        Effects of Combining Feed Grade Urea and a Slow-release Urea Product on Characteristics of Digestion, Microbial Protein Synthesis and Digestible Energy in Steers Fed Diets with Different Starch:ADF Ratios

        As a result of the cost of grains, the replacement of grains by co-products (i.e. DDGS) in feedlot diets is a common practice. This change produces diets that contain a lower amount of starch and greater amount of fibre. Hypothetically, combining feed grade urea (U) with slow release urea (Optigen) in this type of diet should elicit a better synchrony between starch (high-rate of digestion) and fibre (low-rate of digestion) promoting a better microbial protein synthesis and ruminal digestion with increasing the digestible energy of the diet. Four cannulated Holstein steers ($213{\pm}4$ kg) were used in a $4{\times}4$ Latin square design to examine the combination of Optigen and U in a finishing diet containing different starch:acid detergent fibre ratios (S:F) on the characteristics of digestive function. Three S:F ratios (3.0, 4.5, and 6.0) were tested using a combination of U (0.80%) and Optigen (1.0%). Additionally, a treatment of 4.5 S:F ratio with urea (0.80% in ration) as the sole source of non-protein nitrogen was used to compare the effect of urea combination at same S:F ratio. The S:F ratio of the diet was manipulated by replacing the corn grain by dried distillers grain with solubles and roughage. Urea combination did not affect ruminal pH. The S:F ratio did not affect ruminal pH at 0 and 2 h post-feeding but, at 4 and 6 h, the ruminal pH decreased as the S:F ratio increased (linear, p<0.05). Ruminal digestion of OM, starch and feed N were not affected by urea combination or S:F ratio. The urea combination did not affect ADF ruminal digestion. ADF ruminal digestion decreased linearly (p = 0.02) as the S:F ratio increased. Compared to the urea treatment (p<0.05) and within the urea combination treatment (quadratic, p<0.01), the flow of microbial nitrogen (MN) to the small intestine and ruminal microbial efficiency were greater for the urea combination at a S:F ratio of 4.5. Irrespective of the S:F ratio, the urea combination improved (2.8%, p = 0.02) postruminal N digestion. As S:F ratio increased, OM digestion increased, but ADF total tract digestion decreased. The combination of urea at 4.5 S:F improved (2%, p = 0.04) the digestible energy (DE) more than expected. Combining urea and Optigen resulted in positive effects on the MN flow and DE of the diet, but apparently these advantages are observed only when there is a certain proportion of starch:ADF in the diet.

        Effects of Combining Feed Grade Urea and a Slow-release Urea Product on Performance, Dietary Energetics and Carcass Characteristics of Feedlot Lambs Fed Finishing Diets with Different Starch to Acid Detergent Fiber Ratios

        Recent findings have shown that microbial nitrogen flow and digestible energy of diets are increased when urea is combined with a slow-release urea (SRU) in diets with a starch to acid detergent fibre ratio (S:F) 4:1. This affect is attributable to enhanced synchrony between ruminal N availability for microbial growth and carbohydrate degradation. To verify the magnitude of this effects on lamb performance, an experiment was conducted to evaluate the effects of combining urea and a SRU in diets containing S:F ratios of 3:1, 4:1, or 5:1 on performance, dietary energetics and carcass characteristics of finishing lambs. For that, 40 Pelibuey${\times}$Katahdin lambs ($36.65{\pm}3kg$) were assigned to one of five weight groupings in 20 pens (5 repetition/treatments). The S:F ratio in the diet was manipulated by partially replacing the corn grain and dried distiller's grain with solubles by forage (wheat straw) and soybean meal to reach S:F ratios of 3:1, 4:1 or 5:1. An additional treatment of 4:1 S:F ratio with 0.8% urea as the sole source of non-protein nitrogen was used as a reference for comparing the effect of urea combination vs. conventional urea at the same S:F ratio. There were no treatment effects on dry matter intake (DMI). Compared the urea combination vs urea at the same S:F ratio, urea combination increased (p<0.01) average daily gain (ADG, 18.3%), gain for feed (G:F, 9.5%), and apparent energy retention per unit DMI (8.2%). Irrespective of the S:F ratio, the urea combination improved the observed-to-expected dietary ratio and apparent retention per unit DMI was maximal (quadratic effect, $p{\leq}0.03$) at an S:F ratio of 4:1, while the conventional urea treatment did not modify the observed-to-expected net energy ratio nor the apparent retention per unit DMI at 4:1 S:F ratio. Urea combination group tended (3.8%, p = 0.08) to have heavier carcasses with no effects on the rest of carcass characteristics. As S:F ratio increased, ADG, G:F, dietary net energy, carcass weight, dressing percentage and longissimus thoracis (LM) area increased linearly ($p{\leq}0.02$). Combining urea and a slow-release urea product results in positive effects on growth performance and dietary energetics, but the best responses are apparently observed when there is a certain proportion (S:F ratio = 4:1) of starch to acid detergent fibre in the diet.

