        Purification and Characterization of Chinese Cabbage Pectinesterase

        배추로부터 얻은 펙틴에스테라제 조효소액을 DEAE-셀를로오스 이온교환 수지를 통과시켜 두개의 분획F-A와 F-B를 분리했으며. 그중 주종을 이루고 있는 F-A는 DEAE-셀룰로오스 이온교환수지. CM-셀를로오스 이온교환수지를 통과시켜서 약 340배 정제하였다. F-A와 F-B는 비슷한 염효과를 나타내었는데, 최적 염농도는 $Ca^{++}$ $Mg^{++}$등 2가 양이온의 경우 $20{\sim}50mM$ $K^{+}$, $Na^{+}$등 1가 양이온의 경우 250mM부근이었다. 최적pH는 F-A와 F-B에 대해서 각각 8.0 및 $7.5{\sim}8.0$이였으며. 이들 두 분획은 pH $5.0{\sim}8.0$에서 적어도 한 시간동안 안정했다. F-A에 대한 Km은 펙틴기질에 대해서 0.01%이었다. F-A와 F-B의 최적온도는 각각 $48^{\circ}C$와 $55^{\circ}C$이었으나 $55^{\circ}C$에서 급격히 역가가 감소되었다. 열불활성화 경향은 1차반응을 나타내었으며 $35^{\circ}C{\sim}55^{\circ}C$사이에서 F-B의 D-Value는 F-A보다 커서 F-B가 F-A보다 열에 더 안정한 것으로 나타났다. 한편 Z-value는 F-A 의 경우 $10.5^{\circ}C$, F-B의 경우 $8.3^{\circ}C$이었다. Two fractions of pectinesterase from Chinese cabbage were isolated by ammonium sulfate fractionation, ion exchange chromatography on DEAE-cellulose and Sephadex G-150 gel filtration. The fraction F-A and F-B were purified approximately 340- and 10-fold. The similar salt effects and pH optima (pH 7.5-8.0) were obtained for the two pectinesterase fractions. The maximum activity of both two. fractions were obtained at 20-50mM of divalent rations and at 250mM of monovalent rations. The apparent Michaelis constant of the F-A was 0.01% for citrus pectin. The temperature optima for F-A and F-B were $48^{\circ}$ and $55^{\circ}C$, respectively and both fractions were stable in the region of pH 5.0-8.0 at room temperature. The thermal inactivation of the two fractions followed the first order reaction kinetics. From D and Z-values obtained the thermal resistance of the two fractions were characterized.

        BIM 기반 CAFM시스템 구축을 위한 프로세스 개발에 관한 연구

        Communication problems are very popular in the construction industries because participants of the construction projects including designers, contractors, subcontractors, and manufacturers are very diverse and work separately, thereby in consequence making it difficult to organize and reuse the information produced by each participant. Construction industries have been increasingly dependent on Building Information Modeling(BIM) technologies to mitigate the problems. While BIM technologies have been considered as mainly a design tool, many industry professionals have concerns with developing a 3D CAFM (Computer Aided Facility Management) system in conjunction with BIM technologies to enhance efficiency in handling building maintenance. The main objective of this study is to develop a construction process of BIM-based CAFM. The study is composed of three steps including a literature review, a case study for extraction of existing CAFM problems and a development of BIM-based CAFM construction process with Information Delivery Manual.

      • 궤도밀착형 철도용 신호기의 적용에 대한 연구

        철도용 신호기는 주신호기, 종속신호기, 신호부속기로 분류된다. 철도용 신호기는 열차가 소속한 선로의 바로 위 또는 선로의 주변 좌측에 설치되고, 신호기의 사용목적에 따라 특정위치에 설치된다. 하지만 철도시설물이 신호기 인접에 설치됨에 따라 새로 가설되는 구조물에 가려져 승무원이 원거리에서 신호기를 인식하지 못하거나 적색을 띄는 센서 등의 빛과 혼동하는 문제점이 발생될 가능성이 있다. 또한 다수의 레일들 사이마다 유치선 신호기가 설치되어 있는 차량기지에서 열차를 진행시키는 경우, 승무원이 좌우측 신호기를 혼동하여 진입여부를 오인하는 경우가 발생할 가능성이 있다. 본 논문에서는 선로밀착형 철도용 신호기를 제시하고, 승무원의 설문조사를 통해 효과를 입증하였다. There are main signal, subsidiary signal and optional signal in signal flags in railway. The signal flags are installed up or left and right tracks of running trains and are installed in special position according to purposes. However, train crew are not realize the colar of signal flags by installing railway equipment nearby the signal flags. Moreover, it is possible to confuse the color of sense which is red with the color of signal flags. When trains are driven in depots where hold track signals are installed between many rails, it is possible that the train crew confuse the left singal flag with the right signal flag. In this paper, contact to track-type signal flag in railway is presented and the effect of the signal flag is demonstrated through a survey to the train crew.

