사과 왜성대목 M.9 및 M.26의 고온, ribavirin, 생장점 배양을 통한 바이러스 제거

사과(Malus pumila)는 국내에서 가장 경제적으로 중요한 과수 중의 하나이다. 하지만 사과 바이러스 감염은 생산량을 감소시키고 수확량 손실과 과일 품질 저하와 같은 심각한 문제를 야기한다. 국내에 감염된 사과 바이러스 및 비로이드 종류는 Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV), Apple stem grooving virus (ASGV), Apple mosaic virus (ApMV)와 Apple scar skin viroid (ASSVd) 등이 알려져 있다. 사과는 바이러스나 비로이드에 감염되어 있어도 대체로 이상한 징후가 발견되지 않아 바이러스로 인해 피해가 많았다. 본 연구는 사과 왜성대목 M.9 및 M.26의 무독묘 생산을 위하여 고온처리($37^{\circ}C$, 6주), 화학처리(Ribavirin) 및 생장점 배양하여 바이러스 제거 처리를 하였다. 바이러스 검출에 일반적으로 사용되는 방법은 효소면역 측정법(ELlSA)과 중합효소연쇄반응(RT-PCR)을 이용하였는데, RT-PCR은 ELlSA방법보다 10 ~ 30% 더 민감하였다. 사과 왜성대목 바이러스 검정 결과, 바이러스 제거 효율은 생장점 배양이 가장 높았다. 생장점 배양 후 바이러스 무병묘의 획득율은 30 ~ 40%로 높게 나타났다. 생장점 배양에서 사과 왜성대목 M.9은 ACLSV, ASPV 및 ASGV의 비율이 각각 45%, 60%, 50%로 높았고, 사과 왜성대목 M.26에서는 ACLSV, ASPV 및 ASGV의 감염율은 각각 40%, 55%, 55%였다. 이상의 결과, 사과 왜성대목에서 무독묘를 생산할 수 있는 가장 효과적인 방법은 생장점 배양에 의한 것으로 판단되었다. Apple (Malus pumila) is one of the most economically important fruits in Korea. but virus infection has decreased the sustainable production of apples and caused serious problems such as yield loss and poor fruit quality. Virus or viroid infection including apple chlorotic leaf spot virus (ACLSV), apple stem pitting virus (ASPV), apple stem grooving virus (ASGV), apple mosaic virus (ApMV) and apple scar skin viroid (ASSVd) have been also reported in Korea. In many cases, as apple gets infected with virus and viroid with no specific symptoms, the damage and symptoms caused by the viruses are not detected. In our research, viruses in the rootstock were eliminated for a virus-free apple dwarfing rootstock of M.9 and M.26. The virus elimination methods were apical meristem culture, thermotherapy ($37^{\circ}C$, 6 weeks) and chemotherapy($Ribavirin^{(R)}$). The detection of apple viruses was accomplished by Enzyme-linked Immuno-Sorbent Assay (ELlSA) and reverse transcription-polymerase chain reaction (RT-PCR). RT- PCR method was 10 ~ 30% more sensitive than the ELISA method. The efficiency of virus elimination was enhanced in apical meristem culture method. The acquisition rate of virus-free apple dwarfing rootstocks was 30 ~ 40% higher in apical meristem culture. After the meristem culturing of M.9, the infection ratio of ACLSV, ASPV and ASGV was 45%, 60% and 50%, respectively. In the apple dwarfing rootstock of M.26, the infection ratio of ACLSV, ASPV and ASGV was 40%, 55% and 55%, respectively. Based on this study, the best method for the production of virus-free apple dwarfing rootstocks was the apical meristem culture.

전통누룩 진균류를 이용한 입국의 제조 및 입국곰팡이의 동정

다양한 향미를 가진 막걸리의 개발을 위해 전통누룩으로부터 분리한 곰팡이로 입국을 제조한 후 품질특성을 분석하여 입국의 규격에 적합하며 이취가 없고 관능이 우수한 9균주를 입국 제조용 우수균주로 최종 선발하였다. 선발된 균주는 Aspergillus oryzae(C1-5-2-2, C20-7-3, CN1.3.1-4, CN16.19.1-1, N152-1, N220-1), Mycocladus corymbiferus (N162-2), Rhizopus oryzae(N20), Lichtheimia corymbifera (N21)로 동정되었으며, 제조한 입국의 산도는 5.0~6.8, 당화력은 128~241sp이었다. Various koji were prepared by fungi isolated from traditional nuruk and their quality characteristics were investigated. Acidity and saccharification power of their koji were ranged in 5.0~6.8 and 128sp~241sp. Nine fungi which were showed good quality and sensory evaluation were identified by analysis of their nucleotide sequences with PCR-amplified 18S rDNA internal transcribed spacer-1(ITS-1) and ITS-4 genes. Among them, six strains were identified as Aspergillus oryzae and the other strains were identified as Mycocladus corymbiferus, Rhizopus oryzae, Lichtheimia corymbifera.

