        고정화 시스템을 이용한 용균효소의 생산

        해양에서 분리된 용균활성이 우수한 균주의 균체고정화에 의한 연속생산을 위한 조건을 검토하였다. Bacillus subtilis SH-1의 균체를 고정화하여 column형 reactor에 의하여 연속적으로 효소생산을 실험하였다. 연속발효의 최적조건은 담체는 3.0% sodium alginate로 직경 2.4mm의 bead를 제조하여 기질의 주입속도는 20ml/h 공기의 주입속도는 2.0ml/min이였다. Column형 reactor(용량 2.8ℓ)에 의한 연속발효시 25일까지 76.5~88.0 units/ml의 최적의 효소활성을 갖는 용균효소를 생산할 수 있었다. Bacillus subtilis SH-1 screened from coastal sea water of South Korea was used to produce bacteriolytic enzyme. The production of bacteriolytic enzyme by immobilized cells was investigated. The optimum conditions for the continuous production of the bacteriolytic enzyme using immobilized cells were 2.4 mm diameter of 0.3% alginate beads, 20 ml/h of substrate feeding rate and 20 ℓ/min of aeration rate. A productivity of 76.5 to 88.0 units/ml could be obtained for 25 days by continuous column reactor under the optimum conditions.

        해양에서 분리한 Bacillus subtilis SH-1이 분비하는 용균효소의 정제 및 특성

        용균활성이 우수한 균주를 사용하여 균주가 생산하는 용균효소를 분리 정제하고 그 특성을 검토하였다. Bacillus subtilis SH-1이 생산하는 효소의 정제는 최적 조건에서 얻은 배양액의 상징액을 황산암모늄 침전, CM-cellulose 및 Sephadex G-100에 의한 gel 여과 및 hydroxylapatite column chromatography를 행하였을 때 본 효소는 약 66.5배로 정제되었으며 회수율은 18.5%였다. 본 효소의 최적 pH는 9.0, 최적온도는 50℃이었고, pH는 6.0~10.0 범위에서 안정하였으며 열 안정성은 30℃이하에서는 안정하나 60℃에서는 완전 실활되었다. 본 효소의 분자량은 23,000 dalton으로, 단일효소 단백질이었다. 식중독의 원인균을 주 대상으로 용균효소에 대한 cell의 용해율을 조사한 결과 대부분의 Gram 음성균에 대해서는 용해율이 31% 이하였으며 Gram 양성균에 대해서는 cell의 용해율이 48% 이상으로 나타났다. 특히 Vibrio sp.와 Listeria sp. 균주에 대한 용해율은 73% 이상이었다. Peptidoglycan 분해산물의 측정결과 탁도가 감소됨에 따라 free amino group이 유리되어 나왔으며 casein의 가수분해능이 있는 것으로 보아 본 효소는 endopeptidase로 추정되었다. The bacteriolytic enzyme produced from Bacillus subtilis SH-1 was purified and characterized, and its molecular weight was determined. The bacteriolytic enzyme activity was increased about 66.5 times via purification with recovery yield of 18.5%. The optimum pH and temperature of this enzyme were 9.0 and 50℃. The enzyme was stable within a pH range of 6.0~10.0 and unstable above 60℃. The molecular weight of the enzyme was estimated to be 23,000 dalton in a form of monomer with no other subunits. Effect of the enzyme on the lysis of bacteria engaged in food posion was tested. The lysis degree was below 31% against Gram negative bacteria and above 48% in Gram positive bacteria. The values higher than 73% were obtained against Vibrio sp. and Listeria sp. As the turbidity of dissolved peptidoglycan decreases, the free amino group levels were increased. And, based on hydrolysis of casein, this enzyme was thought to be an endopeptidase.

