        ATM-dependent chromatin remodeler Rsf-1 facilitates DNA damage checkpoints and homologous recombination repair.

        <P>As a member of imitation switch (ISWI) family in ATP-dependent chromatin remodeling factors, RSF complex consists of SNF2h ATPase and Rsf-1. Although it has been reported that SNF2h ATPase is recruited to DNA damage sites (DSBs) in a poly(ADP-ribosyl) polymerase 1 (PARP1)-dependent manner in DNA damage response (DDR), the function of Rsf-1 is still elusive. Here we show that Rsf-1 is recruited to DSBs confirmed by various cellular analyses. Moreover, the initial recruitment of Rsf-1 and SNF2h to DSBs shows faster kinetics than that of γH2AX after micro-irradiation. Signals of Rsf-1 and SNF2h are retained over 30 min after micro-irradiation, whereas γH2AX signals are gradually reduced at 10 min. In addition, Rsf-1 is accumulated at DSBs in ATM-dependent manner, and the putative pSQ motifs of Rsf-1 by ATM are required for its accumulation at DSBs. Furtheremore, depletion of Rsf-1 attenuates the activation of DNA damage checkpoint signals and cell survival upon DNA damage. Finally, we demonstrate that Rsf-1 promotes homologous recombination repair (HRR) by recruiting resection factors RPA32 and Rad51. Thus, these findings reveal a new function of chromatin remodeler Rsf-1 as a guard in DNA damage checkpoints and homologous recombination repair.</P>

      • 都市主婦의 MAP와 %Fat 調査分析

        The purpose of this study is to analyze a maximal aerobic power(MAP) and % fat of the urban female. The subjects are 73 urban females who were devided into three groups(27 young femeles, 35 middle females, and 11 old age females) I measured Margaria bench stepping, VO_2 max calculated by Margaria's estimation expression, and % fat was calculated by using Nagamine's estimation expresstion. Therefore, The results of this study is to be compared to the cross sectional analysis among three groups. The results are as follows : 1. 「MAP」 Young female groups MAP was 41.4±13.99㎖/㎏·min, middle age females 39.2±13.95㎖/㎏·min, and old age females, 38.2±10.4㎖/㎏·min. This data showed that 3 groups were in fitness. 2. 「% Fat」 %fat in young female groups % fat was 31.4±5.46%, middle age females, 32.5±8.09%, and old age females, 31.3±6.92%. According to the results, 3 groups have no statistical significance(P>.05). These data showed that three groups were over weight(obesity).

