Development of a Simple Cell Lysis Method for Recombinant DNA Using Bacteriophage Lambda Lysis Genes

In this study, we describe the development of a simple and efficient method for cell lysis via the insertion of a bacteriophage lambda lysis gene cluster into the pET22b expression vector in the following order; the T7 promoter, a gene for a target protein intended for production, Sam7 and R. This insertion of R and Sam7 into pET22b exerted no detrimental effects on cellular growth or the production of a target protein. The induction of the T7 promoter did not in itself result in the autolysis of cells in culture but the harvested cells were readily broken by freezing and thawing. We compared the efficiency of the cell lysis technique by freezing and thawing to that observed with sonication, and determined that both methods completely disintegrated the cells and released proteins into the solution. With our modification of pET22b, the lysis of cells became quite simple, efficient, and reliable. This strategy may prove useful for a broad variety of applications, particularly in experiments requiring extensive cell breakage, including library screening and culture condition exploration, in addition to protein purification.
An adaptive remote display scheme to deliver mobile cloud services

<P>Often referred to as a supercomputer in the palm of the hand, mobile cloud computing is expected to be the next big trend in cloud computing research. Effective remote display protocols are one essential technology for enabling the vision of mobile cloud computing. However, existing remote display protocols have been developed for non-mobile computers such as desktops and are not ready to fully embrace the challenges and requirements distinct to mobile devices in mobile cloud computing settings. Recognizing the importance of energy efficiency on mobile devices, this paper investigates the correlation between energy consumption and various encoding types, and proposes a power-aware encoding scheme to maximize the connection lifespan of battery-driven devices to the cloud. The proposed scheme considers the patterns of power consumption by CPUs and wireless network interface cards for the purpose of reducing power usage for remote display activities. Experimental results show that the proposed hybrid encoding scheme outperforms existing approaches in terms of power efficiency.</P>

Competition among golf courses in South Korea for recruiting and retaining customers has increased dramatically over the years as the number of golf courses grew. Therefore, it has become important for golf course managers to understand what attracts golfers to their golf courses. The purpose of this study was to examine the relationships among motivation, service quality, customer satisfaction, and loyalty among recreational golfers in South Korea. Total 563 recreational golfers participated in this study and responded to a self-administered survey measuring the constructs. The result showed that intrinsic motivation and extrinsic motivation were positively associated with service quality whereas amotivation was negatively related to service quality. In addition, golfers` perceived service quality had a positive impact on golfers` satisfaction and loyalty towards golf courses. These results suggest that golf course managers should understand the types and levels of motivation golfers have and develop strategies for enhancing service quality.
Age, Gender, Perceived Social Support, and Organizational Commitment among Fitness Employees

The purpose of this study was to examine the relationships between age, gender, perceived social support, and organizational commitment among fitness employees in the United States. A total of 420 employees completed the questionnaires measuring their demographic information and proposed variables. Confirmatory factor analysis and structural equational modeling technique was employed to test the theoretical model. The findings of this study revealed that fitness employees` gender was positively related to their perceived social support. However, employees` age was not significantly related to their perceived social support. Finally, perceived social support was found to be positively related to their organizational commitment. The results of this study highlighted the importance of gender differences in the perception of social support among fitness employees, which in turn affect their organizational commitment.

Managing human capital in an effective manner is always a challenge for sport managers. The purpose of the study was to examine the relationships among perceived support, collectivism, affective commitment, work effort, and intention to leave among sport instructors across the United States. The data were collected from 379 ACSM certified sport instructors using online questionnaire. The results showed that perceived support, which is represented by coworker support, supervisor support, and organizational support, and collectivism had a significant impact on affective commitment explaining 75.0% and 13.2% of the variances respectively. In addition, affective commitment explained 19.0% of the variance in work effort and 61.9% of the variance in intention to leave. This study significantly contributes to the existing body of knowledge in sport management literature and provides meaningful guidance to sport managers on how to retain valuable employees and elicit the best work effort they could offer.
Revisiting the Model of Sport Consumption: Review of Theories and Suggestions

One of the important tasks of sport marketers is developing effective marketing strategies for increasing game attendance. In order to do so, it is important to understand the overall psychological process of sport consumers. The purpose of this paper is to revisit the model of sport consumption suggested by Trail and James (2008) and review various theories explaining the relationships among the constructs. In particular, identity theory, social identity theory, the theory of planned behavior, attitude theory, decision-making theory, and satisfaction theory are discussed in relation to the model of sport consumption. In addition, possible deficiencies of the model as well as possible inclusions of other constructs and paths that can help enhancing the model are discussed.