      • 無機凝集劑에 依한 Kaolin 縣濁液 凝集에 있어서 Amylose가 미치는 效果

        An investigation was carried out to compare the coagulation effects of inorganic coagulants, amylose and inorganic coagulants + amylose on coagulation of kaolin suspension. Inorganic coagulants used were Al_2(SO_4)_3, K-Alum, CaCl_2, MgCl_2, KCl and NaCl. The results are as follows : 1) When the inorganic coagulant only is added to kaolin suspension, ⅰ) The coagulation effect is superior to the other two cases. ⅱ) The coagulating power of the inorganic coagulants is ranked as follows : Al_2(SO_4)_3> K-Alum> CaCl_2 > MgCL_2 > KCl > NaCl It is consistent with the studies of other investigators. ⅲ) The coagulation values of inorganic coagulants containing counter-ions with charge numbers of 3, 2 and 1 i.e. Al_2(SO_4)_3, MgCl_2 and NaCl are in the ratio of : 1.227 x 10 exp(-3) : 1.446 x 10 exp(-2) : 5.020 x 10 exp(-1) It is consistent with the Schulze & Hardy rule and D.L.V.O. theory. iv) Because of cationic antagonism between potassium ions and aluminium ions in kaolin suspension, the coagulating power of K-Alum is weaker than that of Al_2 (SO_4)_3. v) The coagulation values decrease with increasing of molecular weight in the same ionic coagulants. 2) When an inorganic coagulant with amylose is added to kaolin suspension, the coagulation effect is poorer than that when the inorganic coagulant only is added. As the charge number of the inorganic coagulant increases the coagulation effect is uniformly increased, but the magnitude of the increase is small. 3) When amylose only is added to kaolin suspension, i) The coagulation effect is the worst among the three cases. ii) Only over a narrow range of dilute amylose concentration the coagulation occurs slightly. 4) A logarithmic relationship between the relative turbidity and the settling time is as follows ; log Trel = -m log t + log C°

      • 食用油脂類에 對한 免疫生物學的 硏究

        食用油脂는 健康維持에 必須的인 營養素이다. 그러나 食用油脂는 脂肪을 構成하는 脂肪酸의 種類와 攝取하는 量에 따라 健康을 保衛하는 必須的인 食品의 成分이 되기도 하고 有害한 影響을 끼칠 수도 있어서 脂質代謝와 關連된 疾病 뿐만아니라 老化를 促進하고, 乳房癌, 大腸癌等의 癌發生과도 關連된 것으로 밝혀져 있다. 本 硏究는 우리生活環境에서 食用으로 接할 수 있는 10種의 油脂에 對한 免疫生物學的인 活性에서 究明한 結果 對照群인 참기름食餌群에 比해 올리브유食用群은 全般的으로 體液性 및 細胞性免疫을 有意性있게 增加시켰으나, 大食細胞의 活性과 末梢循環白血球數는 有意性없는 減少를 보였고, 動物性 油脂인 牛脂食餌群과 植物性 油脂인 옥수수기름食餌群, 들기름食餌群, 米糠油食餌群, 大豆油食餌群, 菜種油食餌群 및 고추씨기름食餌群에서는 全般的으로 體液性 및 細胞性免疫, 大食細胞의 活性 및 末梢循環白血球數를 有意性있게 減少시켰다. This study was performed to investigate the effects of edible oils and fats on the immunobiological responses in ICR male mice. Ten groups of experimental diets, such as sesame oil diet, beef tallow diet, lard oil diet, olive oil diet, corn oil diet, perilla oil diet, rice polishing oil diet, soybeen oil diet, rape seed oil diet, and red pepper seed oil diet were fed adlibitum to the ICR male mice for 27 days. The results of this study were summarized as followings. 1. Both humoral and cellular immune responses, phagocyte activity, and number of leukocytes in sesame oil group were increased. However, the increasing rate of body weight and the weight retios of spleen and thymus to body were generally decreased. 2. Hemagglutination titer(HA) and rosette forming cell(RFC) of the beef tallow group and the lard group were significantly lower than the sesame oil group(P<0.01), but the lard group significantly increased Arthus reaction and delayed type hypersensitivity(DTH)(P<0.01). 3. Arthus reaction, DTH, and RFC of the olive oil group were significantly higher than the sesame oil group(P<0.01). 4. DTH of the corn oil group was significantly higher than the sesame oil group(P<0.01) whereas phagochyte activity was significantly lower(P<0.01). But no statistically significant differences were found between the two groups in the humoral immune response. 5. HA, PFC, DTH, RFC, phagocyte activity, and number of leukocytes of the perilla oil group and the rice polishing oil group were significantly lower than the sesame oil group(P<0.01). 6. Both humoral and cellular immune responses, phagocyte activity, and number of leukocytes of the soybeen oil group, the rape seed oil group, and the red pepper oil group were significantly lower than the sesame oil group(P<0.01).

