        A Proteomic Analysis of Leaf Responses to Enhanced Ultraviolet-B Radiation in Two Rice (Oryza sativa L.) Cultivars Differing in UV Sensitivity

        To determine the proteomic response to UV irradiation, two cultivars, i.e., Lemont (UV tolerant) and Dular (UV sensitive), were exposed to natural and enhanced ultraviolet-B (UV-B) irradiation for 1, 7, and 14 days, and two-dimensional gel electrophoresis in combination with mass spectrometry (MS) and bioinformatics were used to compare the different proteomic responses in the leaves of the two cultivars. Thirty-nine proteins were up- or downregulated following the UV-B treatments. Among them, 30 increased or decreased more than 1.5-fold in abundance. They were further tested by using matrix-assisted laser desorption/ionization time of flight MS and performed a database search. Twentyfour proteins were thus identified. These identified proteins were mostly upregulated in Lemont, whereas only 14 of them upregulated in Dular. Nine proteins involved in glycometabolism and fatty acid metabolisms, signal transduction, and protein synthesis and folding in Dular were not changed. These results suggest that there was a complex regulative mechanism on the proteomes in rice leaves upon UV-B exposure.

        Study on Multi-Field Coupled Evolution Mechanism of Laser Irradiated 40Cr Steel Quenching Process Based on Phase Change Induced Plasticity

        40Cr is essential to steel for the manufacture of hardened gears, and optimizing its laser quenching process will be of greatsignificance for gear wear and life extension. However, the complete reliance on the experimental trial-and-error approachdoes not effectively reveal the mechanism of the evolution of the laser quenching process in disk laser. Numerical simulationprovides an effective way to obtain the transient evolution of the laser quenching process. In this paper, the temperaturechanges physical parameters of the quenched material are calculated based on the CALPHAD method. Establish a thermoelastic–plastic multi-field coupling model for the laser hardening process of the disk laser 40Cr gear steel. Perform thenumerical calculations on the transient temperature, martensite phase fraction, and the transformation-induced plasticitystress evolution of the 40Cr laser hardening process. Focus on the mechanism of the mutual coupling between phase changebehavior and plastic strain. Calculations show that there is heat accumulation during quenching, with a peak temperatureof 1448 K. Plastic stress has a “hump” distribution, with a peak value of 834 MPa, which is 1.25 mm from the center ofthe focal point. The experimental analyses of the 40Cr gear steel quenching microstructure and hardness distribution stateby the Axioskop2 scanning electron microscope, the KEYENCE VH-Z100R super depth of field 3D microscope, and theQ10M microhardness tester reveal the mechanical behavior of the material and the phase hardening law during the quenchingprocess. That provided an essential theoretical basis for effective prediction on quenching residual stress and optimizingindustrial production parameters.

        Xin Yu Lou: Unrepeatable Commercial Miracle in Chinese Retailing Industry

        In order to change the impression of businessman, Zhang Hongrui engaged in business by ethics. He set up Xin Yu Lou in 1984, with the representation of honesty and credibility. This article begins with Xin Yu Lou’s ethics: honesty in business and credibility in nature, then exhibits its unique management feature: enjoyable work and happy life. Based to special governance structure, Xin Yu Lou gets along well with all the stakeholders: Xin Yu Lou regards customer as friend, shows consideration to staff, becomes friend with counterpart and safeguards profit of suppliers. The purpose of this case study is to illustrate the miracle of Xin Yu Lou. The first is the position of human nature – unconditional return, no performance assessment, employee holiday in traditional festival, no heritage on stock which is linked with the post. The second is that desire is substituted by belief – credibility is the premise, and honesty is the beginning. The third is the orientation of target – transcending money, creating happiness, holy land in working, guiding development, all-round life fulfillment. Under the whole circumstance of low credibility in retailing industry, the big advantage of Xin Yu Lou depends on solving information asymmetry (such as guaranteed quality, trial commodity, unconditional return, and etc.), which can save transaction cost. The focus of this case analysis is win-win relations among all the stakeholders, including customers, staff, counterparts, suppliers, which is insisting on the principle of benefit sharing. Business longevity, staff happiness and successful life, customer satisfaction, growing up together with suppliers, joint development with business partners, consumer gains the benefit in the end as a result of win-win. The importance of study is to affirm that the most significant winner is the unit that has thought for others since the beginning in repeated game experiments.

