Medicinal Mushroom Ganoderma lucidum (Leyss: Fr) Karst. Triggers Immunomodulatory Effects and Reduces Nitric Oxide Synthesis in Mice

        Rosália Rubel,Herta S. Dalla Santa,Sandro J.R. Bonatto,Se´rgio Bello,Luiz Cla´udio Fernandes,Raffaello Di Bernardi,Juliana Gern,Cid Aimbire´ M. Santos,Carlos Ricardo Soccol 한국식품영양과학회 2010 Journal of medicinal food Vol.13 No.1

This study investigated the effect of Ganoderma lucidum supplementation on lymphocytes and peritoneal macrophages from mice. Our results show that G. lucidum in vivo was able to increase interferon-γ (IFN-γ) concentration but reduced CD3+ and CD8+ spleen lymphocytes. Ex vivo, IFN-γ; and interleukin-10 levels were increased and the tumor necrosis factor-α (TNF-α) level was reduced by peritoneal macrophages from mice fed with G. lucidum. In the absence of stimuli nitric oxide production was reduced in mice fed with G. lucidum, and with lipopolysaccharide stimulation nitric oxide production was increased but was lower than control values (P<.05). G. lucidum was grown by solid-state culture in wheat grain, and a chow containing 10% G. lucidum mycelium was formulated (G10). Swiss male mice were divided into two groups, termed G10 and control groups according to the diet, and were fed for 3 months. Peritoneal macrophages were obtained and investigated with regard to phagocytosis, lysosomal volume, hydrogen peroxide, superoxide anion, and cytokines ex vivo. In the plasma we investigated concentrations of cytokines, and in the spleen we determined subsets of CD3+, CD4+, CD8+, and CD19+ lymphocytes.

Antioxidant Activity and Phenolic and Mineral Content of Rose Grape Juice

        Dani, Caroline,Oliboni, Livia S.,Vanderlinde, Regina,Pra, Daniel,Dias, Johnny F.,Yoneama, Maria Lucia,Bonatto, Diego,Salvador, Mirian,Henriques, Joao A.P. The Korean Society of Food Science and Nutrition 2009 Journal of medicinal food Vol.12 No.1

There are many studies related to the antioxidant activity of grape products; however, they concern only purple and white grape varieties. Up to now, there are no reports of studies on the Goethe rose grape variety, either on its antioxidant activity or on its phenolic and mineral quantification. Thus, the purpose of this study was to evaluate in vitro and in vivo antioxidant activity, as well as to quantify total phenolic compounds, ascorbic acid, and mineral content, in a Goethe rose grape juice. The results obtained showed that the Goethe rose grape juice is a great polyphenol source, which contains catechin, epicatechin, and procyanidins ($B_1$, $B_2$, $B_3$, and $B_4$). Of all metals analyzed, potassium, calcium, magnesium, and iron showed the highest values. We found that this rose grape juice shows an important antioxidant activity in in vitro (2,2-diphenyl-1-picrylhydrazyl radical scavenging activity) and in vivo (using the Saccharomyces cerevisiae yeast cells) assays. The antioxidant activity could be explained by the significant phenolic content and ascorbic acid levels found in the juice. The results showed that rose grape juice is an excellent antioxidant source, which could contribute to the prevention of many diseases related to oxidative stress, such as atherosclerosis and Parkinson's disease.

Hypolipidemic and Antiatherosclerotic Potential of Pleurotus ostreatus, Cultived by Submerged Fermentation in the High-fat Diet Fed Rats

Leandro Freire dos Santos,Ana Lucia Zanatta,Vanete Thomaz Soccol,Maria Fernanda Torres,Sandro José Ribeiro Bonatto,Rosália Rubel,Carlos Ricardo Soccol 한국생물공학회 2013 Biotechnology and Bioprocess Engineering Vol.18 No.1

The ability of Pleurotus ostreatus biomass, cultived by submerged fermentation, to produce beneficial effect on lipid profile and macrophages activity during a high-fat diet (HFD) for a long-term intake was investigated. Blood samples were collected through cardiac puncture to measure the plasma cholesterol, triglycerides, low-density protein (LDL), high-density protein (HDL), aspartate aminotransferase (AST) activity, urea– blood urea nitrogen (BUN)/creatinine ratio of rats fed on an HFD for 4 months. Dosage of lipid hydroperoxides was carried out on methanolic extract of liver tissue. Peritoneal macrophages activity was evaluated in relation to the superoxide anion, hydrogen peroxide and nitric oxide production, phagocytosis and lysosomal volume. The administration of P. ostreatus significantly altered the lipid profile and oxidative stress as related to the LDL and triglycerides decrease and inhibitory effects on superoxide anion and hydrogen peroxide production. All findings of this study lead us to suggest that the P. ostreatus maybe a beneficial agent in the hyperlipidemia and atherosclerosis treatments.

J-PLUS: Identification of low-metallicity stars with artificial neural networks using SPHINX

        Whitten, D. D.,Placco, V. M.,Beers, T. C.,Chies-Santos, A. L.,Bonatto, C.,Varela, J.,Cristó,bal-Hornillos, D.,Ederoclite, A.,Masseron, T.,Lee, Y. S.,Akras, S.,Borges Fernandes, M.,Caballero, J. Springer-Verlag 2019 Astronomy and astrophysics Vol.622 No.-

<P><I>Context.</I> We present a new methodology for the estimation of stellar atmospheric parameters from narrow- and intermediate-band photometry of the Javalambre Photometric Local Universe Survey (J-PLUS), and propose a method for target pre-selection of low-metallicity stars for follow-up spectroscopic studies. Photometric metallicity estimates for stars in the globular cluster M15 are determined using this method.</P><P><I>Aims.</I> By development of a neural-network-based photometry pipeline, we aim to produce estimates of effective temperature, <I>T</I>eff, and metallicity, [Fe/H], for a large subset of stars in the J-PLUS footprint.</P><P><I>Methods.</I> The Stellar Photometric Index Network Explorer, SPHINX, was developed to produce estimates of <I>T</I>eff and [Fe/H], after training on a combination of J-PLUS photometric inputs and synthetic magnitudes computed for medium-resolution (<I>R</I> ~ 2000) spectra of the Sloan Digital Sky Survey. This methodology was applied to J-PLUS photometry of the globular cluster M15.</P><P><I>Results.</I> Effective temperature estimates made with J-PLUS Early Data Release photometry exhibit low scatter, σ(<I>T</I>eff) = 91 K, over the temperature range 4500 < <I>T</I>eff (K) < 8500. For stars from the J-PLUS First Data Release with 4500 < <I>T</I>eff (K) < 6200, 85 ± 3% of stars known to have [Fe/H] < −2.0 are recovered by SPHINX. A mean metallicity of [Fe/H] = − 2.32 ± 0.01, with a residual spread of 0.3 dex, is determined for M15 using J-PLUS photometry of 664 likely cluster members.</P><P><I>Conclusions.</I> We confirm the performance of SPHINX within the ranges specified, and verify its utility as a stand-alone tool for photometric estimation of effective temperature and metallicity, and for pre-selection of metal-poor spectroscopic targets.</P>
