        Evaluation of the plant growth-promoting activity of Pseudomonas nitroreducens in Arabidopsis thaliana and Lactuca sativa

        <P>Plant growth-promoting rhizobacteria (PGPR) enhance plant development through various mechanisms; they improve the uptake of soil resources by plants to greatly promote plant growth. Here, we used Arabidopsis thaliana seedlings and Lactuca sativa to screen the growth enhancement activities of a purified PGPR, Pseudomonas nitroreducens strain IHB B 13561 (PnIHB). When cocultivated with PnIHB, both species of plants exhibited notably improved growth, particularly in regard to biomass. Quantitative reverse transcription polymerase chain reaction analysis indicated high expression levels of the nitrate transporter genes, especially NRT2.1, which plays a major role in the high-affinity nitrate transport system in roots. Moreover, enhanced activity of the cyclin-B1 promoter was observed when wild-type 'Columbia-0' Arabidopsis seedlings were exposed to PnIHB, whereas upregulation of cyclin-B also occurred in the inoculated lettuce seedlings. Overall, these results suggest that PnIHB improves A. thaliana and L. sativa growth via specific pathways involved in the promotion of cell development and enhancement of nitrate uptake.</P>

      • <i>Paenibacillus pabuli</i> strain P7S promotes plant growth and induces anthocyanin accumulation in <i>Arabidopsis thaliana</i>

        <P><B>Abstract</B></P> <P>In this study, a novel plant growth-promoting rhizobacteria (PGPR), the bacterial strain <I>Paenibacillus pabuli</I> P7S (<I>PP7S</I>), showed promising plant growth-promoting effects. Furthermore, it induced anthocyanin accumulation in <I>Arabidopsis</I>. When co-cultivated with <I>PP7S</I>, there was a significant increase in anthocyanin content and biomass of <I>Arabidopsis</I> seedlings compared with those of the control. The quantitative reverse transcription-polymerase chain reaction analysis revealed higher expression of many key genes regulating anthocyanin and flavonoid biosynthesis pathways in <I>PP7S</I>-treated seedlings when compared with that of the control. Furthermore, higher expression of pathogen-related genes and microbe-associated molecular pattern genes was also observed in response to <I>PP7S</I>, indicating that the PGPR triggered the induced systemic response (ISR) in <I>A. thaliana</I>. These results suggest that <I>PP7S</I> promotes plant growth in <I>A. thaliana</I> and increases anthocyanin biosynthesis by triggering specific ISRs in plant.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>PP7S</I> enhance growth promotion of <I>Arabidopsis thaliana</I> via cell growth promotion. </LI> <LI> <I>PP7S</I> upregulates anthocyanin biosynthesis genes expression, subsequently increase anthocyanin accumulation in <I>Arabidopsis</I>. </LI> <LI> <I>PP7S</I> trigger specific Induced Systemic Response in <I>Arabidopsis</I> plant. </LI> <LI> <I>PP7S</I> co-cultivated <I>Arabidopsis</I> plant showed better tolerance to oxidative stress cause by salt. </LI> </UL> </P>

        Control of morphology and Orientation of Electrochemically Grown ZnO Nanorods

        We report the direct electrochemical deposition of ZnO nanorods on an indium tin oxide substrate. Themorphology and orientation of the grown ZnO nanorods were investigated as functions of the currentdensity. It is likely that the concentrations of OH- and Zn2+ ions, which could be controlled by varying thecurrent density, determine the shape and alignment of the ZnO nanorods. The nanorods were tilted, hexagonal,and prismatic at a low current density (0.1 mA/cm2) and vertically aligned and obelisk-shaped at highcurrent densities (greater than 0.6 mA/cm2). By using the low and high current densities sequentially in atwo-step growth process, vertically aligned, hexagonal, and prismatic ZnO nanorods could be grownsuccessfully. The underlying mechanism responsible for the growth of the ZnO nanorods is also discussed.