      • Effects of different biochar amendments on carbon loss and leachate characterization from an agricultural soil

        <P><B>Abstract</B></P> <P>Selection of an appropriate biochar as a soil amendment requires a thorough investigation of the effects on soil ecosystems and adjacent water systems via leaching. Different biochar characteristics influence retention or leaching of different soil and biochar components. A lab lysimeter study was conducted to investigate carbon (C) balance and leachate quality with biochar additions. Biochar made from wood pellets (WP) and sewage sludge (SS) produced at 400 °C (WP400 and SS400) and 700 °C (WP700 and SS700), respectively, were applied to silt loam soil at an application rate of 4%. Fluorescence excitation-emission spectrophotometry (EEMs) was utilized to understand the compositional changes in leachate dissolved organic carbon (DOC). Our results show that DOC contributed the largest portion of C leaching loss. The WP treatments increased DOC mass loss, but did not significantly change leachate DOC quality. SS400, in comparison, increased mass loss of DOC and SS700 decreased it probably due to its higher adsorptive capacity to DOC. Unlike WP treatments, SS treatments significantly changed leachate DOC quality. Chemical oxygen demand (COD) was reduced with SS400 and SS700 biochar additions, which is assumed to be related to SS biochar's high oxygen-containing surface functional groups. Reduction in total nitrogen (TN) leaching by WP700 and SS700 treatments might be related to the higher micropore surface area. Over all, our findings imply that changes in the different components of the leachate from biochar-amended soil are related to different biochar properties, such as labile matter content, total surface area, micropore volume and cation exchange capacity.</P> <P><B>Highlights</B></P> <P> <UL> <LI> DOC contributed the largest portion of C leaching loss from biochar added soils. </LI> <LI> Leachate properties of biochar addition were related to biochar characteristics. </LI> <LI> Wood pellet biochar increased DOC leaching but did not influence DOC quality. </LI> <LI> Sewage sludge biochar (700 °C) reduced COD/TN leaching and retained humic substances. </LI> </UL> </P>

      • Imaging Anatomy of Waldeyer's Ring and PET/CT and MRI Findings of Oropharyngeal Non-Hodgkin's Lymphoma

        Background: This study was conducted to analyze positron emission tomography (PET) / computed tomography (CT) and magnetic resonance imaging (MRI) performance with oropharyngeal non-Hodgkin's lymphoma (ONHL).Materials and Methods: The complete image data of 30 ONHL cases were analyzed, all patients were performed PET / CT and MRI examination before the treatment, with the time interval of these two inspections not exceeding 14 days. The distribution, morphology, MRI signal characteristics, enhancement feature, standardized uptake value (SUV) max value and lymph node metastasis way of the lesions were analyzed. Results: Among the 30 cases, 23 cases were derived from the B-cell (76.7%), 5 cases were derived from the peripheral T cells (16.7%) and 2 cases were derived from the NK/T cells (6.7%). 19 cases exhibited the palatine tonsil involvement (63.3%). As for the lesion appearance, 10 cases appeared as mass, 8 cases were the diffused type and 12 cases were the mixed type. 25 cases exhibited the SUVmax value of PET / CT primary lesions as 11 or more (83.3%). MRI showed that all patients exhibited various degrees of parapharyngeal side-compressed narrowing, but MRI still exhibited the high-signal fat, and the oropharyngeal mucosa was intact. 25 cases were associated with the neck lymph node metastasis, among who 22 cases had no necrosis in the metastatic lymph nodes, while the rest 3 cases exhibited the central necrosis in the metastatic lymph nodes. Conclusions: PET / CT and MRI have important value in diagnosing and determining the lesion extent of ONHL.

