        관상동맥 질환에서 CD14 유전자형에 따른 세균 및 Heat Shock Protein에 대한 반응의 차이

        Background : CD14 is the receptor for lipopolysaccharides and heat shock protein (HSP), which has been suggested being associated with increased risk of coronary artery disease (CAD). We investigated whether the response to infectious agents or HSP is different according to CD14 polymorphism in Koreans. Materials and Methods : Antibody titers to Helicobacter pylori, Chlamydia pneumoniae, and human HSP60 (hHSP60) were measured in 48 patients with stable CAD and in 41 healthy controls by ELISA. CD14 genotype was determined by PCR and high-sensitivity C-reactive protein (hs-CRP) was measured. Results : Seropositivity to C. pneumoniae and H. pylori, and antibody titer to hHSP60 were not significantly associated with the presence of CAD. CD14 genotype distribution was 31 TT (35%), 43 CT (48%), and 15 CC (17%). To compare the response to the infectious organism and hHSP60, we divided study population into 3 groups; CAD patients with non-TT genotype (group I, n=30), CAD patients with TT genotype (group II, n=18), and normal controls (group III, n=41). Seropositivity to C. pneumoniae and H. pylori, and antibody titer to hHSP60 were not significantly different among 3 groups. Though hs-CRP level was significantly different among 3 groups, post-Hoc analysis showed that hs-CRP level was not significantly different between group I and group II (group I: 1.6[1.1-3.5] mg/L and group II: 0.35[0.1-2.0] mg/L). Conclusions : This study suggests that the inflammatory responses to infectious organisms and HSP do not differ according to the CD14 genotype in Koreans. 목적 : 만성적인 세균 감염이나 자가 면역 반응이 동맥경화와 연관되어 있다는 주장이 제기되어 왔다. CD14은 lipopolysaccharides (LPS)와 heat shock protein(HSP)의 수용체로 C(-260)→T 다형성이 관상동맥 질환의 위험과 관련되어 있다고 제안되어 왔다. 본 연구에서는 한국인에서 CD14 다형성에 따른 LPS와 HSP에 대한 반응성을 염증표지자를 측정하여 살펴보고자 하였다. 재료 및 방법 : 안정형 관상동맥 환자 48명과 정상 대조군 41명을 대상으로 Chlamydia pneumoniae, Helicobacter pylori 항체 및 인체 HSP60 (hHSP60) 항체 역가를 enzyme-linked immunosorbent assay (ELISA) 방법으로 측정하였다. CD14 유전자형은 중합효소연쇄반응을 이용하여 결정하였고 hs-CRP를 측정하였다. 결과 : C. pneumonias 및 H. pylori 항체 양성 여부 및 항체 역가, 그리고 hHSP60에 대한 항체 역가는 안정형 관상동맥 질환의 유무와 유의한 상관관계는 없었다. CD14 유전자형의 분포는 TT 30명(39%), CT 31명(40%), 그리고 CC 16명(21%)이었다. 관상동맥 질환 군에서 TT 유전자형은 38%, 대조군에서는 32%로 TT 유전자형과 관상동맥 질환의 유무와는 유의한 상관관계가 발견되지 않았다(P>0.05). CD14 유전자형에 따라 C. pneumonias와 H.pylori 감염 및 hHSP60에 따른 반응에 차이가 있는지 여 부를 알아보기 위해, 관상동맥 질환 군을 다시 TT 유전자형 군과 non-TT 유전자형 군으로 나누어, 전체적으로 세군에서 항체 양성률과 역가를 비교하였다(I군: CD14 TT 유전자형을 가진 안정형 관상동맥 환자, II군: CD14 TT 유전자형을 가진 안정형 관상동맥 환자, III군: 정상 대조군). 세 군에서 C. pneumonias와 H. pylori 항체 양성률 및 hHSP60 항체 역가에는 유의한 차이가 없었다. 세 군사이에 hs-CRP 값에 유의한 차이가 있었지만, post-Hoc분석에서 II군의 hs-CRP 값이 I군에 비해 유의하게 높지는 않았다(0.35 [0.1-2.0] mg/L in group II vs. 1.6 [1.1-3.5]mg/L in group I, P>0.05). 결론 : 본 연구는 한국인에서 CD14 유전자형에 따라 LPS나 HSP에 대한 반응이 유의하게 다르지 않음을 시사한다.

