Overexpression of TTRAP inhibits cell growth and induces apoptosis in osteosarcoma cells

( Cai Hong Zhou ),( Qi Shen ),( Jin Glun Xue ),( Chao Neng Ji ),( Jin Zhong Chen ) 생화학분자생물학회(구 한국생화학분자생물학회) 2013 BMB Reports Vol.46 No.2

TTRAP is a multi-functional protein that is involved in multiple aspects of cellular functions including cell proliferation, apoptosis and the repair of DNA damage. Here, we demonstrated that the lentivirus-mediated overexpression of TTRAP significantly inhibited cell growth and induced apoptosis in osteosarcoma cells. The ectopic TTRAP suppressed the growth and colony formation capacity of two osteosarcoma cell lines, U2OS and Saos-2. Cell apoptosis was induced in U2OS cells and the cell cycle was arrested at G2/M phase in Saos-2 cells. Exogenous expression of TTRAP in serum-starved U2OS and Saos-2 cells induced an increase in caspase-3/-7 activity and a decrease in cyclin B1 expression. In comparison with wild-type TTRAP, mutations in the 5`-tyrosyl-DNA phosphodiesterase activity of TTRAP, in particular TTRAPE152A, showed decreased inhibitory activity on cell growth. These results may aid in clarifying the physiological functions of TTRAP, especially its roles in the regulation of cell growth and tumorigenesis. [BMB Reports 2013; 46(2): 113-118].

DFT and TD-DFT study on structure and properties of organic dye sensitizer TA-St-CA

        Cai-Rong Zhang,Zi-Jiang Liu,Yu-Hong Chen,Hong-Shan Chen,You-Zhi Wu,WangJun Feng,Dao-Bin Wang 한국물리학회 2010 Current Applied Physics Vol.10 No.1

The geometry, electronic structure, polarizability and hyperpolarizability of organic dye sensitizer TA-St-CA, which contains a π-conjugated oligo-phenylenevinylene unit with an electron donor.acceptor moiety, was studied using density functional theory (DFT), and the electronic absorption spectrum was investigated via time-dependent DFT (TD-DFT) with several hybrid functionals. The calculated geometry indicates that the strong conjugated effects are formed in the dye. The TD-DFT results show that the hybrid functional PBE1PBE and MPW1PW91 are more suitable than B3LYP for calculating electronic absorption spectra. The features of electronic absorption spectra were assigned on account of the qualitative agreement between the experiment and the calculations. The absorption bands in visible and near-UV region are related to photoinduced electron transfer processes, and the diphenylaniline group is major chromophore that contributed to the sensitization, and the interfacial electron transfer are electron injection processes from the excited dyes to the semiconductor conduction band. Compared with the similar dye D5, the good performance of TA-St-CA in dye-sensitized solar cells may be resulted from the higher energy level of the lowest unoccupied molecular orbital and the larger oscillator strengths for the most excited states with intramolecular electron transfer character.

Energy and Security in the East China Sea

( Cai Peng Hong ) 한국국방연구원 2007 The Korean Journal of Defense Analysis Vol.19 No.3

The conflict-ridden East China Sea has been attracting attention recently. The two major actors, China and Japan, have conducted nine rounds of negotiations and the priority on the agenda is placed on the legal delimitation and joint development of oil and gas fields. Concrete progress is being made with an accident-preventive initiative to enhance the liaison mechanism between China and Japan. The other is likely a joint report on a joint development plan to be issued by fall 2007. However, security implications and problems exist, with some characteristics. The first one is the security structure in East Asia, which is closely related to the ongoing Cold War climate, which hinders China to engage in regional security activities. China, therefore, is confronting traditional threats as well as non-traditional threats. The second one is domestic politics, which has a strong influence on a country`s maritime security policy. It reflects reality clearly in Japan. The third one is the issue of Sea Lines of Communication (SLOCs), which is closely related to security issues. The last one is the lack of an adequate management regime in this region. China, Korea and Japan can cooperate in the field to contribute to maintaining maritime stability. The author`s argument is that although there are some positive signals between Japan and China trying to alleviate tensions, one cannot see very clearly that both sides would make great concessions from their persistent stands, but a naval clash between China and Japan in the East China Sea may not happen in the near future.

