        Effects of Tween 80 and Fibrolytic Enzymes on Ruminal Fermentation and Digestibility of Feeds in Holstein Cows

        The effects of the nonionic surfactant Tween 80 and a mixture of fibrolytic enzymes on total tract digestion, in situ disappearance (ISD) and ruminal fermentation characteristics of orchardgrass hay and barley grain were investigated in a 4${\times}$4 Latin square experiment with 4 non-lactating Holstein cows and 4 diets in 4 periods. Cows were offered a total mixed ration of 50% rolled barley grain and 50% orchardgrass hay treated with either 1) water (control), 2) 0.2% (vol/wt) Tween 80, 3) 0.2% (vol/wt) hydrolytic enzyme, or 4) 0.2% hydrolytic enzyme plus 0.2% Tween 80. Total tract digestibility coefficients of DM, nitrogen, NDF and ADF were not affected (p>0.05) by dietary treatment. Compared to the control, the rate of ISD of DM from orchardgrass hay was faster (p<0.05) in cows receiving diets treated with the enzyme alone or with enzyme plus Tween 80 (0.06/h vs. 0.076 and 0.069/h). The rate of digestion was lower (p<0.05) as compared to control when barley grain was treated with these additives. Ruminal fluid pH and concentrations of total VFA, acetate, isobutyrate and butyrate were not affected (p>0.05) by treatments. Cows that consumed diets treated with enzyme plus Tween 80 had higher (p<0.05) ruminal concentrations of propionate and isovalerate, and lower (p<0.05) acetate:propionate ratios. Compared to the control, microbial protein synthesis tended (p = 0.13) to increase with the addition of enzyme to the diet while nonammonia nitrogen flow to the duodenum increased (p<0.05) with both enzyme and Tween 80 treatments. The study indicated that fibrolytic enzymes alone or in combination with Tween 80 could enhance ISD of orchardgrass hay and ruminal concentrations of propionate, valerate and iso-valerate, but do not affect total tract digestibility.

        Effect of Lactobacillus buchneri 40788 and Buffered Propionic Acid on Preservation and Nutritive Value of Alfalfa and Timothy High-moisture Hay

        The effects of Lactobacillus buchneri 40788 and buffered propionic acid on preservation, intake and digestibility of alfalfa (Medicago sativa) and timothy (Phleum pratense) hay were investigated. During baling, forages were treated with L. buchneri 40788 (1.2${\times}$10$^6$ CFU/g) as a liquid (LLB) or as a granular preparation (GLB), with buffered propionic acid (10 mL/kg, BPA), or left untreated (control). Triplicate 500 kg round bales of each treatment were put up at two moisture levels for each forage: 17%${\pm}$0.33% and 20%${\pm}$0.30% for timothy and 17%${\pm}$0.20% and 19%${\pm}$0.27% for alfalfa (mean${\pm}$SD). Bales were sampled for chemical and microbiological analyses after 0, 30 and 60 d of storage. Compared to controls, all preservatives reduced (p<0.05) heating of both forages at all moisture levels with the exception of alfalfa baled at 19% moisture. After 60 d of storage, GLB reduced (p<0.05) moulds in 17% timothy hay as compared to other treatments, but at 20% moisture, moulds were reduced in LLB- and BPA-treated timothy as compared to controls. In alfalfa at 17% moisture, total bacteria were lower (p<0.05) in GLB-treated bales than LLB or control bales, but yeast and total bacteria were only reduced in BPA-treated alfalfa at 19% moisture. In situ DM disappearance of timothy (both moisture levels) and alfalfa (19% moisture level) increased (p<0.05) with LLB treatment compared to control. Digestibility of both forages did not differ (p>0.05) among treatments, however, voluntary DM intake of LLB-treated timothy (1.32 kg/d) was 22.3% higher (p<0.05) than control, and 14.1% higher than BPA-treated timothy. Treating timothy and alfalfa hay with L. buchneri 40788 or buffered propionic acid may improve the nutritive value of the hay when baled at 17 to 20% moisture.

