rises to confusion in managering curriculum. Fourthly, the edition of textoook, which is memorizing oriented, is the majority, rather than the content which promotescientific thinking and cause motivation.

        Induction of Apoptosis by Cordycepin via Reactive Oxygen Species Generation in Human Leukemia Cells

        Cordycepin (3’-deoxyadenosin), a polyadenylation specific inhibitor, is the main functional component in Cordyceps militaris which is one of the top three famous traditional Chinese medicine. It has been shown to possess many pharmacological activities including immunologically stimulating, anti-cancer, anti-bacterial, and anti-virus, anti-infection effects. However, its anti-cancer molecular mechanisms are poorly understood. In this study, the apoptotic effects by cordycepin were investigates in human leukemia cells. Treatment of cordycepin significantly inhibited cells growth in a concentrationdependent manner by inducing apoptosis, as evidenced by morphological change and apoptotic cell death such as formation of apoptotic bodies, DNA fragmentation and increased populations of sub-G1. Induction of apoptosis by cordycepin was associated with modulation of Bcl-2 and inhibitor of apoptosis proteins (IAP) family expression. Cordycepin also increased reactive oxygen species (ROS) generation, activation of casepase-3, caspase-8, caspase-9, cleavage of poly(ADP-ribose) polymerase (PARP), β-catenin and phospholipase C (PLC)-γ1 protein. The quenching of ROS generation by N-acetyl-L-cysteine administration, a scavenger of ROS, reversed the cordycepin-induced apoptosis effects. Theresults suggested that cordycepin may be a potential chemotherapeutic agent for the treatment of leukemia patients [This work was supported by Blue-Bio Industry RIC at Dong-Eui University as a RIC (08-06-07) program of ITEP under Ministry of Knowledge Economy].

        Tool Wear Performance in Face Milling Inconel 182 using Minimum Quantity Lubrication with Different Nozzle Positions

        Minimum quantity lubrication (MQL) is one of the promising Green Manufacturing Technology to meet the challenge of machining difficult-to-cut material Inconel 182 which is deposited by Shield Metal Arc Welding (SMAW). Unlike the flood lubrication, in MQL, only a small amount of non-toxic biodegradable oil are directly sprayed to the cutting zone by compressed air, which can reduce friction coefficient and temperature. Wear performance of uncoated tool inserts and Physical Vapor Deposition (PVD) coated tool inserts are evaluated by comparison during up and down face milling Inconel 182 with different MQL nozzle positions. Results indicate that uncoated tool inserts are not suitable for face milling Inconel 182 in all lubrication conditions due to the severe flank wear and the catastrophic breakage. On the contrary, PVD coated tool inserts have longer tool life compared to uncoated ones in all lubrication conditions. MQL nozzle positioned at tool cut into workpiece and positioned at tool cut out of workpiece in down milling as well as MQL nozzle positioned at tool cut out of workpiece in up milling can effectively prolong tool life of PVD coated tool inserts, which can be selected as the optimal lubrication solution for face milling Inconel 182.

      • Antitumor Activity of Histone Deacetylase Inhibitor Trichostatin A in Osteosarcoma Cells

        Background: Histone deacetylase (HDAC) inhibitors have been reported to induce cell growth arrest, apoptosis and differentiation of tumor cells. The present study aimed to examine the effects of trichostatin A (TSA), one such inhibitor, on the cell cycle, apoptosis and invasiveness of osteosarcoma cells. Methods: MG-63 cells were treated with TSA at various concentrations. Then, cell growth and apoptosis were determined by 3-(4, 5-dimethyl-2-thiazolyl)-2H-tetrazolium bromide (MTT) and TUNEL assays, respectively; cell cycling was assessed by flow cytometry; invasion assays were performed with the transwell Boyden Chamber system. Results: MTT assays revealed that TSA significantly inhibited the growth of MG-63 cells in a concentration and time dependent manner. TSA treated cells demonstrated morphological changes indicative of apoptosis and TUNEL assays revealed increased apoptosis of MG-63 cells after TSA treatment. Flow cytometry showed that TSA arrested the cell cycle in G1/G2 phase and annexin V positive apoptotic cells increased markedly. In addition, the invasiveness of MG-63 cells was inhibited by TSA in a concentration dependent manner. Conclusion: Our findings demonstrate that TSA inhibits the proliferation, induces apoptosis and inhibits invasiveness of osteosarcoma cells in vitro. HDAC inhibitors may thus have promise to become new therapeutic agents against osteosarcoma.