      • Survivin Gene Functions and Relationships between Expression and Prognosis in Patients with Nasopharyngeal Carcinoma

        Cai, Jun-Hong,Fu, Sheng-Miao,Tu, Zhi-Hua,Deng, Li-Qun,Liang, Zhu,Chen, Xin-Ping,Gong, Xuan-Ju,Wan, Li-Hua Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.6

This study aimed to investigate the relationship between prognosis and protein and mRNA expression of an apoptotic inhibitor gene, survivin, in patients with nasopharyngeal carcinoma. Furthermore, functions of the survivin gene in the CNE2 nasopharyngeal carcinoma cell line were assessed. Immunohistochemistry and in situ hybridization were used in detecting the survivin protein and mRNA in 44 nasopharyngeal carcinoma specimens, and 30 chronic nasopharyngitis samples as controls. Survivin gene expression in CNE2 cell line was suppressed with an shRNA (short hairpin RNA). The positive ratios of expression for survivin protein and mRNA in nasopharyngeal carcinoma were 79.5% and 75.0% respectively, obviously higher than in the control group (p<0.01), and there is very good consistency between the two methods. The mean survival time of patients with higher survivin protein or mRNA expression was shorter than in patients with lower levelsv(p<0.01). Proliferation of the CNE2 cell line was distinctly inhibited by the shRNA. The results indicate that overexpression of the survivin gene plays an important role in onset and development of nasopharyngeal carcinoma, and it may be helpful for prognostic appraisal.

      • HiF-1α siRNA and Cisplatin in Combination SuppressTumor Growth in a Nude Mice Model of Esophageal Squamous Cell Carcinoma

        Liao, Hong-Ying,Wang, Gui-Ping,Gu, Li-Jia,Huang, Shao-Hong,Chen, Xiu-Ling,Li, Yun,Cai, Song-Wang Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.2

Introduction: The esophagus squamous cell carcinoma (ESCC) is one of the most deadly malignances, and a current challenge is the development of effective therapeutic agents. Our present work addressed the effect of HIF-$1{\alpha}$ siRNA alone or in combination with cisplatin on the growth of ESCC in nude mice. Materials and Methods: Xenografts were established by inoculating ESCC TE-1 cells in nude mice, and transplanted tumors were treated with HIF-$1{\alpha}$ siRNA, cisplatin alone or together. Growth was assessed by measuring tumor volume. HIF-$1{\alpha}$ mRNA and protein expression were detected using RT-PCR and immunohistochemistry, respectively. Apoptosis of ESCC TE-1 cells was analyzed by flow cytometry. Results: In our nude mice model, HIF-$1{\alpha}$ siRNA effectively inhibited the growth of transplanted ESCC, downregulating HIF-$1{\alpha}$ mRNA and protein expression, and inducing ESCC TE-1 cell apoptosis. Notably when combinated with cisplatin, HIF-$1{\alpha}$ siRNA showed synergistic interaction in suppressing tumor growth. Furthermore, the proportion of apoptotic cells in HIF-$1{\alpha}$ siRNA plus cisplatin group was significantly higher than that in cisplatin or HIF-$1{\alpha}$ siRNA-treated groups (P<0.05). Conclusions: Down-regulated HIF-$1{\alpha}$ expression induced by siRNA could effectively suppress the growth of transplanted ESCC $in$ $vivo$. HIF-$1{\alpha}$ siRNA could enhance the cytotoxicity of cisplatin, which suggests that a combination of these two agents may have potential for therapy of advanced ESCC.

