소나무재선충(Pine wood nematode, PWN)은 아시아와 유럽의 소나무류들에 침입하여 고사시키는 심각해 병원성 선충이다. 가장 완벽한 방제 방법은 감염목을 소각/분쇄의 방법으로 PWN 감염목을 제거하는 것이다. 현재 PWN의 다른 종과 차이를 가지는 유전자서열을 이용한 진단 방법이 다양하게 개발되어져서 실험실에서의 종 판명은 가능한 실정이다. 하지만, 대부분의 분자진단방법은 소나무 목편에서부터의 시료 추출, 그리고 분자진단 시약과 혼합하고, PCR을 이용하여 증폭을 진행 한 후, 결과를 확인하는 과정이 필요하다. 그러므로, 분자진단법을 현장에 적용하기 위해서는 향 후 많은 노력이 필요하다. 이에 반하여 항원/항체 반응을 이용한 진단 방법의 경우는 항원 추출 후, 일정량을 신속진단키트에 떨어 뜨려서 반응을 확인하는 과정을 거치므로, 전문적인 지식이나 기술이 필요하지 않다. 하지만, PWN과 PWN 감염목에 특이적인 항원이 밝혀지지 않은 상황이다. 최근에 우리는 PWN단백질 중에서 분비되는 Aspartic peptidase 1 (ASP1)을 항원으로 선정을 하고, 전장단백질을 Baculovirus Expression System으로 발현을 하였다. PWN-ASP1을 항원으로 단클론항체를 분비하는 세포 주를 확립하였다. 이러한 단클론항체는 향후, PWN의 소나무 침입에 관한 연구와 신속진단키트의 개발에 활용될 수 있을 것이다. (본 연구는 국립산림과학원의 연구비 지원으로 진행되었음)

The biochemical characteristics of pine wood nematode (PWN)-secretory antigen 571 (PWN-SA571) identified from the PWN secretome and PWN-infected pine tree extracts (PWNIPT) were investigated. PWN-SA571 has homologs in various nematodes in diverse families and a signal peptide at the amino-terminal end. Since it was not possible to express the full-length PWN-SA571 protein, two peptides showing high antigenicity were synthesized and used as antigens for generating monoclonal antibodies (Mabs). Mab-secreting fusion hybridoma cell lines were selected based on immunoreactivity to free peptide and BSA-conjugated peptide (BSA pep) antigens by enzyme-linked immunosorbent assay for establishing Mab-secreting hybridoma lines. Ascites containing PWN-SA571-specific Mabs showed stronger immunoreactivity to PWNIPT than to healthy pine tree extracts. In addition, PWN-SA571-specific Mabs could recognize less than 20 ng of BSA pep in lateral flow assays. Furthermore, purified biotin-conjugated PWN-SA571-specific Mab IgG or IgM were able to detect BSA pep (< 15 ng) and PWN extracts (< 600 ng). Taken together, these results demonstrate that PWN-SA571-specific Mabs could detect PWN or PWNIPT extracts by ELISA, LFA, or dot blot analysis.

Red seaweed Chondracanthus canaliculatus, anunderexploited algae species, was used as a potentialsource for the obtaining of carrageenan. Seaweed wastreated under alkaline conditions using ultrasound alone orcombined with conventional procedures, to improve theyield extraction. Color, syneresis behavior, water retentioncapacity, and functional groups of the gelling and nongellingfractions of carrageenan were determined; theseproperties were compared with those of commercial carrageenansnamed A and B. Ultrasound alone or with heatsignificantly (p\0.05) increased the yield extraction up to41–45% and influenced color parameters, in comparisonwith conventional treatments. Functional groups kappa andiota, and alginates, were confirmed in both carrageenanfractions. Syneresis behavior was well fitted to a thirddegreepolynomial equation within days 1 to 6, after which,it reached a plateau. While, the use of ultrasound at roomtemperature gave carrageenan properties more similar tothose of the commercial carrageenan type A.
The Modernization of Khasanskii Raion in the Russian Far East: Potential, Problems, and Perspectives

In our article, we discuss development issues in a strategically important Russian territory-Khasanskii Raion (district). We focus on the implementation of infrastructure projects within the framework of establishing the International Transportation Corridor Primor’e-2, and on subjective assessments of modernization in terms of the common good. We conclude that an assessment of the rhetoric and practice of proposed projects reveals a close relationship with other experiences of post-Soviet transformation, showing that modernization itself is largely selective, nonsystemic, and controversial in its social consequences.
Metronidazole in the Management of Post-Open Haemorrhoidectomy Pain: Systematic Review

Purpose: Open haemorrhoidectomy is associated with significant postoperative pain. Metronidazole is commonly prescribed in the postoperative period as an adjunct to analgesia in pain management. Methods: In our systematic review, studies were identified using PubMed/MEDLINE, Embase/Ovid and Cochrane Register of Controlled Trials databases. Studies were included if they were randomised controlled trials (RCTs) involving interventions with oral metronidazole at any dose over any time period. The primary outcome was pain score (visual analogue scale, VAS) after open haemorrhoidectomy. Secondary outcomes included time to return to normal daily activities, additional analgesia usage, and postoperative complications. Results: Of 14 RCTs reviewed, 4 met inclusion criteria and were selected. The studies comprised 336 study subjects and 169 subjects were randomised to metronidazole while 167 were in the control group. There was a significant reduction in VAS across all time points, with maximal reduction seen on day 5 posthaemorrhoidectomy (mean difference, -2.28; 95% confidence interval, -2.49 to -2.08; P < 0.001). There was no difference in incidence of complications (P = 0.13). The Cochrane Risk of Bias Tool showed 3 of 4 of the studies had a risk of bias. Conclusion: Metronidazole may be associated with decreased pain but there is insufficient evidence from RCTs to provide a strong grade of recommendation. Further RCTs are required.
