      • CXCL12-CXCR4 Promotes Proliferation and Invasion of Pancreatic Cancer Cells

        Objective: CXCL12 exerts a wide variety of chemotactic effects on cells. Evidence indicates that CXCL12, in conjunction with its receptor, CXCR4, promotes invasion and metastasis of tumor cells. Our objective was to explore whether the CXCL12-CXCR4 biological axis might influence biological behavior of pancreatic cancer cells. Methods: Miapaca-2 human pancreatic cancer cells were cultured under three different conditions: normal medium (control), medium + recombinant CXCL12 (CXCL12 group), or medium + CXCR4-inhibitor AMD3100 (AMD3100 group). RT-PCR was applied to detect mRNA expression levels of CXCL12, CXCR4, matrix metalloproteinase 2 (MMP-2), MMP-9, and human urokinase plasminogen activator (uPA). Additionally, cell proliferation and invasion were performed using CCK-8 colorimetry and transwell invasion assays, respectively. Results: CXCL12 was not expressed in Miapaca-2 cells, but CXCR4 was detected, indicating that these cells are capable of receiving signals from CXCL12. Expression of extracellular matrix-degrading enzymes MMP-2, MMP-9, and uPA was upregulated in cells exposed to exogenous CXCL12 (P<0.05). Additionally, both proliferation and invasion of pancreatic cancer cells were enhanced in the presence of exogenous CXCL12, but AMD3100 intervention effectively inhibited these processes (P<0.05). Conclusions: The CXCL12-CXCR4 biological axis plays an important role in promoting proliferation and invasion of pancreatic cancer cells.

      • The Influence of Sharing Evaluation Information on Consumer Buying Behavior in Social Commerce

        Social commerce is a new generation of e-commerce. Compared to traditional commerce, social commerce, treating social media and social network as a carrier, promotes online trading and information exchange related to commercial activities. Information sharing in social commerce affects buying behavior of consumers in all aspects. Therefore, research on information sharing in social commerce has important implications for further investigation in consumer behavior and business marketing strategy. In this paper, the authors propose a method of multi-agent simulation based on FIRE model, to discuss the influence of sharing evaluation information on consumer buying behavior in social commerce. This study established a simulation model of consumer behavior based on the information, took full account of the effect given by situational factors and information sources, set up rules on consumer decision of purchase on the basis of consumer sub-model and merchant sub-model, realized the simulation for two different information sharing situation by using Repast, and drew relevant conclusions according to simulation results about the total number of transactions and trading volatility of each type of merchants.

        Removal characteristics of nitrogen oxides and particulates of a novel Mn-Ce-Nb-Ox/P84 catalytic filter applied for cement kiln

        A novel Mn-Ce-Nb-Ox/P84 catalyticfilter for synergetic removal of particulates and NO was designed andprepared by a new method using functional foaming coating. Removal characteristics of nitrogen oxides(including catalyst loading amount, O2 concentration, NH3/NO molar ratio, H2O and SO2) andparticulates, were systematically studied. Results showed that the NO removal efficiency of catalyticfilter reached 95.3% at 200 C, as the catalyst loading amount,filtration velocity and mole ratio of NH3/NOwere set to be 450 g/m2, 1 m/min and 1, respectively. The removal characteristic of particulates of Mn-Ce-Nb-Ox/P84 catalyticfilter belonged surfacefiltration and the PM2.5 removal efficiency reached 99.98%.

      • Mechanism of HN-3 cell apoptosis induced by carboplatin: Combination of mitochondrial pathway associated with Ca <sup>2+</sup> and the nucleus pathways

        <P>Laryngeal carcinomas have been recognized as a serious health threat worldwide. In the present study, the mechanism of apoptosis in HN-3 cells induced by carboplatin (CBCDA), a widely used anti-cancer drug, was investigated. The pro-apoptotic effect of CBCDA in HN-3 cells was demonstrated to be time- and dose-dependent. Therefore, the stages of apoptosis were investigated in chronological order. The results demonstrated that excessive generation of cytosolic Ca<SUP>2+</SUP> in HN-3 cells was initially triggered when cells were exposed to CBCDA, followed by the appearance of mitochondrial depolarization and oxidative stress, leading to the release of apoptosis-inducing factor. At later stages, expression of caspase-8 was increased due to the apoptotic signals originating from CBCDA-induced DNA damage, as well as caspase-9 and poly ADP ribose polymerase (PARP) expression upregulation. Glutathione decreased the available CBDCA concentration, decreased apoptosis and alleviating oxidative stress, thus reducing the actual effective concentration. Mechanistic research may benefit the rational design of more efficient therapeutic strategies as well as development of novel platinum-based agents.</P>

      • Application on Web Page Filtering Technology

        Web page filtering technology intends to filter out the large number of the repeated and theme-unrelated noise information and obtain useful information. Some web filtering methods cannot make full use of the layout and visual features. In view of the new mainstream “DIV+CSS” designing style of modern commercial web sites, this paper summarizes that elements laying in the same div blocks have common semantic features and proposed a DIV_FOREST model to represent the web pages. And in combination with the Vision-based Page Segmentation Algorithm, a DVPS Algorithm which considers both layout features and visual features was proposed to improve web page filtering efficiency.