With the wide usage of the Internet, there use of pre-developed software is becoming important more and more in the development of various communication software. In this paper, the architecture to reuse the communication software is proposed. It is applied to the development of MIB-II system group of SNMP agent software for reusing. If Communication software is developed using this architecture, the software development life cycle will be abridged and the development cost will be reduced.

In this paper, we propose an improve regression testing method, which use method as the basic unit of changing. The test method consists of three steps. We represent the relationship of classes using the notation of UML(Unified Modeling Language), find the nodes of the modified and affected methods by node analysis, and then select changed test cases from the original test cases. The proposed object-oriented regression testing method can reduce the number of test cases, testing time and through reuse of test cases.

특권의 울타리 밖으로: 『어둠이 오기 전 여름』에 나타난 여성 주체성과 자유의 문제

Doris Lessing’s The Summer Before the Dark prefigures the third-wave feminism’s critique of the unitary notion of female subject hood that undermined earlier women’s liberation movements. Centered on Kate Brown’s evolving consciousness, the novel illumines her spiritual journey to the recognition that while she has been sacrificed as an angel-in-the-house, she nevertheless is a beneficiary in the global system of exploitation. Lessing, in the earlier half of the novel, posits the protagonist along the two familiar lines of women’s liberation narratives: career pursuit and sexual autonomy. It becomes clear, though, neither work nor sex will salvage Kate from her deep sense of failure and intense spiritual crisis. Instead, Lessing allows her to experience a series of mirroring encounter; people of diverse social status induce her to reflect on what she really is in a way that opens her eyes to crucial facets of socioeconomic formation and operation, illuminating how they have determined her seemingly unique personality, not to mention of her life devoted to her family. Further, Kate learns to symbolically align herself with the underprivileged through a per formative act of living as an ignored and therefore ‘invisible’ woman owing to unattractive look. Lessing ends this novel without presenting how Kate’s experience may result in significant changes in her life. Nevertheless, in the final act of returning home is embedded a potential of alternative forms of female subjectivity and liberation, rather than a debilitating failure or a gloomy social vision.

기이(旣異)함의 정동과 탈식민주의 생태비평: 레싱의 아프리카 이야기 읽기

This essay proposes a redefinition of the uncanny as an affect in order to foreground the implications of environmental texts from the perspective of postcolonial ecocriticism. I examine the conceptual history of the uncanny, beginning with Freud’s 1919 essay and the deconstructionist reconfiguration of the uncanny as literariness as such. The affective turn, I argue, encourages a re-evaluation of the Freudian uncanny as an affect rather than a personal emotional experience. With this theoretical discussion, I move on to an analysis of Doris Lessing’s two African stories. In “The Old Chief Mshlanga,” the uncanny affect arises when the protagonist-narrator “I” unexpectedly encounters a stunning African landscape beyond her parents’ farm. The uncanny affect conveyed by the African landscape suggests that African lands cannot be fully domesticated by white settlers and will remain as wild as they have always been. In “A Sunrise in the Veld,” the protagonist experiences a crucial moment of the affective uncanny when he witnesses a dying buck. The buck as a symbol for African nature dissolves the dichotomy between man and nature, evoking strong feelings of empathy in the boy and demonstrating the inherent connection between humans and non-human creatures.

