Solanum aethiopicum is used in ethnomedicine for the treatment of overweight, constipation and anaemia. This study evaluated the ameliorative effect of aqueous leaf extract of S. aethiopicum on phenylhydrazine-induced anaemia in rats. Acute toxicity was determined in male and female rats (n = 5/group/sex) by oral administration of single dose of up to 5000 mg/ kg of the S. aethiopicum extract. The experimental rats were randomly grouped into five (5) groups of 6 rats each. Group (i) served as normal control, group (ii) negative control, group (iii) standard drug-5 mg/kg ferrous sulphate, groups (iv) and (v), 200 and 400 mg/kg of S. aethiopicum extract respectively. Phenylhydrazine (PHZ) was administered intraperitoneally at the dose of 50 mg/kg body weight for two consecutive days to groups (ii–v). After 14 days, the rats were sacrificed; blood, liver and kidney were collected. The haematological, lipid profile, liver and kidney function parameters were determined and the histopathology of the liver and kidney were examined. In acute toxicity study, no signs of toxicity or death were recorded. The study shows an observable significant (P < 0.05) increase in packed cell volume, haemoglobin and red blood cell counts at 400 mg/kg S. aethiopicum extract in both the male and female rats when compared to other groups. Solanum aethiopicum extract at the dose of 400 mg/kg reduced aspartate aminotransferase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), urea, creatinine and chloride. The results of this study lent credence to the use of S. aethiopicum leaf as an anti-anaemic tonic with a wide margin of safety and hepato/reno-protective potentials.

Characterization of a Calcitonin Gene-Related Peptide Release Assay in Rat Isolated Distal Colon

The release of calcitonin gene-related peptide (CGRP) plays a key role gastrointestinal tract homeostasis. We aimed to investigate mechanisms that mediate CGRP release from the rat colon in vitro. Colon segments were stimulated and the amount of CGRP released was measured using an enzyme immunoassay. Capsaicin and low pH induced significant increases in CGRP release which was shown to be mediated by TRPV1 activation as demonstrated with the TRPV1 antagonists CTPC and capsazepine. The mast cell degranulator, compound 48/80 significantly increased CGRP release an effect that was blocked in the presence of the mast cell stabilizer, ketotifen and the selective Gi inhibitor benzalkonium chloride. The addition of a mixture of inflammatory mediators containing pro-inflammatory cytokines, 5HT, bradykinin and PGE2 showed no effect at neutral pH but at low pH a significant additive effect was observed. We conclude that CGRP release in the rat distal colon occurs in response to mast cell degranulation, inflammatory mediators, low pH and capsaicin and describe a role for TRPV1 receptors in mediating the response.

Assessment of genetic diversity of Bambara groundnut (Vigna subterranea (L.) Verdcourt) accessions from Nigeria using informative molecular markers has become imperative for their genetic improvement and conservation. Comparative analysis of 30 V. subterranea from different locations in Nigeria was investigated using Directed Amplified Minisatellite DNA (DAMD) and Start codon targeted (SCoT) markers. The DNA was extracted using CTAB method for amplification. Both markers resolved the accessions into eight major groups using dendrograms and six clusters by principal component analysis. Alleles of 25 and 53 were obtained with DAMD and SCoT, respectively. Mean alleles, gene diversity, and polymorphic information content detected with DAMD were 10.2000, 0.6950, and 0.6600, while SCoT yielded 16.200, 0.847, and 0.836, respectively. Polymorphic loci were 130 and 142 in DAMD and SCoT, respectively. Both markers produced high polymorphism of 60.00-80% and 40.33-96.67% in DAMD and SCoT, respectively. Effective alleles (Ne) in both markers (DAMD: 1.1828-1.5927; SCoT: 1.1830-1.6779) were high. The Nei’s value (H) ranged from 0.1959-0.3238 in DAMD and 0.1533- 0.3782 in SCoT. The Shannon’s information index (I) obtained from DAMD and SCoT were 0.3281-0.4635 and 0.1420-0.5557, respectively. Total gene diversity (Ht), diversity within population (Hs), coefficient of gene differentiation (Gst), and estimate of gene flow (Nm) from DAMD were 0.3304, 0.2673, 0.1907, and 2.1215, while SCoT had 0.3927, 0.3163, 0.1945, and 2.0713, respectively. Our study demonstrated that SCoT markers are more competent than DAMD and should be integrated in the exploration of genetic diversity and selection of unique accessions for improvement and utilization of V. subterranea.
Modeling Service Quality Improvement Priorities in Selected Hotels for Efficient Service Delivery

This paper develops an integrated performance model that is aimed at improving service quality and providing accurate indicators that guide hotel managers in improving service priorities that promote customer satisfaction and eliminate resource wastage. The study was guided by three objectives of determining the level of importance of selected service quality attributes the level of satisfaction with selected service quality attributes and establishing the key service quality attributes that aid managers focus their improvement efforts. Quantitative techniques were used based on a sample size of 238 respondents selected from 30 hotels labeled as Common wealth Heads Of Government Meeting hotels in 2007. The findings indicate that out of the 22 service quality items, 13 needed improvements. These were mainly to do with staff skills in handling customers’ requirements and the hotel owners’ inability to provide flexible services to customers which affected service quality. There is need for hotel owners to recruit personnel with skills in customer care in order to improve service quality.

Patient satisfaction with the Emergency Department(ED) in a hospital is related to the length of stay, and especially to the amount of waiting time for medical treatments. ED over-crowding decreases quality and efficiency, therefore affecting hospitals` profitability. This paper presents a forecasting and simulation model for resource management of the ED at Moses H. Cone Memorial Hospital. A linear regression forecasting model is proposed to predict the number of ED patient arrivals, and then a simulation model is provided to estimate the length of stay of ED patients, system throughput, and the utilization of resources such as triage nurses, patient beds, registered nurses, and medical doctors. The near future load level of each resource is presented using the proposed models.
