Dissemination and tracking of Salmonella spp. in integrated broiler operation

Controlling Salmonella in integrated broiler operation is complicated because there are numerous potential sources of Salmonella contamination, including chicks, feed, rodents, wild poultry operations, and the processing plant. The objective of this study was to investigate the distribution of Salmonella through all phases of two integrated broiler operations and to determine the key areas related to the control of all known sources of infection. Two different Salmonella serotypes were observed at integrated broiler chicken company A. S. enteritidis, the predominant company A isolate, was consistently found in the breeder farm, hatcheries, broiler farms, and chicken slaughterhouse. At company B, a total of six different serotypes, S. heidelberg, S. senftenberg, S. enteritidis, S. blockley, S. gallinarum, and S. virchow, were detected. Although S. heidelberg was not found in the broiler farms, it was consistently found in the breeder farm, hatcheries, and chicken slaughterhouse. In addition, S. enteritidis was found in the hatcheries, broiler farm, and chicken slaughterhouse. In order to obtain the genetic clonality, 22 S. enteritidis isolates were digested with XbaI and analyzed by pulsed-field gel electrohporesis (PFGE). A difference in the PFGE pattern was found to be related to the origin of the integrated broiler operation. These data support the critical need to control Salmonella in breeder farms and hatcheries, and demonstrate important points related to the control of infection in large-scale poultry operations of Korea.

Objective: This study aimed to investigate tissue- and serum-based prognostic biomarkers of high-grade serous ovarian carcinoma (HGSOC) through proteomic analysis. Methods: We conducted label-free liquid chromatography-mass spectrometry using chemotherapy-nave, fresh-frozen primary HGSOC specimens(n=12) and compared results between a favorable prognosis group(PFS ≥18 months) and a poor prognosis group (PFS <18 months). We also conducted proteomic profiling of serum samples obtained from chemotherapy-nave, HGSOC patients(n=20). Differentially expressed serum proteins between two groups were compared. Candidate protein biomarkers were validated via immunohistochemistry(IHC) and ELISA using an independent set of chemotherapy-nave primary HGSOC specimens (n=107) and serum samples (n=100), respectively. Results: Among 658 differentially expressed proteins, 288 proteins were upregulated in favorable prognosis group and 370 proteins were upregulated in other group. Using hierarchical clustering, we selected AAT, NFKB, PMVK, VAP1, FABP4, PF4, APOA1, and AGP for further validation via IHC. Survival analyses revealed that high expression of AAT, NFKB, and PMVK were independent biomarkers for favorable PFS. High expression of VAP1, FABP4, and PF4 were identified as independent biomarkers for poor PFS. We constructed models predicting 18-month PFS by combining clinical variables and IHC results. The optimal model was based on initial serum CA-125, germline BRCA1/2 mutations, residual tumors after surgery, FIGO stage, and expression levels of the six proteins. Among six biomarkers, AAT and FABP4 were quantified in serum samples by ELISA. Dose-dependent prognostic significance of each proteins concentration for PFS was observed. A model consisting of serum AAT and FABP4 concentrations and clinical variables showed good performance in predicting poor prognosis groups. Conclusion: The present results elucidate the proteomic landscape of HGSOC and tissue- and serum-based protein biomarkers to predict the prognosis of HGSOC.
Charge-Directed Peptide Backbone Dissociations of o-TEMPO-Bz-C(O)-Peptides

In the present study, we report that the charge-directed (assisted) peptide dissociation products, such as b- and y-type peptide backbone fragments, were the major products in MS/MS and MS3 applications of some o-TEMPO-Bz-C(O)-peptide ions, while radical-driven dissociation products, such as a/x and c/z-type fragments, were previously shown to be the major products in the free radical initiated peptide sequencing mass spectrometry (FRIPS MS). Those o-TEMPO-Bz-C(O)-peptides share a common feature in their sequences, that is, the peptides do not include an arginine residue that has the highest proton affinity among free amino acids. The appearance of b- and y-type fragments as major products in FRIPS MS can be understood in terms of the so-called “mobile-proton model”. When the proton is highly mobilized by the absence of arginine, the chare-directed peptide dissociation pathways appear to be more competitive than the radical-driven dissociation pathways, in our FRIPS experiments.

The family of ClpB protein is a molecular chaperone which protects cellular proteins from being aggregated upon exposure to severe environmental stresses in association with DnaK/DanJ/GrpE in the ATP-dependent manner. In a psychrophilic bacterium which survives at a subzero temperature, any functional role of cold-active ClpB protein can be rather crucial. In order to identify a ClpB encoding gene from a cold-adapted bacterium whose genome sequence has not been fully discovered, we have employed a series of PCR technologies, including a gradient PCR with homologous primers,an inverse PCR and a cassette PCR. The full sequence of PaclpB gene was successfully identified and compared with those of other psychrophilic species. We have further cloned the gene in E.coli expression systems and were able to induce PaClpB protein expression by IPTG, which help us understand a molecular mechanism for survival against extremely cold environments.

We investigate the explicit evaluation for the sum ∑(a,b,x,y)∈N(4), ax+by=n, C(x,y) ab in terms of various divisor functions (where C(x,y) is the set of residue conditions on x and y) for various fixed C(x,y). We also obtain some identities and congruences as interesting ap-plications.