        Characterization of proteinase A excretion from Saccharomyces cerevisiae in high sugar stress conditions

        High sugar concentration culturing was commonly used in modern fermentation industry. However, it leads to the reduction of the foam stability in beer brewing due to the excess secretion of proteinase A. To better understand the characterization of proteinase A excretion from Saccharomyces cerevisiae in high sugar stress conditions, the cultures were grown by using YNB medium with a high concentration of glucose. Pro-PrA isolated from the medium was purified by gel exclusion chromatography, and the PrA activity was detected using fluorescent substrate analysis. The relative molecular weight of the purified PrA and pro-PrA was estimated at 42 and 54 kDa, respectively, by SDS-PAGE. It indicated that the metabolic behavior of PrA in the high glucose culturing was quite different from the normal conditions, and glucose concentration may have a big influence on its secreted process. Further study showed that PrA was released at the logarithmic growth phase of the culturing, and the amount of PrA was 11 times higher compared with the normal culturing. PrA was considered to be activated by itself under acidic conditions. And it was also confirmed in this work that the step-wise pathway for the autoactivation known as a pseudo-PrA has a major contribution to the autoactivation process of PrA zymogen outside the cell.

        Antioxidant Effects of Cheonggukjang Containing Phellinus linteus Extract

        This study was carried out to examine the antioxidant effects of cheonggukjang combined with Phellinus linteus extract. The electron-donating activity (EDA) of cheonggukjang containing 0.3% P. linteus extract (0.3% CPLE) was higher than that of cheonggukjang only. EDA of the ethanol extract from cheonggukjang was higher than that of the water extract. The water and the ethanol extracts showed strong antioxidant activity with regard to peroxide value. However, the ethanol extract showed a higher peroxide value than the water extract. The nitrite scavenging activity of the ethanol extract was greater than that of the water extract, corresponding to the EDA and peroxide values for each extract. Therefore, the antioxidant effects were enhanced by adding 0.3% of extract from P. linteus in manufacturing cheonggukjang. It is suggested that P. linteus extract could be put into practice as an effective antioxidant agent.

      • Effect of FeNi30 Powder Catalyst by Water Atomizing on Synthesis High-grade Diamond

        This paper described the preparation method for composing high-grade synthetic diamond by water atomizing using FeNi30 powder catalyst. The objective of this article is about powder making process using super high water atomizing in the atmosphere of inert gas, and then corroded the powder with a corrosion inhibitor. Finally, FeNi30 catalyst powder with lower oxygen content and good sphericity is produced. The experiment of making diamonds by using cubic press and the performance of the diamonds are also discussed.

        Bcl-2 Overexpression Attenuates Agaricus blazei Murill-Induced Apoptosis in U937 Cells by Inhibition of Caspase-3 Activit

        Agaricus blazeiMurill is an edible mushroom distributed in Brazil and presently cultivated in other areas, including Korea, Japan, and China. Its chemical components, including steroids and lectin and various polysaccharides have been widely studied. For this, we used U937/vector and U937/Bcl-2 cells, which were generated by transfection of the cDNA of the Bcl-2 gene. As compared with U937/vector, U937/Bcl-2 cells exhibited a 4-fold greater expression of Bcl-2. Treatment with 0.5 or 4 mg/ml A. blazei Murill for 24 h produced morphological features of apoptosis in U937/vector cells, respectively. This was associated with caspase-3 activation and PARP degradation. In contrast, A. blazei Murill-induced caspase-3 activation and PARP degradation and apoptosis were significantly inhibited by z-DEVD-fmk in U937 cells. Bcl-2 overexpressing cells exhibited sustained caspase-3 activation and expression levels of the Bcl-2 proteins during A. blazei Murill-induced apoptosis. In addition, these findings indicate that Bcl-2 inhibits A. blazei Murill-induced apoptosis by a mechanism that interferes with Bcl-2 degradation and activity of caspase-3 that is involved in the execution of apoptosi.

      • Comparison of the Gene Expression Profiles Between Smokers With and Without Lung Cancer Using RNA-Seq

        Lung cancer seriously threatens human health, so it is important to investigate gene expression changes in affected individuals in comparison with healthy people. Here we compared the gene expression profiles between smokers with and without lung cancer. We found that the majority of the expressed genes (threshold was set as 0.1 RPKM) were the same in the two samples, with a small portion of the remainder being unique to smokers with and without lung cancer. Expression distribution patterns showed that most of the genes in smokers with and without lung cancer are expressed at low or moderate levels. We also found that the expression levels of the genes in smokers with lung cancer were lower than in smokers without lung cancer in general. Then we detected 27 differentially expressed genes in smokers with versus without lung cancer, and these differentially expressed genes were foudn to be involved in diverse processes. Our study provided detail expression profiles and expression changes between smokers with and without lung cancer.