      • β-elemene Induces Caspase-dependent Apoptosis in Human Glioma Cells in vitro through the Upregulation of Bax and Fas/FasL and Downregulation of Bcl-2

        Background: ${\beta}$-elemene, extracted from herb medicine Curcuma wenyujin has potent anti-tumor effects in various cancer cell lines. However, the activity of ${\beta}$-elemene against glioma cells remains unclear. In the present study, we assessed effects of ${\beta}$-elemene on human glioma cells and explored the underlying mechanism. Materials and Methods: Human glioma U87 cells were used. Cell proliferation was determined with MTT assay and colony formation assay to detect the effect of ${\beta}$-elemene at different doses and times. Fluorescence microscopy was used to observe cell apoptosis with Hoechst 33258 staining and change of glioma apoptosis and cell cycling were analyzed by flow cytometry. Real-time quantitative PCR and Western-blotting assay were performed to investigated the influence of ${\beta}$-elemene on expression levels of Fas/FasL, caspase-3, Bcl-2 and Bax. The experiment was divided into two groups: the blank control group and ${\beta}$-elemne treatment group. Results: With increase in the concentration of ${\beta}$-elemene, cytotoxic effects were enhanced in the glioma cell line and the concentration of inhibited cell viability ($IC_{50}$) was $48.5{\mu}g/mL$ for 24h. ${\beta}$-elemene could induce cell cycle arrest in the G0/G1 phase. With Hoechst 33258 staining, apoptotic nuclear morphological changes were observed. Activation of caspase-3,-8 and -9 was increased and the pro-apoptotic factors Fas/FasL and Bax were upregulated, while the anti-apoptotic Bcl-2 was downregulated after treatment with ${\beta}$-elemene at both mRNA and protein levels. Furthermore, proliferation and colony formation by U87 cells were inhibited by ${\beta}$-elemene in a time and does-dependent manner. Conclusions: Our results indicate that ${\beta}$-elemene inhibits growth and induces apoptosis of human glioma cells in vitro. The induction of apoptosis appears to be related with the upregulation of Fas/FasL and Bax, activation of caspase-3,-8 and -9 and downregulation of Bcl-2, which then trigger major apoptotic cascades.

        The Feasibility of Using Biomarkers Derived from Circulating Tumor DNA Sequencing as Predictive Classifiers in Patients with Small-Cell Lung Cancer

        Purpose To investigate the feasibility of biomarkers based on dynamic circulating tumor DNA (ctDNA) to classify small cell lung cancer (SCLC) into different subtypes. Materials and Methods Tumor and longitudinal plasma ctDNA samples were analyzed by next-generation sequencing of 1,021 genes. PyClone was used to infer the molecular tumor burden index (mTBI). Pre-treatment tumor tissues [T1] and serial plasma samples were collected (pre-treatment [B1], after two [B2], six [B3] cycles of chemotherapy and at progression [B4]). Results Overall concordance between T1 and B1 sequencing (n=30) was 66.5%, and 89.5% in the gene of <i>RB1</i>. A classification method was designed according to the changes of <i>RB1</i> mutation, named as subtype Ⅰ (both positive at B1 and B2), subtype Ⅱ (positive at B1 but negative at B2), and subtype Ⅲ (both negative at B1 and B2). The median progressive-free survival for subtype Ⅰ patients (4.5 months [95%CI: 2.6-5.8]) was inferior to subtype Ⅱ (not reached, p<0.0001) and subtype Ⅲ (10.8 months [95%CI: 6.0-14.4], p=0.002). The median overall survival for subtype Ⅰ patients (16.3 months [95%CI: 5.3-22.9]) was inferior to subtype Ⅱ (not reached, p=0.01) and subtype Ⅲ (not reached, p=0.02). Patients with a mTBI dropped to zero at B2 had longer median overall survival (not reached vs. 19.5 months, p=0.01). The changes of mTBI from B4 to B1 were sensitive to predict new metastases, with a sensitivity of 100% and a specificity of 85.7%. Conclusion Monitoring ctDNA based <i>RB1</i> mutation and mTBI provided a feasible tool to predict the prognosis of SCLC.

