Detection of Cleavage Sites on 5S rRNA by Methidiumpropyl-EDTA-Iron(II)

The affinity cleavage reagent Methidiumpropyl-EDTA-Iron(II) is applied to the structural analysis of 5S rRNA. Analysis of cleavage sites induced by MPE-Fe(II) on 5S rRNA shows that MPE intercalates easily between the unstable base pairs or into the bulges, thereby it strongly cuts the nucleosides nearby. The stable helical stems A, B, D and E as well as loop d are weakly cut. Most of the single-stranded loops are not cleaved. Based on the cleavage pattern of the 5S rRNA by MPE-Fe(II) and RNase V1, we suggest that MPE-Fe(II) may be used as a potential chemical probe in searching for the unstable helical regions of RNA, and for the sequences that appear to be involved in folding and distorting 5S rRNA.

두개강내의 뇌동정맥 기형에 대한 새로운 외과적인 수술 수기와 치료 방법은 다양하게 개발 ㆍ 발전되어 왔다. 저자들은 1990년 1월부터 1995년 12월까지 서울백병원 신경외과에서 외과적 수술후 평균 43.9개월 동안 추적, 관찰한 뇌동정맥 기형 환자 32명을 임상적 측면을 검토하고 수술후의 상태와 비교하여 외과적 측면에서 수술에 대한 예후 인자를 분석하였다. 가장 흔한 증상은 두개강내 출혈(52%)이었고, 그 중 뇌실질내 출혈의 빈도가 가장 높았다. 위치는 천막상부의 전두엽 부위가 가장 흔하였다. 낮은 Spetzler-Martin 등급, 작은 크기의 뇌동정맥 기형 종괴, 적은 수의 영양 동맥과 유출 정맥의 양상을 보일 때 외과적 수술 결과는 양호하였다. 영양 동맥이나 유출 정맥의 수가 많거나 크기가 큰 뇌동정맥 기형은 수술적으로 완전 제거는 어려웠으나, 일단 수술적으로 완전 제거후의 이환율은 작은 크기의 뇌동정맥 기형과 크게 다르지는 않았다. 상기의 관찰에서 볼 때, 뇌동정맥 기형의 치료는 외과적인 완전 제거가 주를 이루며 높은 Spetzler-Martin 등급(IV,V)의 뇌동정맥기형은 외과적 수술이외에도 단계적인 외과적인 수술과 함께 뇌정위적 방사선 치료나 혈관 색전술과 같은 다른 치료전략이 세워져야 할 것이다. Development of new surgical techniques and treatment modalities for intracranial arteriovenous malformations(AVM) has offered various methods of treatment available. The authors have followed up for 43.9 months and have reviewed clinical and surgical prognostic factors for 32 AVM patients, surgically managed from January 1990 through December 1995. The most common presenting symptom was hemorrhage(52%), of which the presenting hemorrhagic event, intracerebral hemorrhage was the most frequent. Supratentorial frontal type was the most common intracranial AVMs location. Our result shows that a good surgical outcome is anticipated under the following circumstances : low Spetzler-Martin grade, small-sized nidus and few feeding arteries. Many numbers of feeding arteries, and of draining veins and of large-sized AVM are very difficult to remove totally. After successful removal of large AVMs, postoperative morbidity are not different from small AVM. On the basis of these observations, we conclude that the treatment of AVM is principally total removal by surgical method. AVMs of the high Spetzler-Martin grade(IV, V) needs another treatment modalities such as a radiosurgery or a embolization wish or without a stage operation.

(η6-mesitylene) manganese (Ⅰ) tricarbonyl hexafluorophosphate[MTH-Mn (Ⅰ)]과 황산 이메틸, 피로탄산 이에틸, 과망간산 칼륨 따위 화학탐침들을 사용하여 Pseudomonas alcaligenes 5S rRNA의 고차원 구조를 분석하였다. 5S rRNA의 삼차구조에서 MTH-Mn (Ⅰ)이 강하게 절단하는 자리들은 a고리의 $G_{12}AUGG_{16}$, b-C 구역의 3'쪽 가닥, 즉$G_{51}AAGUGAAGC_{60}$, B-a구역의 $U_{65}-AGCG_{69}$, d고리의 5'쪽 가닥의 $G_{72}AUGG_{76}$ 연속부분 들이다. MTH-Mn(Ⅰ)과 그밖의 화학 탐침들을 사용하여 얻은 절단 양식들에서 우리는, MTH-Mn(Ⅰ)으로 강하게 절단되는 연속부분들이 $tRNA^{Phe}$의 L자 구조의 모서리 부분에서와 같은 주머니 구조를 이룰 것이며, 이러한 구조를 형성할 때 b-C구역과 d고리가 돌쩌귀 구실을 하는 것으로 추정한다. The higher order structure of Pseudomonas alcaligenes 5S rRNA has been investigated by using ($\eta^{6}$-mesitylene) manganese (Ⅰ) tricarbonyl hexafluorophosphate[MTH-Mn (Ⅰ)], dimethylpyrocarbonate, potassium permanganate as chemical probes. The sequences cleaved strongly by MTH-Mn (Ⅰ) on the tertiary structure of the 5S rRNA are $G_{12}AUGG_{16}$ of loop a, $G_{51}AAGUGAAGC_{60}$ of the region b-C, $U_{65}-AGCG_{69}$. of the region B-a, and $G_{72}AUGG_{76}$ of loop d. Based on such cleavage patterns of 5S rRNA by MTH-Mn(Ⅰ) and other chemical probes, we presume that the sequences strongly cleaved form pocket-like structure as in the the corner of L structure of $tRNA^{Phe}$. We also presume that the region b-C and loop d together play a role of hinge in forming the pocket-like structure in the 5S rRNA.

Seasonal variation of phytoplankton community was investigated at inner and outer stations around Jeju harbor from March to December 2000 in terms of standing crops and chlorophyll a. Among a total of 171 taxa of Phytoplankton identified, diatoms occupied 67.8%. dinoflagellates 15.2% and phytoflagellates 17.0% in species composition. Two diatoms were seasonally most dominant, that is, Chaetocoros socialis was predominant in March and October. and Skeletonema costatum in May. October and December. Mean standing crops were higher in inner stations with 5.1×106 cells 1-1 than in outer ones with 3.9×104 cells 1-1. Chlorophyll a concentration was also three times higher in inner stations than outer ones. Water temperature and salinity exhibited little difference seasonally in vertical distribution. but they were different between inner and outer stations. Higher water temperature and lower salinity prevailed in the inner station throughout the season. It suggests that the inner harbor has no good circulation and is affected directly by freshwater inflow from the neighboring stream. That accelerates the eutrophication of the inner harbor and thus sometimes causes high concentrations of Phytoplankton in a small red-tide scale.
