<P>Sorting of mitochondrial inner membrane proteins is a complex process in which translocons and proteases function in a concerted way. Many inner membrane proteins insert into the membrane via the TIM23 translocon, and some are then further acted upon by the mitochondrial m-AAA protease, a molecular motor capable of dislocating proteins from the inner membrane. This raises the possibility that the threshold hydrophobicity for the retention of transmembrane segments in the inner membrane is different depending on whether they belong to membrane proteins that are m-AAA protease substrates or not. Here, using model transmembrane segments engineered into m-AAA protease-dependent proteins, we show that the threshold hydrophobicity for membrane retention measured in yeast cells in the absence of a functional m-AAA protease is markedly lower than that measured in its presence. Whether a given hydrophobic segment in a mitochondrial inner membrane protein will ultimately form a transmembrane helix may therefore depend on whether or not it will be exposed to the pulling force exerted by the m-AAA protease during biogenesis.</P>
Seismic design and assessment of steel-concrete frame structures with welded dissipative fuses

This research presents the design and numerical assessment of composite steel-concrete frame structures with welded dissipative fuses. The assessment has been carried out based on linear response spectrum, nonlinear static pushover and time history procedures. The analytical expressions which define the envelope of the nonlinear response of the dissipative fuses are first presented and calibrated against experimental results available in literature. The assessment is then carried out according to a design methodology proposed herein. Outcomes of the numerical assessment indicate that the use of welded dissipative fuses successfully limited damage within the replaceable parts. Furthermore, although structures with dissipative fuses present lower strength and, generally, lower displacement capacity, their displacement ductility and global dissipative performance are generally higher than conventional structures, especially when the structure with dissipative fuses presents a dissipative configuration adjusted to the bending moment distribution diagram calculated for the applied seismic action.

In recent years there has been increasing international interest about designing structures thatcost less to repair after they have been subjected to strong earthquakes. Considering this interest, aninnovative repairable fuse device has been developed for dissipative beam-to-column connections inmoment-resisting composite steel and concrete frames. The seismic performance of the device was assessedthrough an extensive experimental program comprising ten cyclic and two monotonic tests. These tests wereconducted on a single beam-to-column specimen with different fuse devices for each test. The devicesvaried in terms of the chosen geometric and mechanical parameters. The tests showed that the devices wereable to concentrate plasticity and to dissipate large amounts of energy through non-linear behavior.Numerical models were developed with Abaqus and simplified design models are also proposed.
Cyclic behaviour of beam-to-column welded

In this paper the results of an experimental program devoted to the assessment of the cyclic behaviour of full scale, European type, beam-column subassemblages with welded connections are presented. Six tests (five cyclic and one monotonic) have been carried out on three different series of specimens, encompassing a total of eighteen tests. The three specimen series have been designed with the aim of defining the effect of the column size on the connection behaviour, under different applied loading histories. The tests have evidenced the effect of the column size and panel zone design and of the applied loading history on the cyclic behaviour and failure modes of the connections.

The article describes the development of a novel dissipative bracing connection device (identified by the acronym DRBrC) for concentrically braced frames in steel and composite structures. The origins of the device trace back to the seminal work of Kelly, Skinner and Heine (1972), and, more directly related, to the PIN-INERD device, overcoming some of its limitations and greatly improving the replaceability characteristics. The connection device is composed of a rigid housing, connected to both the brace and the beam-column connection (or just the column), in which the axial force transfer is achieved by four-point bending of a dissipative pin. The experimental validation stages, presented in detail, consisted of a preliminary testing campaign, resulting in successive improvements of the original device design, followed by a systematic parametric testing campaign. That final campaign was devised to study the influence of the constituent materials (S235 and Stainless Steel, for the pin, and S355 and High Strength Steel, for the housing), of the geometry (four-point bending intermediate spans) and of the loading history (constant amplitude or increasing cyclic alternate). The main conclusions point to the most promising DRBrC device configurations, also presenting some suggestions in terms of the replaceability requirements.
Effects of Hydrocarbon Additions on Gas-liquid Mass Transfer Coefficients in Biphasic Bioreactors

The effects of aliphatic hydrocarbons (n-hexadecane and n-dodecane) on the volumetric oxygen mass transfer coefficient $(k_L\;a)$ were studied in flat alveolar airlift reactor and continuous stirred tank reactors (CSTRs). In the flat alveolar airlift reactor, high aeration rates (>2vvm) were required in order to obtain efficient organic-aqueous phase dispersion and reliable $k_La$ measurements. Addition of 1% (v/v) n-hexadecane or n-dodecane increased the $k_La$ 1.55- and 1.33-fold, respectively, compared to the control (superficial velocity: $25.8{\times}10^{-3}m/s$, sparger orifice diameter: 0.5 mm). Analysis of the gas-liquid interfacial area a and the liquid film mass transfer coefficient $k_L$ suggests that the observed $k_La$ increase was a function of the media's liquid film mass transfer. Addition of 1% (v/v) n-hexadecane or n-dodecane to analogous setups using CSTRs led to a $k_La$ increase by a factor of 1.68 and 1.36, respectively (superficial velocity: $2.1{\times}10^{-3}m/s$, stirring rate: 250 rpm). These results propose that low-concentration addition of oxygen-vectors to aerobic microbial cultures has additional benefit relative to incubation in purely aqueous media.
Experimental study on the cyclic behaviour of bolted end-plate joints

In this paper an experimental study is performed on end-plate type joints. The test arrangement represents a column-base joint of a steel frame. Altogether six specimens were tested, each of them subjected to cyclic loading. The specimens were carefully designed by performing detailed preliminary calculations so that they would present typical behaviour types of end-plate joints. On the basis of the experimentally established moment-rotation relationship, the cyclic characteristics of each specimen have been calculated and compared to one another. The results are evaluated, qualitative and quantitative conclusions are drawn.

Background: Cupping therapy is used to treat musculoskeletal conditions, including low back pain. Objectives: The study assessed the effects of dry cupping on pain and functional disability from persistent nonspecific low back pain. Methods: This was a randomized controlled trial, where participants were allocated to a cupping therapy (n = 19) or sham (n = 18) group, for five 10-minute sessions of cupping therapy, twice a week, to stimulate the acupoints related to low back pain (GV4, BL23, BL24, BL25, and BL30, BL40 and BL58) and emotional aspects (HT3 and ST36). All participants were assessed at baseline, post-treatment and follow up (a finalization period of four weeks) using a visual analogue scale (VAS) and the Oswestry Disability Index (ODI). Groups were compared using the analysis of covariance (ANCOVA) and the effect size was calculated using Cohen ̓s d. Results: The cupping therapy group presented a lower mean VAS when compared to the sham, at post-treatment (mean difference: –2.36; standard error [SE]: 0.58; p < 0.001; “large” effect size: –0.94) and follow up (mean difference: –1.71; SE: 0.81; p < 0.042; ‘large’ effect size: –0.83). The cupping therapy group presented a lower mean ODI when compared to the sham post-treatment (mean difference: –4.68; SE: 1.85; p: 0.017; ‘large’ effect size: –0.87), although in follow-up, there was no difference between the groups (mean difference: 4.16; SE: 2.97; p: 0.17; “medium” effect size: –0.70). Conclusion: Dry cupping was more effective in improving pain and functional disability in people with persistent nonspecific low back pain when compared to the sham.
