        Complete mitochondrial genome of Capnia zijinshana (Plecoptera: Capniidae) and phylogenetic analysis among stoneflies

        The complete mitochondrial genome (mitogenome) of the winter stonefly, Capnia zijinshana (Plecoptera: Capniidae), was sequenced and compared with other stoneflies. The mitogenome of C. zijinshana is 16,310 bp long and contains 37 genes including 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes, and a control region (CR). The gene arrangement of C. zijinshana and other 12 stonefly mitogenomes is identical with the putative ancestral mitogenome in Drosophila yakuba. Twenty-one of the 22 tRNAs in the C. zijinshana mitogenome could be folded into the typical cloverleaf secondary structure, while the dihydrouridine (DHU) arm of trnS (AGN) was missing. The 1513-bp control region of C. zijinshana contained two potential stem-loop structures and a tandem repeat region. The phylogenetic relationships of 13 species from Plecoptera were analyzed based on the nucleotide sequences of 13 PCGs. The phylogenetic relationships among 13 stoneflies and two mayflies were inferred by Bayesian inference and maximum likelihood methods. C. zijinshana was grouped with another two winter stoneflies, Mesocapnia arizonensis and Apteroperla tikumana. The two analyses both supported the close relationship of Pteronarcyidae and Chloroperlidae, and also supported the sister-group relationship of Styloperlidae and Perlidae.

        Altered mRNA Levels of MOV10, A3G, and IFN-α in Patients with Chronic Hepatitis B

        To explore the relationship of the MOV10, A3G, and IFN-αmRNA levels with chronic hepatitis B virus (HBV) infection,Blood samples from 96 patients with chronic hepatitis B(CHB) and 21 healthy individuals as control were collected. HBV DNA load and aminotransferase in the serum weretested using real time PCR and velocity methods, respectively. The MOV10, A3G, and IFN-α mRNA levels in theperipheral blood mononuclear cells (PBMC) were examinedthrough qRT-PCR. The MOV10, A3G, and IFN-α mRNAlevels in CHB group was significantly lower than those inthe control group (P<0.01, P<0.05, P<0.01, respectively). TheA3G mRNA level in the high-HBV DNA load group waslower than that in the low-HBV DNA load group (P<0.05). However, no statistical difference was found in the MOV10and IFN-α mRNA levels between the two HBV DNA loadgroups. Furthermore, the MOV10 mRNA level showed positivecorrelation with IFN-α in the control group. These resultsindicated that the expression of the innate immune factorsMOV10, A3G, and IFN-α is affected by chronic HBV infection.

        Three partial mitochondrial genomes from Ips (Coleoptera: Curculionidae, Scolytinae) contribute to the phylogeny of Scolytinae

        Partial mitogenomes of three species from the genus Ips (Coleoptera; Curculionidae), Ips grandicollis, Ips sexdentatus and Ips typographus were sequenced and analyzed. The three mitogenomes are 11,553-bp with 28 genes, 13,135-bp with 30 genes, and 13,502-bp with 30 genes, respectively. Their gene arrangements are consistent with that of Drosophila yakuba. The three Ips species shared the conserved overlapping sequence motif “ATGATAG” between ATP8 and ATP6, using TTG as start codon for ND1 genes and TAG as stop codon for ATP8 genes. All tRNA genes could fold into typical cloverleaf structures except for tRNASer1. Three tRNASer1 genes lack the DHU arm and instead formed the D-loop. The phylogenetic analyses indicate that Scolytinae is paraphyly and belongs to Curculionidae. The Eurasian I. sexdentatus is sister group to the North American I. grandicollis and the Eurasian I. typographus. The phylogeny of Ips based on COI genes indicates that Ips is paraphyly, Ips perroti and Orthotomicus suturalis are sister groups and Ips spinifer is grouped with (Orthotomicus chaokhao + Orthotomicus erosus).