        Effects of Homolactic Bacterial Inoculant Alone or Combined with an Anionic Surfactant on Fermentation, Aerobic Stability and In situ Ruminal Degradability of Barley Silage

        The effect of a homolactic inoculant containing a blend of Lactobacillus plantarum, Pediococcus acidilactici and Enterococcus faecium or, the anionic surfactant, sodium dodecyl sulphate (SDS), alone or in combination on fermentation characteristics, aerobic stability and in situ DM, OM and NDF degradability of barley silage was investigated. Barley (Hordeum vulgare, L.) was harvested (45% DM), chopped and treated with water at 24 ml/kg forage (Control), inoculant at $1.09{\times}10^5$ cfu/g forage (I), SDS at 0.125% (wt/wt) of forage (S) or with the inoculant ($1.09{\times}10^5$ cfu/g) plus SDS (0.125% wt/wt; I+S). The treated forages were ensiled in triplicate mini silos and opened for chemical and microbiological analyses on d 1, 2, 3, 7, 14, 42 and 77. Silage samples from d 77 were opened and aerobically exposed for 7 d. The in situ rumen degradability characteristics of silage DM, OM and NDF were also determined. The terminal concentration of NDF in S and I+S was lower (p<0.001) than in other treatments. Lactate concentration was higher (p<0.001) and the rate and extent of pH decline were greater (p<0.001) in I and I+S than S and Control silages. A homolactic pathway of fermentation in I and I+S was evidenced by reduced (p<0.001) water-soluble carbohydrates concentration, higher lactate (p<0.01), lower acetate (p<0.01) and lower pH values (p<0.001) than in S and Control silages. All silages remained stable over 7 d of exposure to air as indicated by lower temperatures and moulds, and by non-detectable yeast populations. The treated silages had lower DM and OM degradability than in the Control but NDF degradation characteristics of I+S were improved compared to other treatments. It is concluded that the inoculant alone improved the fermentation characteristics whereas the combination of the inoculant with SDS improved both fermentation and NDF degradability of barley silage.

        Effects of Microbial Inoculants on the Fermentation, Nutrient Retention, and Aerobic Stability of Barley Silage

        Fermentation characteristics, nutrient retention and aerobic stability of barley silages prepared using 6 commercial inoculants were evaluated using 126 mini-silos (3-L) in a completely randomized design. Whole barley forage was chopped, wilted to 39% DM and treated with water (control, S) or one of six inoculants: A (containing Lactobacillus plantarum); B (L. plantarum and Enterococcus faecium); C (L. plantarum and Pediococcus cerevisiae); D (L. plantarum, Pediococcus pentosaceus and Propionibacterium freudenreichii, plus hydrolytic enzymes); E (Lactobacillus buchneri plus hydrolytic enzymes); F (L. buchneri and P. pentosaceus plus hydrolytic enzymes). Samples of treated forage were collected for analysis at the time of ensiling, and then 18 silos of each treatment were filled, capped and weighed. Triplicate silos were weighed and opened after 1, 3, 5, 7, 33, and 61 d. On d 61, $400{\pm}5g$ of material from each silo was placed in 1-L styrofoam containers, covered with cheesecloth and held at room temperature. Silage temperature was recorded hourly for 14 d via implanted thermocouple probes. Chemical composition of the forage at ensiling was consistent with previously reported values. At d 61, pH was lowest (p<0.01) in silage S. Ammonia-N was lower (p<0.05) in silage A than in silages S, B, E, or F. Compared to pre-ensiling values, water soluble carbohydrate concentrations were elevated in silages S, A, B, C and D, and decreased in E and F. Lactic acid concentrations were similar (p>0.10) across treatments. Acetic acid levels were highest (p<0.01) in silage E and lowest (p<0.01) in silage D. Recovery of DM was lower (p<0.01) in silage F than in silages S, A, B, C, or D. On d 61, yeasts were most numerous (p<0.01) in silage D, which was the only silage in which temperature rose more than $2^{\circ}C$ above ambient during aerobic exposure. Silage D also had the highest (p<0.01) pH and ADIN content after aerobic exposure. Lactic acid and WSC content of silage D decreased dramatically during the 14-d aerobic exposure period. Yeast counts (at d 14 of exposure) were lowest (p<0.01) in silages E and F. In general, the commercial inoculants did not appear to enhance the fermentation of barley silage to any appreciable extent in laboratory silos.