      • 위치추적 기술을 응용한 검지시스템 적용에 대한 연구

        도시철도 및 철도 운영기관에서 발생하는 안전사고는 심야 유지보수 중에 발생빈도가 높다. 이를 방지하기 위하여 다양한 안전관련 시스템 및 기기개발이 이루어지고 있으나 심야시간에 이루어지는 특수성과 선로 내외 터널에서 작업을 고려한 안전성과 신뢰성을 확보한 시스템은 아직까지 제작되지 못하고 있는 실정이다. 이로 인하여 심야에 유지보수차량(모타카) 운행 중 작업자와의 충돌 추돌사고가 발생하고 있으며 이는 인사사고로 이어져 작업장 안전관리의 문제점과 근무자의 사기를 저하시키는 경우가 빈번히 발생하고 있다. 이에 본 논문에서는 중앙 집중식 통제시스템이 없으며 작업장내 별도의 기기를 설치하지 않고 휴대성과 편의성을 충분히 고려한 RF 모듈레이션 방식을 적용한 거리 측정기술과 근거리 위치추적 기술을 적용한 양방향 검지시스템을 제시하였다. Accidents that occur in urban rail and railway operation agency, there is a high frequency of occurrence in the maintenance of late. To prevent this, the development of systems and equipment associated with various safety have been carried out and to ensure the safety and reliability considering the work on the tunnel and out of line with particularity to be performed in time midnight system is not being produced so far is the reality. Thus, the conflict with the people in service (Motor-Car) vehicle maintenance, rear-end collisions have occurred in the middle of the night, which may reduce the morale of the workers and the problems of safety management in the workplace lead to personal injury it frequently occurs. Therefore, in this paper, there is no centralized control system, tracking the position of the short-range technology and distance measurement has been applied modulation scheme RF without having to install another device in the work environment, taking into account sufficiently the convenience and portability I presented an interactive detection system technology has been applied.

      • 신호장치에 의한 ATS 신호장치 오동작 방지에 대한 연구

        Promotion of the line no.2 in Seoul Metro was changing from the existing signaling facilities for ATS(Automatic Train Stop) vehicles to the up-to-date signaling facilities for ATO(Automatic Train Operation). But, in consequence of conducting a trial run after being equipped with the ATO signaling facilities, the matter related to mix-operation with the existing ATS signaling facilities appeared. The operation of the existing ATS signaling system in combination with the ATO signaling system has made improper working related to frequency recognition of the ATS On-board Computerized Equipment. This obstructs operation of a working ATS vehicle. That is, as barring operation of an ATS vehicle that should proceed, it may make the proceeding ATS vehicle stop suddenly and after all, it will cause safety concerns. In this paper, we designed a wayside track occupancy detector that previously prevents improper working related to frequency recognition of the ATS On-board Computerized Equipment by gripping classification and working processes of operating trains throughout transmission of local signaling information from the existing facilities, which does not need to change or replace the existing signaling facilities. Furthermore, we described general characteristics of the wayside track occupancy detector and modeled the IFC(InterFace Contrivance) device and the logical circuit recognizing signal information. Then, we made an application program of PLC(programmable Logic Computer) based on the stated model. We, in relation to data transfer method, used the frame in TCP/IP transfer mode as the standard, and we demonstrated that ATO transmission frequency is intercepted.

        Along with international regulatory action to protect the global environment, the refrigeration and air conditioning industry has been challenged continuously to curb the use of fluoridated greenhouse gases. EU countries are introducing F-gas quota system to strengthen environmental regulations on HFCs, which have a significant impact on global warming. These regulations are affecting heat pump and airconditioners in which R410A refrigerants were used. For this reason, residential and light commercial R32 products are gradually expanding in the European market. This study aimed to compare the primary cooling and heating performance of R32 based on the R410A system and to guide R32 system design by comparing core components. The thermodynamic performance of the R32 is expected to be around 104% and can be developed without major changes in core components when switching to an inverter. Reductions in refrigerant charge are expected to decrease by 15 ~ 25% and CO2 reductions are expected to be 72.5 ~ 75.8%.
