        Evaluation of Antioxidant and Free Radical Scavenging Capacities of Some Nigerian Indigenous Medicinal Plants

        Methanolic extracts of 10 selected Nigerian medicinal plants—Psidium guajava, Alstonia boonei, Cassia alata, Newbouldia laevis, Spondias mombin, Globimetula cupulatum, Chromolaena odorata, Securidaca longepedunculata, Ocimum gratissimum, and Morinda lucida—widely used in ethnomedicine, were assessed for phytochemical constituents and antioxidant and free radical scavenging activities using seven different antioxidant assay methods. Phytochemical screening gave positive tests for steroids, terpenoids, and cardiac glycosides, alkaloids, saponins, tannins, and flavonoids contained in the extracts. P. guajava contained the highest amount of total phenolics (380.08±4.40mg/L gallic acid equivalents), and the highest amounts of total flavonoids were found in the leaf extracts of C. alata (275.16±1.62μg/mL quercetin equivalents [QE]), C. odorata (272.12±2.32μg/mL QE), and P. guajava (269.72±2.78μg/mL QE). Percentage 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity was highest in S. mombin (88.58±3.04%) and P. guajava (82.79±2.84%) and compared with values obtained for ascorbic acid and gallic acid. All the extracts, generally, had low nitric oxide radical scavenging activities, and G. cupulatum had the highest hydroxyl radical scavenging activity (63.84±0.97%). The extracts in general demonstrated high lipid peroxidation inhibitory activity, with only M. lucida (38.74±1.99%) and A. boonei (47.16±0.59%) being exceptions. The reductive potential was highest in P. guajava (0.79±0.04) and least in S. longepedunculata (0.26±0.00). DPPH assay correlated well with total phenolic contents (r2=0.76) and reductive potential (r2=0.81) and fairly with lipid peroxidation inhibitory activity (r2=0.51). There was a good correlation between total phenolic contents and reductive potential (r2=0.79) and a fair correlation between total phenolic contents and lipid peroxidation inhibitory activity (r2=0.55). These results suggest that the methanolic extracts of the studied plant parts possess significant antioxidant and radical scavenging activities that may be due to the phytochemical content of the plants and as such make them potential candidates as natural chemoprophylactic agents. In addition, multiple assay methods should be used in comparing antioxidant capacities of samples to have a reliable result.

        Local Condiment, Iru, Obtained from the Fermentation of Parkia biglobosa Seed Substantially Reduced the Serum Cholesterol Level of Wister Rats

        Hypercholesterolaemia is one of the risk factors in coronary heart disease. Hence, this research was designed to investigate the effect of iru on cholesterol levels in hypercholesterolaemic rats. The fermented condiment iru was produced naturally and with starter cultures of Lactobacillus plantarum, Bacillus subtilis, and Leuconostoc sp.. The hypercholesterolaemic rats were fed a diet supplemented with iru for 28 days, and total cholesterol, triglyceride (TG), high density lipoproteins (HDL), and low density lipoproteins (LDL) were determined before and after feeding. Cholesterol levels in hypercholesterolaemic rats (100.80 mg/dL) were reduced to 56.99∼80.21 ㎎/dL after feeding with iru supplementation while rats not placed on the iru diet had cholesterol levels of 119 ㎎/dL. There were also significant reductions (P<0.05) in serum TG (78.77∼32.57 ㎎/dL) and LDL (28.43∼6.63 ㎎/dL) levels in rats fed the iru diet compared with the control (63.36 ㎎/dL). Higher and significantly different (P<0.05) HDL was found in rats fed with iru fermented with L. plantarum (44.01 ㎎/dL) while the least was found in the untreated group (28.93 ㎎/dL). The results from this study suggest that supplementing the diet with iru obtained by the fermentation of Parkia biglobosa seeds may significantly reduce cholesterol level in the blood stream.

        Tetrapleura tetraptera curtails oxidative and proinflammatory biochemical events in lithium-pilocarpine model of status epilepticus

        Tetrapleura tetraptera Taub. (Fabaceae), commonly known as Aidan is ethnopharmacologically used for the management of health conditions such as diabetes mellitus, leprosy, epilepsy, and stroke in Nigeria. This study evaluated the anti-oxidoinflammatory properties of T. tetraptera methanol leaf extract (TTE) in lithium-pilocarpine-induced status epilepticus in Wistar rats. The extract was phytochemically screened and HPLC fingerprinting was performed. Animals were intraperitoneally administered with 127 mg/kg lithium chloride followed by 25 mg/kg pilocarpine 20 h later to induce status epilepticus. The animals were post treated with 50, 100 and 200 mg/kg TTE with 10 mg/kg valproic acid as the reference standard drug. Phytochemical screening of TTE confirmed the presence of tannins, phenols, alkaloids, terpenoids, flavonoids, saponins, glycosides and steroids. Quantitative phytochemical and antioxidant analyses of the extract indicated significant in vitro antioxidant and radical scavenging activity. HPLC analysis of the extract confirmed the presence of aridanin and polyphenols. TTE ameliorated redox imbalance by increasing markers for oxidative stress such as ferric reducing antioxidant power, glutathione level, catalase, glutathione S-transferase and superoxide dismutase activities. Moreover, TTE ameliorated pro-inflammatory events by reducing the level of the pro-inflammatory mediator nitric oxide, attenuating lipid peroxidation (which produces inflammatory lipid peroxidation-derived aldehydes), and decreasing the activities of xanthine oxidase and lactate dehydrogenase in the brain. These results indicated that the leaf of Tetrapleura tetraptera has therapeutic potential against status epilepticus by reversing oxidoinflammatory events. Tetrapleura tetraptera leaf extracts could be used to produce novel plant-based pharmaceuticals for treating status epilepticus and associated disorders.