        Synthesis, Fastness and Spectral Properties of Some New Azo Pyrazole and Pyrazolotriazole Derivatives

        Coupling of 5-amino-1,3-diaryl-pyrazoles 1a-c with diazonium salts of different aryl amines gave a series of novel 1,3-diaryl-5-amino-4-arylazopyrazoles 3a-l. Such compounds could be also obtained by reaction of 5-amino-1,3-diaryl-4-nitroso- 1H-pyrazoles 2a-c with different aryl amines in alkaline medium. Oxidation of azo derivatives 3a-l with cupric acetate, in dimethyl formamide and stream of air, gave 2,4,6-triaryl-2,4-dihydropyrazolo [4,3-d]-1,2,3-triazoles 4a-l. and the fluorescence properties of the cyclic triazoles were studied. Diazotization of 5-amino-1,3-diaryl-1H-pyrazoles 1a-c by sodium nitrite in ortho-phosphoric acid followed by coupling with some aryl amines gave o-aminoazo compounds 5a-f. Cyclisation of compounds 5a-f in pyridine and cupric acetate gave the corresponding triazoles 6a-f. The coupling of compounds 6a-f with different aryl diazonium salts gave compounds 7a-j. The synthesized dyes were applied to polyesters as disperse dyes and the fastness properties were evaluated. 5-Amino-1,3-diaryl-pyrazoles 1a-c 와 다양한 aryl amine의diazonium salts를 반응시켜서 1,3-diaryl-5-amino-4-arylazopyrazoles 3a-l을 합성하였으며, 몇 가지 화합물은 5-amino-1,3-diaryl-4-nitroso-1H-pyrazoles 2a-c와 aryl amine의 diazonium salts를 반응시켜서 얻었다. 합성한 azo 유도체 화합물 3a-l을 DMF 용매 속에서 cupric acetate와 산화반응시켜서 2,4,6-triaryl-2,4-dihydropyrazolo [4,3-d]-1,2,3-triazoles 4a-l을 합성하였으며, 합성한 cyclic triazoles에 대한 형광 특성을 측정하였다. 한편, Diazotization of sodium nitrite/ortho-phosphoric acid 조건에서 5-amino-1,3-diaryl-1H-pyrazoles 1a-c를 diazotization화 반응시킨 다음에, aryl amines과 반응시켜서 o-aminoazo compounds 5a-f를 합성하였다. 합성한 화합물 5a-f를 pyridine/cupric acetate 반응 조건에서 반응시켜서 triazole 6a-f들을 합성하였으며, 얻어진 화합물 6a-f을 aryl diazonium salts과 반응시켜서 화합물 7a-j을 합성하였다. 합성한 염료 화합물을 polyesters에 분산염료와 정착성을 측정하였다.

        <P>The IL-1 family member 7b (IL-1F7b) is a novel homolog of the IL-1 cytokine family discovered by computational cloning. We have reported that IL-1F7b shares critical amino acid residues with IL-18 and binds the IL-18-binding protein; in doing so, IL-1F7b augments the inhibition of IFN-gamma by the IL-18-binding protein. IL-1F7b also binds IL-18Ralpha but neither induces signal nor acts as a receptor antagonist. Hence, the function of IL-1F7b remains unknown. In the present study, we analyzed the intracellular expression pattern of IL-1F7b. Using two variants of GFP fusion constructs of human IL-1F7b stably expressed in RAW macrophages, only the postcleavage mature form of the IL-1F7b precursor-but not the N-terminal propiece-specifically translocates to the nucleus following LPS stimulation. IL-1F7b, like IL-1beta, IL-18, and IL-33, is processed by caspase-1 to generate the mature cytokines. Therefore, we tested whether caspase-1-mediated cleavage of the IL-1F7b precursor is required for mature IL-1F7b to translocate actively into the nucleus. Indeed, a specific caspase-1 inhibitor markedly reduced nuclear entry of IL-1F7b. In stable transfectants of human IL-1F7b in RAW macrophages stimulated with LPS, levels of TNF-alpha, IL-1alpha, IL-6, as well as the chemokine MIP-2, were substantially reduced (72-98%) compared with LPS-stimulated cells transfected with the empty plasmid. These results demonstrate that IL-1F7b translocates to the nucleus after caspase-1 processing and may act as a transcriptional modulator reducing the production of LPS-stimulated proinflammatory cytokines, consistent with IL-1F7b being an anti-inflammatory member of the IL-1 family.</P>

