        Identification of highly pathogenic Beauveria bassiana strain against Pieris rapae larvae

        Seven different strains of Beauveria bassiana were used in a bioassay on Pieris rapae larvae. The results showed that an B. bassiana strain showed relatively high pathogenicity towards P. rapae larvae. The adjusted mortality rate was 92.86 %, and the infection rate was 85.71 % in 10 days post inoculation. Molecular identification was performed to identify the unknown strain. Internal Transcribed Spacer sequence analysis showed that the polymerase chain reaction amplicon length of the unknown strain of Beauveria sp. was 573 bp, and sequence similarity to the known B. bassiana sequences in the NCBI database was 99 %. The B. bassiana strain was named Bb01. The changes of proteins and PPO of P. rapae larvae infected by B. bassiana Bb01 strain at different times was determined. The activity of PPO increased in 1–6 d and decreased in 7 d again after inoculation. The B. bassiana invaded into the insect body affected the balance of the proteins and PPO.

        Lycium barbarum Polysaccharides Induce Apoptosis in Human Prostate Cancer Cells and Inhibits Prostate Cancer Growth in a Xenograft Mouse Model of Human Prostate Cancer

        Lycium barbarum polysaccharides (LBPs) are important functional constituents in red-colored fruits of L. barbarum (Guo Qi Zi, a well-known traditional Chinese medicinal plant commonly known as Goji berry or wolfberry). The influence of LBP on human prostate cancer cells was systematically investigated in vitro and in vivo. The in vitro effects of LBP on two cell lines (PC-3 and DU-145) were examined by using trypan blue exclusion staining, single-cell gel electrophoresis, flow cytometry, terminal dUTP nick-end labeling assay, and immunohistochemical assay (assessment of Bcl-2 and Bax expression). The in vivo effect of LBP on PC-3 cells was assessed in the nude mouse xenograft tumor model. The in vitro results showed that LBP can dose- and time-dependently inhibit the growth of both PC-3 and DU-145 cells. LBP caused the breakage of DNA strands of PC-3 and DU-145 cells; the tail frequency and tail length were significantly higher than that of control cells. LBP also markedly induced PC-3 and DU-145 cell apoptosis, with the highest apoptosis rates at 41.5% and 35.5%, respectively. The ratio of Bcl-2/Bax protein expression following LBP treatments decreased significantly with a dose–effect relationship, which suggested that LBP can regulate the expression of Bcl-2 and Bax to induce apoptosis of PC-3 and DU-145 cells. The in vivo experimental results indicate that LBP might significantly inhibit PC-3 tumor growth in nude mice. Both the tumor volume and weight of the LBP treatment group were significantly lower than those of the control group.

        A Position Sensorless Control of Switched Reluctance Motors Based on Phase Inductance Slope

        A new sensorless position estimation method for switched reluctance motor (SRM) drives is presented in this paper. This method uses the change of the slope of the phase inductance to detect the aligned position. Since the aligned positions for successive electrical cycle of each phase are apart by a fixed mechanical angle $45^{\circ}$ in the case of 12/8 SRM, the speed of the machine can be calculated to estimate the rotor position. Since no prior knowledge of motor parameters is required, the method is easy for implementation without adding any additional hardware or memory. In order to verify the validity of this technique, effects such as changes in the advanced angle and phase lacking faults are examined. In addition, an inductance threshold based sensorless starting scheme is also proposed. Experimental results demonstrate the validity of the proposed method.

      • Survivin Gene Functions and Relationships between Expression and Prognosis in Patients with Nasopharyngeal Carcinoma

        This study aimed to investigate the relationship between prognosis and protein and mRNA expression of an apoptotic inhibitor gene, survivin, in patients with nasopharyngeal carcinoma. Furthermore, functions of the survivin gene in the CNE2 nasopharyngeal carcinoma cell line were assessed. Immunohistochemistry and in situ hybridization were used in detecting the survivin protein and mRNA in 44 nasopharyngeal carcinoma specimens, and 30 chronic nasopharyngitis samples as controls. Survivin gene expression in CNE2 cell line was suppressed with an shRNA (short hairpin RNA). The positive ratios of expression for survivin protein and mRNA in nasopharyngeal carcinoma were 79.5% and 75.0% respectively, obviously higher than in the control group (p<0.01), and there is very good consistency between the two methods. The mean survival time of patients with higher survivin protein or mRNA expression was shorter than in patients with lower levelsv(p<0.01). Proliferation of the CNE2 cell line was distinctly inhibited by the shRNA. The results indicate that overexpression of the survivin gene plays an important role in onset and development of nasopharyngeal carcinoma, and it may be helpful for prognostic appraisal.

        Model Test and Numerical Simulation Study on Bearing Characteristics of Pervious Concrete Pile Composite Foundation

        When pervious concrete is used as pile material, both high strength and water permeability will be available. By comparing the strength and permeability of pervious concrete with different aggregate size (3 − 5 mm and 4 − 6 mm) and porosity (20%, 25%, 30% and 35%), the recommended aggregate size (3 − 5 mm) and porosity (30%) can be achieved. To investigate the installation effect and bearing characteristics of the pervious concrete pile composite foundation, the excess pore water pressure, settlement, pile-soil stress ratio and skin friction were recorded in the model test. Results show that, compared with the impervious pile, the pervious pile can significantly accelerate the dissipation of excess pore water pressure after pile installation, and in the loading test can shorten the consolidation time by 30.3%, increase the skin friction 0.12 kPa averagely, reduce the peak value of excess pore water pressure by 30.2%, and increase the characteristic value of bearing capacity by 43.2%. When the linear penalty method is applied, and the friction coefficient is 0.6, compared with the test, the deviation of simulated skin friction is the largest, being about 15%, so in general the model test results are reliable.