        UPLC-QTOF-MS/MS Analysis for Steaming Times-dependent Profiling of Steamed Panax quinquefolius and its Ginsenosides Transfromations induced by Repetitious Steaming

        The metabolic profiles of Panax quinquefolius and its associated therapeutic values are critically affected by the repetitious steaming times. The times-dependent steaming effect of P. quinquefolius is not well-characterized and there is also no official guidelilne on its times of steaming, In this paper, a UPLC-Q-TOF-MS/MS method was developed for the qualitative profiling of multi-parametric metabolic changes of raw P. quinquefolius durilng the repetitious steaming process. Our method was successful in discriminating the differentially multi-steamed herbs. Meantime, the repetitious steaming-inducing chemical transformations in the preparation of black American ginseng (American ginseng that was subjected to 9 cycles of steaming treatment) were evaluated by this UPLC-Q-TOF-MS/MS based chemical profiling method. Under the optimized UPLC-Q-TOF-MS/MS conditions, 29 major ginsenosides were unambiguously identified and/or tentatively assigned in both raw and multi-steamed P. quinquefolius within 19 min, among them 18 ginsenosides were detected to bo newly generated during the preparatory process of black American ginseng. The mechanisms involved were further deduced to be hydrolysis, dehydration, decarboxylation and addition reactions of the original ginsenosiders in raw P. quinquefolius through analyzing 9 cycles of steaming extracts of 14 pure reference ginsenosides. Our novel steaming times-dependent metabolic profiling represents the paradigm shift in the global quality control of multi-steamed P. quinquefolius products.

        Field monitoring of splitting failure for surrounding rock masses and applications of energy dissipation model

        Due to high in-situ stress and brittleness of rock mass, the surrounding rock masses of underground caverns are prone to appear splitting failure. In this paper, a kind of loading-unloading variable elastic modulus model has been initially proposed and developed based on energy dissipation principle, and the stress state of elements has been determined by a splitting failure criterion. Then the underground caverns of Dagangshan hydropower station is analyzed using the above model. For comparing with the monitoring results, the entire process of rock splitting failure has been achieved through monitoring the splitting failure on side walls of large-scale caverns in Dagangshan via borehole TV, micro-meter and deformation resistivity instrument. It shows that the maximum depth of splitting area in the downstream sidewall of the main power house is approximately 14 m, which is close to the numerical results, about 12.5 m based on the energy dissipation model. As monitoring result, the calculation indicates that the key point displacement of caverns decreases firstly with the distance from main powerhouse downstream side wall rising, and then increases, because this area gets close to the side wall of main transformer house and another smaller splitting zone formed here. Therefore it is concluded that the energy dissipation model can preferably present deformation and fracture zones in engineering, and be very useful for similar projects.

      • The impact of total retrieved lymph nodes on staging and survival of patients with pT3 gastric cancer

        <B>BACKGROUND.</B><P>The incidence of lymph node metastasis is high in patients who have pT3 gastric cancer. However, the impact of total retrieved lymph nodes (tLNs) on staging and survival of these patients is not clear.</P><B>METHODS.</B><P>For this study, the authors examined 1895 patients with pT3 gastric cancer who underwent surgery at Yonsei University Medical College from January 1987 to June 2000.</P><B>RESULTS.</B><P>Four hundred sixty of 1895 patients (24.3%) were diagnosed with pT3N0 gastric cancer. Patients who had < 31 tLNs (25th percentile) had less advanced lymph node (N) stage than the other patients (P < .001). Lymph node metastasis had a positive association with the number of tLNs in a logistic regression analysis (P < .001; hazards ratio, 1.014; 95% confidence interval, 1.006–1.021). With a median follow-up of 61.1 months, the overall 10-year survival rate (10-YSR) was 42.8%. Patients with pT3N0 disease who had < 31 tLNs had a 10-YSR of only 55.4%. Although this 10-YSR did not differ significantly from the rate for patients with N0 disease who had ≥31 tLNs (65.8%; P = .108), it approached the rate for the N1 group (53.3%; P = .207). In multivariable analyses, the number of tLNs emerged as an independent prognostic predictor in patients with pT3N2 and pT3N3 disease, but not in patients with pT3N0 or pT3N1 disease.</P><B>CONCLUSIONS.</B><P>Increasing numbers of tLNs may improve the accuracy of staging in patients who have pT3 gastric cancer. Because preoperative lymph node staging is difficult, a thorough lymph node dissection is mandatory in all serosa-positive patients. Cancer 2007. © 2007 American Cancer Society.</P>