      • ppGalNAc T1 as a Potential Novel Marker for Human Bladder Cancer

        Objectives: To investigate the effect of glycopeptide-preferring polypeptide GalNAc transferase 1 (ppGalNAc T1 ) targeted RNA interference (RNAi) on the growth and migration of human bladder carcinoma EJ cells in vitro and in vivo. Methods: DNA microarray assays were performed to determine ppGalNAc Ts(ppGalNAc T1-9) expression in human bladder cancer and normal bladder tissues. We transfected the EJ bladder cancer cell line with well-designed ppGalNAc T1 siRNA. Boyden chamber and Wound healing assays were used to investigate changes of shppGalNAc T1-EJ cell migration. Proliferation of shppGalNAc T1-EJ cells in vitro was assessed using [3H]-thymidine incorporation assay and soft agar colony formation assays. Subcutaneous bladder tumors in BALB/c nude mice were induced by inoculation of shppGalNAc T1-EJ cells and after inoculation diameters of tumors were measured every 5 days to determine gross tumor volumes. Results: ppGalNAc T1 mRNA in bladder cancer tissues was 11.2-fold higher than in normal bladder tissues. When ppGalNAc T1 expression in EJ cells was knocked down through transfection by pSUPER-shppGalNAc T1 vector, markedly reduced incorporation of [3H]-thymidine into DNA of EJ cells was observed at all time points compared with the empty vector transfected control cells. However, ppGalNAc T1 knockdown did not significantly inhibited cell migration (only 12.3%). Silenced ppGalNAc T1 expression significantly inhibited subcutaneous tumor growth compared with the control groups injected with empty vector transfected control cells. At the end of observation course (40 days), the inhibitory rate of cancerous growth for ppGalNAc T1 knockdown was 52.5%. Conclusion: ppGalNAc T1 might be a potential novel marker for human bladder cancer. Although ppGalNAc T1 knockdown caused no remarkable change in cell migration, silenced expression significantly inhibited proliferation and tumor growth of the bladder cancer EJ cell line.

        Five-Year Follow-Up Results of a Randomized Controlled Trial Comparing Bipolar Plasmakinetic and Monopolar Transurethral Resection of the Prostate

        Purpose: To report the 5-year follow-up results of a randomized controlled trial comparing bipolar transurethral resection of the prostate (TURP) with standard monopolar TURP for the treatment of benign prostatic obstruction (BPO). Materials and Methods: A total of 220 patients were randomized to bipolar plasmakinetic TURP (PK-TURP) or monopolar TURP (M-TURP). Catheterization time was the primary endpoint of this study. Secondary outcomes included operation time, hospital stay, as well as decline in postoperative serum sodium and hemoglobin levels. All patients were assessed preoperatively and followed-up at 1, 6, 12, 24, 36, 48, and 60 months postoperatively. Parameters assessed included quality of life, transrectal ultrasound, serum prostate-specific antigen level, postvoid residual urine volume, maximum urinary flow rates (Qmax), and International Prostate Symptom Score. Patient baseline characteristics, perioperative data including complications, and postoperative outcomes were compared. Complication occurrence was graded according to the modified Clavien classification system. Results: PK-TURP was significantly superior to M-TURP in terms of operation time, intraoperative irrigation volume, resected tissue weight, decreases in hemoglobin and sodium, postoperative irrigation volume and time, catheterization time, and hospital stay. At 5 years postoperatively, efficacy was comparable between arms. No differences were detected in safety outcomes except that the clot retention rate was significantly greater after M-TURP. Conclusion: Our results indicate that PK-TURP is equally as effective in the treatment of BPO, but has a more favorable safety profile in comparison to M-TURP. The clinical efficacy of PK-TURP is long-lasting and comparable with M-TURP.