        Parameter optimization method for antimisalignment of inductive power transfer system based on genetic algorithm

        The mutual inductance parameters change from time to time. When conducting dynamic wireless power transfer using an inductive power transfer system, it results in larger fluctuation of output power. Therefore, a parameter optimization method is necessary to improve the stability of inductive power transfer system during dynamic misalignment. In this study, a nonlinear programing model with objective function of minimum voltage gain difference was established by taking S-LCC topology as an example. Genetic algorithm and nonlinear programming were combined to optimize the compensating parameters of the system and to realize minimum fluctuation of output voltage gain of the system within any given range of mutual inductance parameters. Optimization results show that output stability can be realized by adjusting the compensation capacitance in the primary side. The feasibility of the theory was verified through stimulation and test prototype. Test results show that when the mutual induction range is from 29.3 μH to 84.3 μH, the voltage gain of the system varies from 0.67 to 0.77. The fluctuation ratio of voltage gain is 6.7%, and the fluctuation ratio of voltage gain under the circumstance of resonance parameters is 40.2%.

      • Prognostic Significance of Overexpression of EZH2 and H3k27me3 Proteins in Gastric Cancer

        The enhancer of zeste homolog 2 (EZH2) methyl transferase and histone 3 lysine 27 (H3K27me3) protein can repress gene transcription, and their aberrant expression has been observed in various human cancers. This study determined their expression levels in gastric cancer tissues with reference to clinicopathological features and patient survival. We collected 117 gastric cancer and corresponding normal tissues for immunohistochemistry analysis. In gastric cancers, 82/117 (70.1%) were positive for EZH2 and 66/117 (56.4%) for H3K27me3 proteins in contrast to only 5.41% and 7.25% of normal gastric mucosa specimens, respectively. Kaplan-Meier survival data showed the average overall and disease-free survival of EZH2 high expression patients was 25.2 and 20.2 months, respectively, shorter than that with EZH2 low expression (40.5 and 35.9 months). The average overall survival and disease-free survival of high H3K27me3 expression patients was 23.4 and 17.4 months, shorter than without H3K27me3 expression (37.6 and 34.5 months). The average overall survival and disease-free survival of patients with both EZH2 and H3K27me3 expression was 18.8 and 12.9 months, respectively, shorter than that with either alone (34.7 and 31.2 months) or with low levels of both (43.9 and 39.9 months). Multivariate Cox regression analysis showed that H3K27me3 and EZH2 expression, tumor size differentiation and clinical stage were all independent prognostic factors for predicting patient survival. This study demonstrated that detection of both EZH2 and H3K27me3 proteins can predict poor survival of gastric cancer patients, superior to single protein detection. In addition, H3K27me3 and EZH2 protein expression could predict lymph node metastasis.

        Enhanced photocatalytic hydrogen evolution under visible light using noble metalfree ZnS NPs/Ni@Trimellitic acid porous microsphere heterojunction

        The construction of late-model non-noble metal catalysts with above average performance and stability isthe best choice to implement visible light decomposition of water for hydrogen production and solve the problem ofclean energy. Herein, novel ZnS nanoparticles (ZnS NPs) grown in situ on the surface of porous Ni@Trimellitic acid(Ni-TA) microspheres were successfully synthesized. The structure, optical properties, element composition and othersof ZnS/Ni-TA composites were systematically analyzed by experimental characterization. The experimental resultsshowed that pure ZnS showed very weak photocatalytic performance. However, the photocatalytic performance wasgreatly increased with the addition of Ni-TA. The yield of the best sample (3% ZnS/Ni-TA) reached 1,098 mol/g/h,about 12 times higher than that of ZnS. Among them, Ni-TA not only can be used as the main body of exotic metalnanoparticles, but also the porous channels can prevent the agglomeration of nanoparticles. The enhanced H2 yield ismainly attributed to the resulting tight interface contact and well-matched band position which are conducive to effectivecarrier separation; moreover, the electrons quickly diverted to the exposed edge of Ni-TA for reducing to producehydrogen. The combination of inorganic and new organic semiconductors provides an idea for hydrogen productionunder visible light.

        Molecular Biology and Omics : Cloning and Characterization of a Novel α-Amylase from a Fecal Microbial Metagenome

        To isolate novel and useful microbial enzymes from uncultured gastrointestinal microorganisms, a fecal microbial metagenomic library of the pygmy loris was constructed. The library was screened for amylolytic activity, and 8 of 50,000 recombinant clones showed amylolytic activity. Subcloning and sequence analysis of a positive clone led to the identification a novel gene (amyPL) coding for α-amylase. AmyPL was expressed in Escherichia coli BL21 (DE3) and the purified AmyPL was enzymatically characterized. This study is the first to report the molecular and biochemical characterization of a novel α-amylase from a gastrointestinal metagenomic library.