      • 우울증 환자 치료에 있어서 Tianeptine과 Sertraline의 효과와 안전성 비교

        연구목적: 주요 우울증의 치료에 있어서 티아넵틴과 써트랄린의 임상적 효과와 안정성을 비교하기 위하여 연구를 시행하였다. 방법: DSM-IV 진단 기준에 의거하여 주요 우울증의 진단기준에 부합되고 17항목 해밀턴 우울척도 점수가 14점 이상인 환자를 대상으로 하였다. 총 40명의 환자를 무작위적 방식에 따라 티아넵틴 치료군과 써트랄린 치료군으로 분류하였다. 선탠된 환자에서 다른 항우울제를 복용하고 있는 경우 7일 이상의 약물 배설기간을 가진 후 티아넵틴과 써트랄린을 각각 6주간 투여하였으며, 기준점, 1주후, 2주 후, 4주 후, 6주 후에 HAM-D, MADRS, 그리고 CGI, COVI 등을 사용하여 평가하였다. 본 연구기간 중 발생된 모든 부작용은 부작용의 발생 및 소실 시기, 심한 정도, 발생 빈도, 관련 조치 및 결과에 대하여 증례기록서에 기록하도록 하였다. 결과: 30명(티아넵틴군 15명 ; 써트랄린군 15명)이 6주간의 연구를 완결하였고 티아넵틴 치료군에서는 1일 37.5mg을 투여하였으며 써트랄린군에서는 1일 평균 64.0±22.5mg을 투여하였다. 본 연구 결과 HAM-D, MADRS, CGI 상에서 두 군 모두에서 치료 1주 후부터 유의한 감소를 나타내었으며 이는 2주 후, 4주 후, 6주 후에도 지속적인 감소를 보였고 두 군간에 항우울 효과의 차이는 없었다. 활력징후, 일반 혈액학, 생화학, 심전도 검사 등에서는 두 군 모두 유의한 변화는 없었다. 티아넵틴과 관련된 부작용으로 흔히 보고한 증상은 오심(33.3%), 복부 불쾌감(26.7%), 구강 건조(20.0%), 두통(13.3%)등 이었고 써트랄린 치료군에서는 구강 건조(53.3%), 두통(46.7%), 오심(33.3%), 그리고 식욕 부진(33.3%)등으로 티아넵틴군에서 보다 적은 발현빈도를 보였다. 결론: 티아넵틴은 우울증의 치료에 있어서 효과적이고, 내약성이 우수한, 안전한 항우울제이다. Objective: This study was designed to evaluate the efficacy and safety of tianeptine and sertraline in the treatment of patients with depression. Method: The study was done on the patients with major depression diagnosed by DSM-IV, who had aHamilton Rating Scale for Depression(HAM-D) score ≥ 14 on the first 17 items of the HAM-D. A total of 40 patients were randomly assigned to tianeptine group and sertraline group. Tianeptine and sertraline were prescribed to each group. 6 weeks of each medication was carried out after 7 days of drug excretion period. Evaluation using 17 item HAM-D, Montgomery and Åsberg Depression Rating Scale(MADRS), Clinical Global Impression Scale (CGI), and Covi Scale was done on the baseline and after 1 week, 2 weeks, 4 weeks, and 6 weeks. Regrding all side effects that had occurred during the period of our study such as their developed/disappeaed time, severitien, incidences, managements and results have been recorded. Results: A total of 30 patients(tianeptine group 15 ; sertraline froup 15) finished the 6 weeks of research. 37.5mg of the daily dose was regularly prescribed to the tianeptine group, the average amount of 64.0±22.5mg of the final daily dose was prescribed to the sertraline group. Total 17 item HAM-D scores, total points of MADRS and CGI showed significant decrease after 1 week in each treatment froup and continous decrease after 2, 4 and 6 weeks ; and no difference was found between tianeptine group and sertaline group in the antidepressant efficacy. Also there were no significant changes in vital sign, CBC, chemistry, and EKG in each treatment group. The common reported side effects of tianeptine were nausea(33.3%), epigastic distress(26.7%), dry mouth(20.0%), headache(13.3%) and those of sertraline were dry mouth (53.3%), headache(46.7%), nausea(33.3%), anorexia(33.3%). Conclusion: According to the results, tianeptine was effective in improvement of depressive symptoms and was well tolerated and safe in patients with depression.

      • Mucor mucedo C-7이 생산한 정제 응유효소의 성질(II) : 정제 응우효소의 pH 안정성의 성질과 특성

        These experiments were conducted to investigate the properties and characteristics on the pH stability of the purified milk clotting enzyme from Mucor mucede C-7. The results obtained were as follows: 1.The Dependance on pH which effect on milk clotting activity(MCA) of this enzyme was increased along with a decrease in pH of the substrate solution. 2.In pH 6.0 and 3.0 of enzyme solution, MCA was not inactivated by heat treatment for 60min at 35℃ but was inactivated over 50% in pH 6.0 and over 80% in pH 3.0 by heat treatment for 10min at 60℃ respectively, 3.In pH 6.0, Thermo-stability of MCA and PA was showed but in pH 3.0 was not showed. On the other hand, MCA was higher in pH 3.0 then in pH 6.0. 4.pH-stability of MCA was within pH 3.5-5.5 and pH-stability of PA was within pH 3.0-6.0.

      • Geometric effect of the hydrogel grid structure on in vitro formation of homogeneous MIN6 cell clusters.

        <P>A microstructure-based hydrogel was employed to study the relationship between spatial specificity and cellular behavior, including cell fate, proliferation, morphology, and insulin secretion in pancreatic β-cells. To effectively form homogeneous cell clusters in vitro, we made cell-containing hydrogel membrane constructs with an adapted grid structure based on a hexagonal micropattern. Homogeneous cell clusters (average diameter: 83.6 14.2 μm) of pancreatic insulinoma (MIN6) cells were spontaneously generated in the floating hydrogel membrane constructs, including a hexagonal grid structure (size of cavity: 100 μm, interval between cavities: 30 μm). Interestingly, 3D clustering of MIN6 cells mimicking the structure of pancreatic islets was coalesced into a merged aggregate attaching to each hexagonal cavity of the hydrogel grid structure. The fate and insulin secretion of homogeneous cell clusters in the hydrogel grid structure were also assessed. The results of these designable hydrogel-cell membrane constructs suggest that facultative in vitro β-cell proliferation and maintenance can be applied to biofunctional assessments.</P>