배추무름병 원인균 분리 및 특성 연구

배추 무름병을 대상으로 식물병원균 감염모델을 확립하기 위하여 무름병변을 가진 배추조직으로부터 80개의 독립된 세균집락을 순수 분리하였다. 이들 균을 감염되지 않은 배추 잎의 주맥부위에 접종하여 24~48 시간 만에 무름병변을 나타내는 8개의 균주를 1차 선별하였다. 다양한 미생물학적, 생화학적, 형태학적인 시험을 통하여 분석한 결과 서로 다른 특성을 나타내는 균이라고 여겨지는 3개의 균을 최종 선정하였고 이들은 모두 그람 음성균으로 판명되었고 RB1, RB2 및 RB6로 명명하였다. API 20E시험, VITEK 2 COMPACT 분석, 16S rRNA 염기서열 분석 등을 종합할 때 RB1 및 RB2는 Erwinia carotovorum subsp. odoriferum 아종으로, RB6는 Erwinia carotovorum subsp. carotovorum 아종으로 추정되었다. 이들 균들은 30℃, 생리적 pH인 중성 pH에서 최적생육을 하였다. 이들은 배추의 상처 유무에 상관없이 초기 접종양과 비례하여 무름병을 유발하였으며, 상처가 없는 경우보다 상처가 있는 경우에 더욱 명확한 병변을 유발하였다. RB1의 경우 8.0×10? CFU/㎖, RB2 균주는 10? CFU/㎖, RB6는 4.7×10? CFU/㎖에서 최초로 무름 증상을 나타냈다. 이들 균의 숙주 특이성을 관찰하기 위하여 14종의 야채에 접종한 결과 배추, 가지 파프리카에서만 무름병변을 유발하는 숙주 특이성을 나타내었다. 이 연구에서 사용된 실험재료들 및 감염모델은 향후 식물병원균의 감염기작해석에 크게 기여할 것으로 예상된다. In order to establish in vitro infection model for research of plant pathogen based on tissue softening disease in napa cabbage, eighty independent bacterial strains were isolated from the softened napa cabbage tissues. Eight bacterial isolates were primarily screened with the generation of reproducible tissue softening disease to fresh napa cabbages within 24~48 hours after inoculation. Through various microbiological, biochemical and morphological examinations, three Gram (-) isolates which harbor independent biological properties were finally chosen, and named as RB1, RB2 and RB6. Collective results obtained from API 20E test and analyses of VITEK 2 COMPACT and nucleotide sequences of 16S rRNA of each isolate proposed that isolates RB1 and RB2 are close to the Erwinia carotovora subsp. odorifera, and RB6 is close to the Erwinia carotovora subsp. carotovora. These isolates grew optimally at 30℃ with neutral pH culture condition. The isolates caused softening tissue disease with dose-dependent manner regardless of pre-surface damages of napa cabbage. Minimum dose to cause soft rot disease for RB1, RB2 or RB6 were 8.0×10? CFU/㎖, 10? CFU/㎖ or 4.7×10? CFU/㎖, respectively. These isolates caused tissue softening disease to eggplant, paprika and napa cabbage out of 14 different tested vegetables, indicating that these isolates damages specific plant tissues. The bacterial isolates obtained in this research and in vitro plant infection model will be adapted in the understanding of the mechanism of pathogenesis by plant pathogen.

『연애하는 여자들』의 원시주의 서사와 정동

This essay suggests that the affective dimension of primitivism has a crucial part in D. H. Lawrence’s innovative characterization in Women in Love. Building on Michael Bell’s and Ben Etherington’s astute commentaries on Lawrentian primitivism, while drawing on Brian Massumi’s notion of affect, it foregrounds Gudrun and Birkin as major focalizers of the primitive narrative. First, in her relationship with Gerald, Gudrun turns him into a primitive object of her sadomasochistic desire. It is Lawrence’s narrative primitivism that makes this psychosexual mechanism feasible, generating precognitive and inexpressible affects, which nevertheless are embodied in the narrative as intensities arresting the characters. As Gerald becomes “primitive” through the logic of Gudrun’s desire, it is potently revealed that he is deeply subordinated to the ‘savagery’ of modern capitalist system, along with the machinery and the coal-miners. In case of Birkin, his primitivist aspiration brings forth alternative modes of sensual experience beyond the pale of heterosexual normativity. Right after the climactic moment of his conflicted relationship with Hermione, he undergoes blissful feelings of fulfillment in an erotic, yet asexual encounter with nature. Further, the wrestling scene with Gerald presents similarly intense and deeply gratifying feelings accompanying genuinely uninhibited sensuality. Yet, the ideological binary of primitivism as a discourse―into degenerative and regenerative ones―affects Birkin to repress his queer desire, and hold onto the safe ground of heterosexuality. Nonetheless, it deserves our attention that rather than an artistic credo or a part of imperial discourse, primitivism as a driving force of narrative enables Lawrence to embody affective realities of his characters, thus generating a unique aesthetics of the novel.