        Ultrasound and clinical findings from metacarpophalangeal joint assessments in show jumping horses in training

Objectives: The aims of this study were to determine the morphological patterns, physical examination characteristics and ultrasound findings of show jumping horses in training and to establish a score-based examination model for physical and ultrasound follow-ups of metacarpophalangeal joint changes in these animals. Methods: A total of 52 metacarpophalangeal joints from 26 horses who were initially in the taming stage were evaluated, and the horses’ athletic progression was monitored. The horses were evaluated by a physical examination and by B-mode and Doppler-mode ultrasound examinations, starting at time zero (T0), which occurred concomitantly with the beginning of training, and every 3 months thereafter for a follow-up period of 18 months. Results: The standardized examination model revealed an increase in the maximum joint flexion angles and higher scores on the physical and ultrasound examinations after scoring was performed by predefined assessment tools, especially between 3 and 6 months of evaluation, which was immediately after the horses started more intense training. The lameness score and the ultrasound examination score were slightly higher at the end of the study.       • The Shiant Isles Main Sill : Nd and Sr Isotopes Tell the Tale of Two Magmas and Sub-solidus Alteration

        The Shinat Isles Main Sill, of Tertialry age in the Outer hebrides, is a classical example of a composite, differentiated alkaline basic sill. The 170m thick body is composed of four intrusions, oldest to youngest;a 2m olivine teschenite; a 24m thick picrite sill; a subsequent intrusion into the picrite before it was completely solidified producing a 140m thick picrodolerite-crinanite unit; and, a 2m thick granular olivine picrodolerite, emplaced into the upper crinanites befor efull solidification of this host. nd and Sr isotopic study of the sill, from the bottom to top, clarifies the petrogenesis and the extensive sub-solidus alteration by hydrothermal fluids that produced extensive formation of analcime and zeolites. Overall, the initial 87Sr/86Sr ratios show considerable dispersion from about 0.7036 to about 0.7057 while initial 143Nd/144Nd ratios vary from about 0.51252 to 0.51285. Much of the scatter in 87Sr/86Sr for whole-rock samples reflects the uniquitous sub-solidus, aqueous alteration (as deduced by leaching rock samples to remove analcime) during which more radiogenic Sr was introduced into the sill rocks, especially along the margins. in contrast, Nd was immobile during fluid interaction so that the sill 143Nd/144Nd ratios were not affected by the alteratiohn reactions. These values along with the 87Sr/86Sr ratios for leached rock samples outline original magmatic signatures where 87Sr/86Sr and 143Nd/144Nd ratios are inversely and linarly well correlated. Samples cluster at the ends of this trend with only a few in between. The magmatic trend indicates the sill was formed by magmas with two distinct isotopic compositions ofr 143Nd/144Nd and 87Sr/86Sr; a more primitive magma at 0.51285 and 0.7035 that produced the teschenite and picrite, and a more evolved melt at 0.51252 and 0.7048 that produced the picrodolerite-crinanite. There was only very limited mixing of the two magmas which was restricted to the near the contacts between the intrusions and apparently occurred by intercumulus melt migration. The more evolved crinanitc magma was probably produced from the more primitive picritic one by a small degree of crustal contamination and crystal fractionation during a very short interval of residence in the deep crust prior to ascent and emplacement into the picritic intrusion.

        Spectrophotometric evaluation of restorative composite shades and their match with a classical shade guide