      • 알코올 의존 환자의 Tryptophan Hydroxylase 유전자 다형성

        연구목적 : 알코올 의존 환자에서 세로토닌 합성 조절 효소인 tryptophan hydroxylase(TPH) 유전자 다형성 빈도를 정상 대조군과 비교함으로써 알코올 의존의 유전적 요인을 추적하고, 임상변인과 이 유전자 다형성과의 관련성을 알아보고자 하였다. 방 법 : DSM-IV진단기준에 부합되는 알코올 의존 환자 100명과 정상 대조군 100명을 대상으로 TPH유전자 다형성을 증합효소 연쇄반응과 제한효소 처리법을 이용하여 동정하였다. 여기서 분리된 대립 유전자와 유전자형에 따른 빈도의 차이를 서로 비교하였으며, 알코올 의존 환자군의 여러 임상 변인에 따른 차이를 비교하였다. 결 과 : 알코올 의존 환자군과 대조군 간에 TPH의 A216C 유전자형 및 대립유전자 빈도에서의 통계적으로 유의한 차이는 관찰되지 않았으나, 조기 발병한 환자의 경우는 유전자형의 빈도가 AA,AC,CC형이 0.57, 0.39, 0.04, 후기 발병한 환자의 경우 0.34, 0.45, -.21로, 조기 발병한 환자군에서 대조군과 비교하여 통계적으로 유의하게 A 대립 유전자의 빈도가 높은 것이 관찰되었다.(by chi-square test, p<0.05). 결 론 : 이는 조기 발병형 알코올 의존의 경우 TPH유전자 다형성과 관련이 있으며, 일부 알코올 의존 환자에서 유전적으로 세로토닌계의 이상이 있다는 사실을 시사한다. Objectives : This study was performed to explore the association of tryptophan hydroxylase (TPH) gene with diagnosis of alcohol dependence and/or clinical characteristics such as age of onset, family history, and severity of symptoms in Korean alcoholics. Methods : The genotype and allele frequencies of TPH in 100 male hospitalized patients who met DSM-IV criteria for alcohol dependence were investigated using polymerase chain reaction and restriction fragment length plymorphism and were compared with 100 age-matched healthy male control subjects. And the associations between gene polymorphisms and clinical characteristics in alcoholic patients were explored. Results : The distributions of TPH genotype and allele in alcohol dependent patients were not different from control subjects. However, the frequencies of TPH genotype in early-onset alcoholic patients, which were 0.57, 0.39, and 0.04(AA, AC and CC, respectively), were significantly different from those of late-onset alcoholics(0.34, 0.45, and 0.21, respectively). "A" allele was found more frequent in early-onset alcoholics. Conclusion : The result suggests that TPH gene polymorphism is associated with early-onset alcioholic patients possibly related with inherited abnormalities of serotonin system.

        Experimental Restoration of a Salt Marsh with Some Comments on Ecological Restoration of Coastal Vegetated Ecosystems in Korea

        Since the 1980s, the coastal wetlands in Korea have been rapidly degraded and destroyed mainly due to reclamation and landfills for coastal development. In order to recover damaged coastal environments and to develop wetland restoration technologies, a 4-year study on ecological the restoration of coastal vegetated ecosystems was started in 1998. As one of a series of studies, a small-scale experiment on salt marsh restoration was carried out from April 2000 to August 2001. The experiment was designed to find effective means of ecological restoration through a comparison of the changes in environmental components and species structure between two different experimental plots created using sediment fences, one with and one without small canals. Temporal variation in surface elevation, sedimentary facies, and benthic species were measured seasonally in each plot and in the adjacent natural reference sites. Monthly exposure occurred from 330 cm to mean sea level, which represents the critical tidal level (CTL) at which salt marsh plants colonize. Vegetation, especially Suaeda japonica, colonized the site the following spring and recovered to a similar extent in the natural marshes 16 months later. The sedimentary results indicated that the sediment fences had effects on particle size and sediment accumulation, especially in the plot with small canals. This experiment also showed that tidal height, especially that exceeding the CTL, is an important factor in the recovery of the benthic fauna of salt marshes. From these results, we suggested that designs for the restoration of salt marsh ecosystems must consider the inclusion of a tidal height exceeding CTL, as this may allow reconstruction of the previous natural ecosystem without artificial transplanting.