        Soil organic carbon stocks in three Canadian agroforestry systems: From surface organic to deeper mineral soils

        <P><B>Abstract</B></P> <P>Our understanding of the effect of agroforestry systems on soil organic carbon (SOC) is largely limited to the upper layer of the mineral soil, while LFH (litter, partially decomposed litter and humus) and deeper soil layers are poorly studied. In this study, the effects of three different agroforestry systems (hedgerow, shelterbelt, and silvopasture) and their component land-cover types (treed area and adjacent herbland) on SOC stock in LFH and mineral soil layers (0–75 cm) were investigated across 36 sites in central Alberta, Canada. The SOC stock of mineral soil (0–75 cm) was not affected by agroforestry systems but by land-cover type. The treed area had greater (<I>p</I> < 0.001) SOC in the 0–75 cm mineral soil (25.5 kg C m<SUP>−2</SUP>) than the herbland (19.4 kg C m<SUP>−2</SUP>), driven by the greater (<I>p</I> < 0.001) SOC level in the top 0–30 cm rather than that in the deeper layers (30–75 cm). Within the treed area, the silvopasture system that was dominated by broad-leaf deciduous trees had 56–70% more SOC in the 0–10 cm soil than in the hedgerow and shelterbelt systems. The SOC stock in the 0–10 cm layer was positively (<I>p</I> = 0.025) related to the C stock of the overlying LFH layer in the silvopasture system. These results together with the 22–24% higher dissolved organic carbon (DOC) concentration in the silvopasture than in the other systems suggest that the greater SOC stock in the 0–10 cm mineral soil could be attributed to the higher rates of translocation of DOC from the LFH in the silvopasture than that in shelterbelt or hedgerow. We conclude that SOC stock in the top mineral soil (e.g., 0–30 cm) is more responsive to changes in land-cover type and the LFH layer plays an important role in increasing SOC stock in the surface mineral soil of the agroforestry systems in central Alberta.</P> <P><B>Highlights</B></P> <P> <UL> <LI> In agroforestry systems, treed area had greater SOC than herbland. </LI> <LI> Silvopasture had greater SOC than hedgerow and shelterbelt. </LI> <LI> LFH layer played a critical role in increasing SOC in mineral layer. </LI> <LI> SOC storage in 0–30 cm mineral layer was most responsive to land-cover type. </LI> </UL> </P>

        Effects of Tween 80 on In Vitro Fermentation of Silages and Interactive Effects of Tween 80, Monensin and Exogenous Fibrolytic Enzymes on Growth Performance by Feedlot Cattle

        The effects of monensin, Tween 80 and exogenous fibrolytic enzymes on ruminal fermentation and animal performance were studied in vitro and in vivo. In Expt 1, the effects of the surfactant Tween 80 (0.2% wt/wt, DM basis) on ruminal fermentation of alfalfa, corn and orchardgrass silages were investigated using in vitro gas production techniques. Tween 80 did not affect (p>0.05) cumulative gas production at 24 h, but it reduced (p<0.05) the lag in fermentation of all three silages. With corn silage and orchardgrass silage, gas production rates and concentrations of total volatile fatty acids (VFA) were increased (p<0.05) by Tween 80; with alfalfa silage, they were reduced (p<0.05). Tween 80 increased (p<0.05) the proportion of propionate in total VFA, and reduced (p<0.05) acetate to propionate ratios (A:P) with all three silages. In Expt 2, exogenous fibrolytic enzymes (E; at 0, 37.5 or 75 g/tonne DM), monensin (M; at 0 or 25 ppm and Tween 80 (T; at 0 or 2 L/tonne DM) were added alone or in combination to backgrounding and finishing diets fed to 320 crossbred steers in a feeding trial with a $3{\times}2{\times}$2 factorial arrangement of treatments. The backgrounding and finishing diets contained barley grain and barley silage in ratios of 57.8:42.2 and 93.5:6.5 (DM basis), respectively. Added alone, none of the additives affected DM intake (p>0.1) in the backgrounding or in the finishing period, but interactive $M{\times}T$ effects were observed in the finishing period (p=0.02) and overall (p=0.04). In the finishing period, T without M tended to reduce DM intake (p=0.11), but T with M increased (p=0.05) DM intake. Monensin increased average daily gain (ADG) during backgrounding (p=0.07) and finishing (p=0.01), and this ionophore also improved overall feed efficiency (p=0.02). Warm carcass weight was increased (p<0.001) by M, but dressing percentage was reduced (p=0.07). In the backgrounding period, T increased ADG by 7% (p=0.06). Enzymes increased (p=0.07) ADG by 5 and 6% (low and high application rates, respectively) during backgrounding, but did not affect (p>0.10) ADG during finishing, or overall feed efficiency. Whereas T enhanced the positive effects of M on ADG during backgrounding (p=0.04) and overall (p=0.05), it had no impact (p>0.1) on the effects of E. Interactions between M and T suggest that the surfactant may have potential for enhancing the positive effects of monensin on beef production, but this requires further research.