      • A Phenome-Based Functional Analysis of Transcription Factors in the Cereal Head Blight Fungus, <i>Fusarium graminearum</i>

        <▼1><P><I>Fusarium graminearum</I> is an important plant pathogen that causes head blight of major cereal crops. The fungus produces mycotoxins that are harmful to animal and human. In this study, a systematic analysis of 17 phenotypes of the mutants in 657 <I>Fusarium graminearum</I> genes encoding putative transcription factors (TFs) resulted in a database of over 11,000 phenotypes (phenome). This database provides comprehensive insights into how this cereal pathogen of global significance regulates traits important for growth, development, stress response, pathogenesis, and toxin production and how transcriptional regulations of these traits are interconnected. In-depth analysis of TFs involved in sexual development revealed that mutations causing defects in perithecia development frequently affect multiple other phenotypes, and the TFs associated with sexual development tend to be highly conserved in the fungal kingdom. Besides providing many new insights into understanding the function of <I>F. graminearum</I> TFs, this mutant library and phenome will be a valuable resource for characterizing the gene expression network in this fungus and serve as a reference for studying how different fungi have evolved to control various cellular processes at the transcriptional level.</P></▼1><▼2><P><B>Author Summary</B></P><P>Large collections of mutant lines allow for identification of gene functions. Here we constructed a mutant library of 657 putative transcription factors (TFs) through homologous recombination in the head blight fungus, <I>Fusarium graminearum</I>, providing a resource for understanding gene regulation in fungus. By screening these mutants in 17 phenotypic categories, we constructed a dataset of over 11,000 phenotypes. This study provides new insight into understanding multiple phenotypes caused by single TF as well as regulation of gene expression at the transcription level in <I>F. graminearum</I>. Furthermore, our TF mutant library will be a valuable resource for fungal studies through the distribution of mutants and easy access to our phenotypic and genetic data.</P></▼2>

        다문화 청소년이 지각한 사회적 지지, 자아존중감, 삶의 만족도의 관계 : 자아존중감의 종단적 매개효과를 중심으로

        목적 본 연구는 다문화 청소년이 지각한 사회적 지지(부모지지, 친구지지, 교사지지), 자아존중감, 삶의 만족도 간의 종단적 인과관계 검증을 목적으로 실시되었다. 방법 이를 위하여 한국청소년정책연구원에서 구축한 다문화청소년패널조사(MAPS)의 4차년도(2014)부터 6차년도(2016)까지의 데이터를 활용하여 자기회귀교차지연모형으로 분석하였다. 결과 첫째, 세 시점에 걸쳐 사회적 지지(부모지지, 친구지지, 교사지지), 자아존중감, 삶의 만족도의 자기회귀계수를 측정한 결과, 이전 시점의 사회적 지지(부모지지, 친구지지, 교사지지), 자아존중감, 삶의 만족도가 각각 이후 시점에 유의한 영향을 미치고 있었다. 둘째, 이전 시점의 사회적 지지(부모지지, 친구지지, 교사지지)는 이후 시점의 자아존중감에 통계적으로 유의한 영향을 미치는 것으로 나타났다. 셋째, 이전 시점의 자아존중감은 이후 시점의 삶의 만족도에 통계적으로 유의한 영향을 미치는 것으로 나타났다. 넷째, 자아존중감은 사회적 지지(부모지지, 친구지지, 교사지지)와 삶의 만족도의 관계에서 종단적 매개효과가 있는 것으로 나타났다. 결론 첫째, 사회적 지지(부모지지, 친구지지, 교사지지), 자아존중감, 삶의 만족도는 특정한 시기에 경험하는 것이 아니라 이전 상태가 이후 상태에 지속적인 영향을 줄 수 있음을 확인한 것으로 이전 시점의 상태가 잘 형성되어야 함을 의미한다. 둘째, 사회적 지지(부모지지, 친구지지, 교사지지)가 자아존중감을 매개로 삶의 만족도에 중학교 시기 동안 지속적으로 유지되는 것을 볼 수 있다. Objectives The purpose of study is to find out the logitudinal relationship among social support (parent support, friend support, teacher support), self-esteem, and life satisfaction. Methods To do this, autoregressive cross-lagged model(ARCL) was adapted in our study. Three waves(2014~2016) of data from the Multi-cultural Adolescents Panel Survey (MAPS) were analyzed in this study. Results First, the previous social support (parent support, friend support, teacher support), self-esteem, and life satisfaction had significant effect on subsequent social support (parent support, friend support, teacher support), self- esteem, and life satisfaction respectively. Second, the previous social support (parent support, friend support, teacher support) had significant effect on the subsequent self-esteem. Third, the previous self-esteem had significant effect on the subsequent life satisfaction. Forth, Self-esteem had longitudinal mediating effect in the relationship between social support (parent support, friend support, teacher support) and life satisfaction. Conclusions First, social support(parent support, friend support, teacher support), self-esteem, and life satisfaction are not experienced at a particular time, but rather confirmed that the previous state can have a lasting effect on the subsequent state, which means that the state at the previous time should be well formed. Second, It can be seen that social support(parent support, friend support, and teacher support) continues to be maintained during middle school years in the satisfaction level of life through self-esteem..