        Benthic Pollution Assessment Based on Macrobenthic Community Structure in Gamak Bay, Southern Coast of Korea

        Benthic pollution assessment based on macrobenthic community structure with environmental variables was carried out at twelve stations during two periods on a presumed pollution gradient in Gamak Bay. Univariate and multivariate methods were applied to investigate structural changes in the benthic communities. A clear gradient of pollution effects on the macrobenthic community was observed from the interior to the exterior of the bay. The community on the northwestern basin was severely disturbed due to a low level of hydrodynamics and a large amount of pollutant input from nearby cities. Exterior regions on the southern basin appeared to have the best benthic environmental characteristics among all stations according to most methods of analysis. Central ridge regions and two stations around the islets in the mouth of the bay exhibited intermediate levels of perturbation when compared to the more disturbed interior and undisturbed exterior regions. Pollution effects on the communities were attenuated at the southern area of the central ridge during spring compared to those of summer, where aquacultural farming was densely distributed. The environmental variables primarily correlated to the macrobenthic community structure were total organic carbon (C), organochlorine pesticides (OCPs), and tributyltins (TBTs), contents found on the surface sediment, as anthropogenic variables indicating organic materials.

        Fibrinolytic Activity of a Novel Serine Protease from the Hemolymph of a Polychaeta, Periserrula Leucophryna

        We purified and characterized a novel protease with fibrinolytic activity from the hemolymph of a polychaeta, Periserrula leucophryna. The enzyme was isolated by chromatographic methods using Phenyl-Sepharose and Benzamidine-Sepharose. SDS-PAGE and gel filtration revealed a single polypeptide chain with a molecular weight of 30 kDa. The N-terminal sequence was determined to be IVGGQNARQGEFPWQV. The purified enzyme preferentially cleaved the synthetic substrates that had Lys (rather than Arg) at the P1 position and did not efficiently cleave substrates with nonpolar amino acids. Among chromogenic protease substrates, the substrate that was most susceptible to hydrolysis by Periserrula leucophryna fibrinolytic protease (PLFP) was Val-Leu-LyspNA (substrate for plasmin). The inhibition profile revealed the protease belongs to a family of serine proteases and has plasmin-like activity that is strongly inhibited by α2-antiplasmin. The purified PLFP was able to dissolve the artificial fibrin, and its fibrinolytic behavior is similar to that of plasmin. In conclusion, PLFP is a novel protease and has potential for practical applications in thrombolytic therapy.

        The Sediment-Water Interface Increment due to the Complex Burrows of Macrofauna in a Tidal Flat

        The architecture of macrofaunal burrows and the total area of the sediment-water interface created by biogenic structure were investigated in the Donggeomdo tidal flat on the west coast of Korea. Resin casting methods were applied to recover burrows of four dominant species, Macrophthalmus japonicus, Cleistostoma dilatatum, Perinereis aibuhitensis, and Periserrula leucophryna, and whole burrows within the casting area at three sites in different tidal levels. P. leucophryna excavated the largest burrow in terms of a surface area among them. In the case of whole burrow casting, the space occupied by the biogenic structure was extended into deeper and expanded more greatly at the higher tidal level. In the uppermost flat, the burrow wall surface area within sediment was more extensive than the sediment surface area. Increased oxygen supply through the extended interface could enhance the degradation rates of organic carbon and also change the pathways of degradation. Quantifying the relationship between the extended interface and mineralization rate and pathway requires more extensive study.