        Methane Production of Different Forages in In vitro Ruminal Fermentation

        An in vitro rumen batch culture study was completed to compare effects of common grasses, leguminous shrubs and non-leguminous shrubs used for livestock grazing in Australia and Ghana on $CH_4$ production and fermentation characteristics. Grass species included Andropodon gayanus, Brachiaria ruziziensis and Pennisetum purpureum. Leguminous shrub species included Cajanus cajan, Cratylia argentea, Gliricidia sepium, Leucaena leucocephala and Stylosanthes guianensis and non-leguminous shrub species included Annona senegalensis, Moringa oleifera, Securinega virosa and Vitellaria paradoxa. Leaves were harvested, dried at $55^{\circ}C$ and ground through a 1 mm screen. Serum bottles containing 500 mg of forage, modified McDougall's buffer and rumen fluid were incubated under anaerobic conditions at $39^{\circ}C$ for 24 h. Samples of each forage type were removed after 0, 2, 6, 12 and 24 h of incubation for determination of cumulative gas production. Methane production, ammonia concentration and proportions of VFA were measured at 24 h. Concentration of aNDF (g/kg DM) ranged from 671 to 713 (grasses), 377 to 590 (leguminous shrubs) and 288 to 517 (non-leguminous shrubs). After 24 h of in vitro incubation, cumulative gas, $CH_4$ production, ammonia concentration, proportion of propionate in VFA and IVDMD differed (p<0.05) within each forage type. B. ruziziensis and G. sepium produced the highest cumulative gas, IVDMD, total VFA, proportion of propionate in VFA and the lowest A:P ratios within their forage types. Consequently, these two species produced moderate $CH_4$ emissions without compromising digestion. Grazing of these two species may be a strategy to reduce $CH_4$ emissions however further assessment in in vivo trials and at different stages of maturity is recommended.

        Effects of Surfactant Tween 80 on Enzymatic Accessibility and Degradation of Orchardgrass (Dactylis glomerata L.) at Different Growth Stages

        The study evaluates the enzymatic dry matter (DM) degradability and water holding capacity of leaf and stem fractions of orchardgrass (Dactylis glomerata L.) at different growth stages with or without the presence of surfactant Tween 80. While Tween 80 significantly (p<0.05) increased water and enzyme holding capacities in the leaf blades fraction, less was observed in the fraction of leaf sheath and stem of orchardgrass. The enzyme holding capacity in the leaves was also altered more than that for water holding capacity. This resulted in the increased rate and extent of enzymatic hydrolysis of the leaf blade fractions at two growth stages, whereas little was with leaf sheath and stem fractions. It was also observed that at 0.005% concentrations of Tween 80 the enzymatic DM degradability of young leaf blades was higher (p<0.05) by 20-30% compared to that of the control, as well as for water and enzyme holding capacity. For matured leaf blades the DM degradability were increased with over 0.01% concentrations of the surfactant, but the increase was less than leaf blades of young orchardgrass. This result suggests the possibility of using the surfactant Tween 80 to improve forage digestibility in the rumen.

        In Vitro Activity of Extract and Fractions of Natural Cocoa Powder on Plasmodium falciparum

        Several flavonoids isolated from certain plants have demonstrated antiplasmodial activity, after their initial indigenous use in malaria treatment. Cocoa has been found to be a rich food source of flavonoids in comparison with many common foods and beverages. The aim of this work was to investigate the in vitro activity of natural cocoa powder on the growth of Plasmodium falciparum. Prepared crude methanol extract was partitioned successively with petroleum ether, ethyl acetate,chloroform, and butanol. Total flavonoid concentration in the crude methanol extract and fractions was measured by the AlCl3colorimetric assay. Direct inhibitory activity of the natural cocoa powder was assessed by culturing extract and fractions with P. falciparum in vitro. Greater antiplasmodial activity was observed in nonpolar solvent fractions (chloroform, ethyl acetate, and petroleum ether) compared with polar solvents. The chloroform fraction was most active, with mean – SEM 50% and 90%inhibition concentrations of 48.3 – 0.9 and 417 – 7.8 lg/mL, respectively. The study showed a weak association between total flavonoid concentration and antiplasmodial activity. Early trophozoite (ring-stage) synchronized cultures treated with the chloroform fraction of natural cocoa powder showed a decline in growth. Further reduction in parasitemia was also observed for other erythrocytic stages. These results suggest that natural cocoa powder has measurable direct in vitro inhibitory effect on P. falciparum and support the anecdotal reports of its ability to prevent malaria as a result of regular intake as a beverage.