        To strand river capital planning, flow situation like low-flow in small rivers used to be calculated as the ratio of area by flow situation in other large rivers. Which is because the small rivers don't have a survey about the small watershed area water-level and discharge. Therefore, this study has examined the low-flow estimation computerizing the dearth water discharge amount in small rivers. The low-flow statistics which is calculated by independent variables of the watershed area(A), the average basin slope(S), the void ratio(V), the volume of pores(V_f), the mountain ratio(M) and the year average amount of rainfall(R) is as follows.       • 태권도 지도자의 이미지가 수련생 인성발달에 미치는 영향

        This research sought aimed to determine the effect of the image of a taekwondo Coach upon the development of his Coachs' personality education. Towards this end, the research surveyed 636 students in high schools and lower schools who lived in Daegu City and continued to practice taekwondo for more than tree months in taekwondo training halls. The data collected was statistically analyzed and the following conclusion was induced. First, higher-rank taekwondo trainees made positive evaluation of their Coachs' attitude, professionalism and work-related image. Also, trainees with longer training periods made positive evaluation of attitude, professionalism, and role-related image. Second, the personality development of taekwondo trainees was affirmatively influenced by the image of their Coachs. In particular, the finding revealed that the image of Coachs' professionalism was the most important factor that had effect on self-composure, concentration, self-respect, leadership and etiquette. Also, the attitude image of Coachs' had positive effects on self-confidence, self-composure, self-respect, the occupation awareness image of Coachs' had positive effects on self-composure, the image related to work had positive effects on concentration, leadership, and etiquette, and the role image of Coach had positive effects on self-composure. Third, the personality development of taekwondo trainees received negative effects due to their Coachs' image. Likewise, the finding revealed that the occupation awareness image of Coachs were factors that had negative effects on Coachs' self-confidence, concentration, leadership and etiquette. Also, the professionalism image of Coachs had negative effects on self-confidence, and the image related to work had negative effects on self-composure.

      • 복합물질로 오염된 실트지반의 안정관리 방법에 관한 연구

        This study investigates general movements about the lateral flow through model tests in case the unsymmetrical surcharge is worked on polluted soft silt, and compared the results with the existing theories. The critical surcharge at the point of time when the lateral flow occurs, was q_(cr) = 11.15C_(u)-0.34 and the ultimate surcharge was decided on q_(ult) = 18.55C_(u)-0.52. The formula of the maximum lateral flow pressure of the polluted soft silt was P_(max) =-216q_(ult)²+1.22q_(u1t)+0.29. It was shown that the maximum value of lateral flow pressure occurred mainly at 0.3∼0.4H thick of soil layers and the lateral flow pressure of the earth surface (P_(0)= P_(max)/3) has been show in the pattern of quadrilateral distribution. The displacement obtained in the model soils was applied to the control diagram presented by Matsuo·Kawamura as follows ; lateral displacement / settlement(Ym/Sv) on x-axis was applied to the settlement on y-axis. And the result was that the destructive surcharge according to the standard of Matsuo·Kawamura showed a slightly less value than the ultimate surcharge and by the formula of settlement of the ultimate surcharge obtained in the eight model soils, a regressive analysis of eight values of experiments was carried and the result was Sv=-3.89(Ym/Sv) +7.17.

        This study was performed to investigate the effect of olive oil diet on the immune response in ICR male mice. Experimental diets of 4 groups were fed ad libitum to the ICR male mice for 27 days. The results of this study were summarized as followings: 1. 10% Olive oil diet group as compared with the control diet group significantly decreased liver weight rate but significantly increased hemagglutination titer (HA), Arthus reaction, delayed type hypersensitivity reaction (DTH), rosette forming cell (RFC), and phagocyte activity. 2. 20% Olive oil diet group as compared with the control diet group significantly increased body weight gain, liver weight rate, and HA but significantly decreased Arthus reaction, DTH, RFC, phagocyte activity, and peripheral circulating white blood cell (WBC). 3. 30% Olive oil diet group as compared with the control diet group significantly increased liver weight rate but significantly decreased body weight gain, Arthus reaction, plaque forming cell (PFC), DTH, RFC, phagocyte activity, and WBC. The results showed that the increase of olive oil doses significantly decreased humoral and cellular immune responses, phagocyte activity, and WBC.