        주요우울증에 대한 벤라팍신의 효과 및 안전성

        연구목적 : 주요우울증 환자에서 벤라팍신의 치료 효과와 안정성을 조사하기 위해 다기관 개방연구를 시행하였다. 방 법 : DSM-IV 진단 기준에 의거하여 주요우울증으로 진단된 환자중 벤라팍신 투여에 동의한 환자를 대상으로 하였다. 선택된 환자에서 다른 항우울제를 복용하고 있는 경우 14일 이상의 약물 배설기간을 가진후 벤라팍신을 6주간 투여하였으며, 기준점, 1주, 2주, 4주, 6주 후에 HAM-D, MADRS, 그리고 CGI등을 사용하여 평가하였다. 본 연구기간중 발생된 모든 부작용은 부작용의 발생 및 소설 시기, 심한 정도, 발생 빈도, 벤라팍신과의 관련성, 관련 조치 및 결과에 대하여 증례기록서에 기록하도록 하였다. 결 과 : 총 141명을 대상으로 하였으며, 이중 94명(66.7%)이 6주간의 연구를 완결하였고, 47명(33.3%)이 중도탈락하였다. HAM-D총점수는 벤라팍신 투여 1주후부터 유의한 감소를 나타내었으며, 2주,4주, 6주후에도 지속적인 감소를 보였다. MADRS의 총점수도벤라팍신 투여 1주 후부터 유의한 호전을 나타내었으며, 2주, 4주, 6주 후에도 지속적인 감소를 보였다. CGI에서도 기준점, 1주, 2주, 4주, 6주 후 각각의 시점 사이에 유의한 호전이 보였다. 활력징후, 일반 혈액학, 생화학, 심전도 검사 등에서 유의한 변화는 없었다. 벤라팍신과 관련된 부작용으로 흔히 보고한 증상은 오심(10.6%), 소화불량(9.5%), 변비(8.5%), 현기증(8.5%)등 이었다. 결 론 : 다기관 개방연구를 통해 주요우울증 환자에 대한 벤라팍신의 투여는 우울증상의 호전에 효과적이었으며 안전하였다. Objective : A Multicenter open-label study was conducted to evaluate the clinical efficacy and safety of venlafaxine for the tretment in patients with major depression. Method : The study was done in patients with major depression diagnosed by DSM-IV who accepted venlafaxine medication. In cases of the patients taking other antidepressants, 6 weeks of venlafaxine medication was carried out after 14 days of drug excretion period and evaluation using HAM-D, MADRS, and CGI was done at baseline, and after 1, 2, 4, and 6 weeks. Regarding all side effects that had occurred during the period of our study such as their developed/disappeared time, severities, incidences, relationship with venlafaxine, managements and results have been putted into the records. Results : A total of 141 patients were enrolled. Among 94 of them finished the 6 weeks of research and 41 of them did not make it through the research. Total HAM-D scores showed significant decrease after 1 week of venlafzxine medication and continous decrease through the study period. Total scores of MADRS also showed significant improvement after 1 week and continuous decrease through the study period. Similarly, CGI showed significant improvement between baseline, 1, 2, 4, and 6 weeks. There were no significant changes in vital sign, CBC, chemistry, and EKG. The commonly reported side effects of venlafaxine were nausea(10.6%), indigestion(9.5%), constipation(8.5%), and dizziness(8.5%). Conclusion : According to the results, venlafaxine was effective and safe in the treatment of patients with major depression.

      • 제빵반죽의 물리적 특성에 미치는 물의 영향

        This study was carried out to investigate the effects of water on the physical characteristics of dough for making white bread The results of the farinogram, the extensogram, and the amylogram measured were as the follow 1. In the farinogram pattern, the water absorption of dough were SW 71.2%, CW 70.9%, and DW 70.5%. water absorption of dough in SW was higher than CW and DW 2. In the development time, both SW and CW showed stability of dough because SW and CW taken 6 minutes whereas DW taken 2-3 min. In the extensogram, the resistance of dough was DW 520 B.U, CW 580 B.U, and SW 560 B.U, and thus CW and SW show excellence in keeping CO_2 formed by dough fermentation since their R/E (Resistance/ Extensibility) ratio was higher than R/E of DW. 3. By the amylogram of analyzing α-amylase activity, the highest viscosity was SW 700 B.U, DW 510 B.U, and CW 525 B.U, which showed SW might have the highest effect in enzyme stalling and starch decomposition in the baking stage.

        Eucheuma spinosum으로부터 다양한 효모를 이용한 바이오에탄올 생산

        Ethanol fermentations were performed using separate hydrolysis and fermentation (SHF) processes with monosaccharides from pretreated seaweed, Eucheuma spinosum as the biomass. The pretreatment was carried out with 11% (w/v) seaweed slurry and 150 mM H₂SO₄ at 121℃ for 40 min. Enzyme hydrolysis after H₂SO₄ pretreatment was performed with Celluclast 1.5 L at 45℃ for 24 h. Five % active charcoal were added to hydrolysate to removed 5-hydroxy methylfurfural. Ethanol fermentation with 11% (w/v) seaweed hydrolysate was performed for 72~96 h using Kluyvermyces marxianus, Pichia stipits, Saccharomyces cervisiae and Candida tropicalis. Ethanol concentration was reached to 18 g/L by K. marxianus, 16 g/L by P. stipitis, 15 g/L by S. cerevisiae and 10 g/L by C. tropicalis, respectively. The ethanol yield from total monosugar was obtained 0.50 and ethanol productivity was obtained 0.38 g/L/h by K. marxianus.