      • Strut-and-tie model for shear capacity of corroded reinforced concrete columns

        An analytical model is developed in this paper to predict the shear capacity of reinforced concrete (RC) columns with corroded transverse reinforcements. The shear strength model for corroded RC columns is proposed based on modifying the existing strut-and-tie model, which considers the deformational compatibility between truss and arch mechanisms. The contributions to the shear strength from both truss and arch mechanisms are incorporated in the proposed model. The effects of corrosion level of transverse reinforcements are considered in the proposed model through the minimum residual cross-sectional area of transverse reinforcements and the reduction of concrete compressive strength for the cover area. The shear strengths calculated from the developed model are compared with the experimental results from Vu's study (2017), which consisted of RC columns with corroded transverse reinforcements showing shear failure under the cyclic loading. The comparison results indicate satisfactory correlations. Parametric studies are conducted based on the developed shear strength model to explore the effects of column axial loading, aspect ratios, transverse reinforcements and the corrosion levels in transverse reinforcements to the shear strength of RC columns with corroded transverse reinforcements.

        Characterization of two leaf rust‑resistant Aegilops tauschii accessions for the synthetic wheat development

        Aegilops tauschii (Ae. tauschii) is a diploid (2n = 2x = 14) wild grass species, which has been reported as the progenitor of hexaploid wheat (Triticum aestivum) with D-genome. In this study, 68 Ae. tauschii accessions with diverse geographical backgrounds were investigated for their resistance to infection by the leaf rust fungi Puccinia triticina. Two Ae. tauschii accessions that exhibited hyper-resistance to leaf rust at both seedling and adult stages were identified. Utilizing two susceptible Ae. tauschii ecotypes and keumkang, a common Korean wheat cultivar known to be susceptible to leaf rust, as the negative control, further investigations were conducted for understanding the mechanism underlying immunity to leaf rust disease of these two resistant accessions. Resistant accessions displayed the increased β-1,3-glucanase activity to prevent fungal penetration and the better peroxidase activity to cope with leaf rustinduced oxidative stress. Moreover, transcriptional analyses reveal the important role of the LRR receptor-like serine/ threonine-protein kinase FLS2 (lrr) to the disease resistance of the two ecotypes. Ae. tauschii is a remarkable genetic source, especially for abiotic and biotic stress resistance genes, as the plant is known for its wide-ranging geographical habitat and adaptability to different environments. This, combined with the fact that Ae. tauschii and wheat share a close evolutionary relationship, is indicative of the immense benefit of using Ae. tauschii as a material for improving the quality of synthetic wheat. Our aim was to identify and evaluate the strongest Ae. tauschii contenders for breeding leaf rust-resistant synthetic wheat.

        Preparation of Silver Core-Chitosan Shell Nanoparticles Using Catechol-Functionalized Chitosan and Antibacterial Studies

        In this paper, we report the preparation and stabilization of colloidal silver nanoparticle solution, with theassistance of chitosan dihydroxyphenyl acetamide (CDHPA), or oligochitosan dihydroxyphenyl acetamide (OCDHPA). The structure of the chitosan derivatives were characterized by nuclear magnetic resonance (1H NMR) spectroscopy. The morphology of the synthesized silver core-chitosan shell nanoparticles were observed by transmission electronmicroscopy (TEM) and X-ray diffraction (XRD) techniques, and showed a well-defined core-shell structure of polymer-coated silver nanoparticles (AgNPs). The core-shell NPs exhibited a strong antibacterial activity against E. coliand S. aureus, at a very low concentration of AgNPs (2.5 ppm). Our studies offer a new method for the preparationand protection of silver nanoparticles for antibacterial applications.