        Objectives The aim of this study was to verify the match between 5 shades of composites from different manufacturers with a shade guide and among the systems using a portable spectrophotometer. Materials and Methods Shade measurements were performed on specimens of Z350 XT (3M ESPE), Charisma Diamond (Heraeus Kulzer GmbH), Esthet X-HD (Dentsply Caulk), and Empress Direct (Ivoclar-Vivadent) for shades A1, A2, A3, B1, and C3 using a Vita Easyshade spectrophotometer (Vita Zahnfabrik) against a white background. Corresponding shades of Vitapan Classical (Vita Zahnfabrik) guide were measured likewise and shade variation (ΔE) was calculated based on International Commission on Illumination L*a*b* parameters. The ΔE of the composites in each shade was compared by one-way analysis of variance and Tukey's post hoc test (α = 0.05). Results All composites presented ΔE > 3.7 compared with the shade guide. Variation in shades A3, B1, and C3 was significantly different for all composites. ΔE of Z350 XT was significantly lower for A1 than for the other shades, whereas ΔE of Z350 XT and Charisma Diamond were significantly lower for A2 than for the other shades. Conclusions No composite shade matched with the shade guide. Equivalent shades of the restorative composite from different manufacturers may show clinically noticeable ΔE.         A MICROSTRUCTURAL MODEL OF THE THERMAL CONDUCTIVITY OF DISPERSION TYPE FUELS WITH A FUEL MATRIX INTERACTION LAYER

        This paper describes a finite element model of the microstructure of dispersion type nuclear fuels, which can be used to determine the effective thermal conductivity of the fuels during irradiation. The model simulates a representative region of the fuel as a prism shaped unit cell made of brick elements. The elements within the unit cell are assigned material properties of either the fuel or the matrix depending on position, in such a way as to represent randomly distributed fuel particles with a size distribution similar to that of the as manufactured fuel. By applying an appropriate heat flux across the unit cell it is possible to determine the effective thermal conductivity of the unit cell as a function of the volume fraction of the fuel particles. The presence of a fuel/matrix interaction layer is simulated by the addition of a third set of material properties that are assigned to the finite elements that surround each fuel particle. In this way the effective thermal conductivity of the material may also be determined as a function of the volume fraction of the interaction layer. Work is on going to add fission gas bubbles in the fuel as a fourth phase to the model.

        Mutations in SEC24D, Encoding a Component of the COPII Machinery, Cause a Syndromic Form of Osteogenesis Imperfecta

        As a result of a whole-exome sequencing study, we report three mutant alleles in SEC24D, a gene encoding a component of the COPII complex involved in protein export from the ER: the truncating mutation c.613C>T (p.Gln205<SUP>*</SUP>) and the missense mutations c.3044C>T (p.Ser1015Phe, located in a cargo-binding pocket) and c.2933A>C (p.Gln978Pro, located in the gelsolin-like domain). Three individuals from two families affected by a similar skeletal phenotype were each compound heterozygous for two of these mutant alleles, with c.3044C>T being embedded in a 14 Mb founder haplotype shared by all three. The affected individuals were a 7-year-old boy with a phenotype most closely resembling Cole-Carpenter syndrome and two fetuses initially suspected to have a severe type of osteogenesis imperfecta. All three displayed a severely disturbed ossification of the skull and multiple fractures with prenatal onset. The 7-year-old boy had short stature and craniofacial malformations including macrocephaly, midface hypoplasia, micrognathia, frontal bossing, and down-slanting palpebral fissures. Electron and immunofluorescence microscopy of skin fibroblasts of this individual revealed that ER export of procollagen was inefficient and that ER tubules were dilated, faithfully reproducing the cellular phenotype of individuals with cranio-lentico-sutural dysplasia (CLSD). CLSD is caused by SEC23A mutations and displays a largely overlapping craniofacial phenotype, but it is not characterized by generalized bone fragility and presented with cataracts in the original family described. The cellular and morphological phenotypes we report are in concordance with the phenotypes described for the Sec24d-deficient fish mutants vbi (medaka) and bulldog (zebrafish).

        <P>A search for dark matter particles is performed by looking for events with large transverse momentum imbalance and a recoiling Higgs boson decaying to either a pair of photons or a pair of tau leptons. The search is based on proton-proton collision data at a center-of-mass energy of 13 TeV collected at the CERN LHC in 2016 and corresponding to an integrated luminosity of 35.9 fb(-1). No significant excess over the expected standard model background is observed. Upper limits at 95% confidence level are presented for the product of the production cross section and branching fraction in the context of two benchmark simplified models. For the Z'-two-Higgs-doublet model (where Z' is a new massive boson mediator) with an intermediate heavy pseudoscalar particle of mass m(A) = 300 GeV and m(DM) = 100 GeV, the Z' masses from 550 GeV to 1265 GeV are excluded. For a baryonic Z' model, with m(DM) = 1 GeV, Z' masses up to 615 GeV are excluded. Results are also presented for the spin-independent cross section for the dark matter-nucleon interaction as a function of the mass of the dark matter particle. This is the first search for dark matter particles produced in association with a Higgs boson decaying to two tau leptons.</P>