        오존 처리에 의해 산소 작용기가 도입된 활성탄소의 세슘 흡착 특성

        세슘은 높은 수용성으로 인하여 인체에 쉽게 침투하여 암 또는 DNA의 변형을 유발하는 잠재적인 독성 오염물질이다. 본 연구에서는 활성탄소의 세슘 흡착 능력을 향상시키고자 오존 처리를 통하여 활성탄소 표면에 산소 작용기를 도입하였다. 오존 처리 시간의 증가에 따라 활성탄소 표면의 산소함량이 증가하였다. 이후 활성탄소와 세슘 사이의 정전기적 상호작용이 더욱 원활하게 이루어져 모든 시료의 세슘 이온 흡착 효율이 향상되었다. 특히 반응기 내부 오존 농도를 50000 ppm으로 하여 7 min 동안 오존 처리한 시료는 약 12%의 높은 산소 작용기 함량을 보이며 97.6%의 가장 높은 세슘 제거 효율을 보였다. 한편, 5 min 동안 처리된 시료는 7 min간 반응한 시료와 비교하여 0.3%의 근소한 세슘 제거율 차이를 보였으며, 이는 오존 기체의 반응 특성으로 인한 두 시료의 표면화학적 유사성에 기인한다. 그러나, 오존 처리된 활성탄소의 세슘 흡착 성능은 활성탄소의 비표면적 및 기공 구조도 중요하지만 표면에 도입된 산소 작용기 양이 주된 영향을 미치는 것으로 판단된다. Cesium is a potential toxic contaminant due to its high solubility, which allows it to easily penetrate the human body and potentially induce cancer or DNA mutations. In this study, oxygen functional groups were introduced on activated carbons (ACs) by ozone treatment to enhance the cesium adsorption capacity. As the ozone treatment time increased, the oxygen content on the ACs surface increased. Subsequently, the electrostatic interaction between ACs and cesium enhanced, resulting in higher cesium ion adsorption efficiency across all samples. In particular, the sample treated with ozone for 7 minutes at an internal ozone concentration of 50000 ppm had roughly 12% greater oxygen functional group content and the highest cesium removal effectiveness (97.6%). Meanwhile, samples treated for 5 minutes showed a 0.3% cesium removal rate difference compared to those treated for 7 minutes, which was caused by the surface chemical similarity of the two samples due to the reactive characteristics of ozone gas. However, the cesium adsorption performance of ozonated activated carbon seems to be mainly influenced by the amount of oxygen functional groups introduced to the surface, although the specific surface area and pore structure of the activated carbon are also important.

        Background: Cardiovascular disease (CVD) is a major clinical problem and the most common cause of morbidity and mortality in patients with chronic kidney disease (CKD). Coronary artery calcifi cation (CAC) is a potential mechanism that explains the association between renal function and cardiovascular mortality. We aimed to evaluate the association between renal function and CAC in apparently healthy Korean subjects. Methods: In 23,617 participants in the health screening program in Kangbuk Samsung Hospital health screening program from January 2010 to December 2011, estimated glomerular fi ltration rate (eGFR) was assessed by Cockcroft-Gault equation. Coronary artery calcium score (CACS) was measured by multi-detector computed tomography (MDCT). Subjects were divided into 3 groups according to the CKD Staging system with eGFR grade; stage 1: eGFR = 90 mL/min/1.73 m2, stage 2: eGFR 60-89 mL/ min/1.73 m2, stage 3: eGFR 30-59 mL/min/1.73 m2. Results: Mean age of the participants was 41.4 years, and mean eGFR was 103.6±21.7 mL/min/1.73 m2. Hypertension and diabetes were noted in 43.7% and 5.5% of the participants, respectively. eGFR showed weakly negative but signifi cant association with CACS in bivariate correlation analysis (r=-0.076, p<0.01). Mean CACS signifi cantly increased from CKD stage 1 to 3. The proportion of subjects who had CAC signifi cantly increased from CKD stage 1 to 3. Although odds ratio for CAC signifi cantly increased from stage 1 to 3 after adjustment for confounding factors, this signifi cance was inversed when age was included in the model. Age was the strongest factor that affects CAC. Conclusions: This study performed in young Korean subjects with relatively normal renal function, age was the mostly strongest effector on the association between eGFR and the degree of CAC.