      • Berberine Hydrochloride Impact on Physiological Processes and Modulation of Twist Levels in Nasopharyngeal Carcinoma CNE-1 Cells

        Li, Cai-Hong,Wu, Dong-Fang,Ding, Hang,Zhao, Yang,Zhou, Ke-Yuan,Xu, De-Feng Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.4

Objective: The main purpose of this work was to investigate the effect of berberine hydrochloride (BH) on the proliferation, apoptosis, migration, and invasion of CNE-1 nasopharyngeal carcinoma cells. Our results shed light on the functional components of traditional Chinese herbs for potential use in modern medicine. Methods: The CNE-1 cell line was treated with different concentrations of BH and effects on cell viability and proliferation were evaluated using the Cell Counting Kit-8 (CCK-8) assay. Anti-migratory and anti-invasive actions of BH were investigated using wound healing assays and the Millicell Hanging cell culture insert system, respectively. Expression of the epithelial-mesenchymal transition (EMT)-related gene twist (Twist) was analyzed by real-time PCR and Western blotting. Apoptosis was estimated with an annexin-V fluorescein (FITC) apoptosis detection kit, as well as with reference to levels of activated caspase-3 of CNE-1 cells before and after treatment with BH utilizing fluorescence spectroscopy. Results: BH was capable of reducing proliferation and viability of CNE-1 cells in a dose- and time-dependent manner, also demonstrating anti-migratory and anti-invasive capacities which correlated with reduction in expression of Twist. Finally, BH was able to induce significant amounts of apoptosis in CNE-1 cells, as demonstrated by an increase in the activity of caspase-3 and in annexin-V staining following treatment. Conclusion: BH extracted from rhizoma coptidis demonstrated an ability to block proliferation, induce apoptosis, and impair the migration and invasion of the CNE-1 cell line Considering these properties, our results suggest that BH could be an important compound for consideration in the treatment of nasopharyngeal carcinoma.

      • MicroRNA-451 Inhibits Growth of Human Colorectal Carcinoma Cells via Downregulation of Pi3k/Akt Pathway

Li, Hong-Yan,Zhang, Yan,Cai, Jian-Hui,Bian, Hong-Lei Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.6

MicroRNAs (MiRNAs) play important roles in coordinating a variety of cellular processes and abnormal expression has been linked to the occurrence of several cancers. The miRNA miR-451 is downregulated in colorectal carcinoma (CRC) cells, suggested by several research groups including our own. In this study, synthetic miR-451 mimics were transfected into the SW620 human CRC cell line using Lipofectamine 2000 and expression of miR-451 was analyzed by real time PCR, while expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2 was analyzed by Western blot, and cell growth was detected by MTT assay. In comparison to the controls, a significant increase in the expression of miR-451 was associated with significantly decreased expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2. The capacity of cell proliferation was significantly decreased by miR-451 expression, which also inhibited cell growth. Our study confirmed that miR-451 has a repressive role in CRC cells by inhibiting cell growth through down-regulating the P13K/AKT pathway.

Laser Joining of Ti3Al-Based Alloy to Ni-Based Superalloy using a Titanium Interlayer

        Xiao-Long Cai,Da-Qian Sun,Hong-Mei Li,Hong-Ling Guo,Yan Zhang,Ying-ying Che 한국정밀공학회 2018 International Journal of Precision Engineering and Vol.19 No.8

Joining Ti3Al-based alloy to Ni-based superalloy is of great interest for applications in the aerospace fields. Direct welding of these two materials was very difficult and the joint usually fractured during laser welding which cannot form an effective welding joint. In this work, a pure titanium interlayer with a thickness of 0.4 mm was used between the base metals. The results indicated that the addition of titanium interlayer has a great effect on the joint performance. No macrocracks were found through the whole joint. The average room-temperature tensile strength of the joint with titanium was 177 MPa which still much lower than the two base metals. The weak link of the dissimilar joint was the Ti3Al/weld interface. The presence of Ti2Ni, α2-Ti3Al and AlNi2Ti brittle intermetallic compounds in the Ti3Al/weld interface deteriorated the joint properties.
