        Class I HDAC s specifically regulate E‐cadherin expression in human renal epithelial cells

        <P><B>Abstract</B></P><P>Epithelial‐mesenchymal transition (EMT) and renal fibrosis are closely involved in chronic kidney disease. Inhibition of histone deacetylase (HDAC) has an anti‐fibrotic effect in various diseases. However, the pathophysiological role of isoform‐specific HDACs or class‐selective HDACs in renal fibrosis remains unknown. Here, we investigated EMT markers and extracellular matrix (ECM) proteins in a human proximal tubular cell line (HK‐2) by using HDAC inhibitors or by knockdown of class I HDACs (HDAC1, 2, 3 and 8). Trichostatin A (TSA), MS275, PCI34051 and LMK235 inhibited ECM proteins such as collagen type I or fibronectin in transforming growth factor β1 (TGF‐β1)‐induced HK2 cells. However, restoration of TGF‐β1‐induced E‐cadherin down‐regulation was only seen in HK‐2 cells treated with TSA or MS275, but not with PCI34051, whereas TGF‐β1‐induced N‐cadherin expression was not affected by the inhibitors. ECM protein and EMT marker levels were prevented or restored by small interfering RNA transfection against HDAC8, but not against other class I HDACs (HDAC1, 2 and 3). E‐cadherin regulation is mediated by HDAC8 expression, but not by HDAC8 enzyme activity. Thus, class I HDACs (HDAC1, 2, 3 and 8) play a major role in regulating ECM and EMT, whereas class IIa HDACs (HDAC4 and 5) are less effective.</P>

        Thermal evaluation of laser exposures in an<i>in vitro</i>retinal model by microthermal sensing

        <P>A temperature detection system using a micropipette thermocouple sensor was developed for use within mammalian cells during laser exposure with an 8.6-μm beam at 532 nm. We have demonstrated the capability of measuring temperatures at a single-cell level in the microscale range by inserting micropipettebased thermal sensors of size ranging from 2 to 4 μm into the membrane of a live retinal pigment epithelium (RPE) cell subjected to a laser beam. We setup the treatment groups of 532-nm laser-irradiated single RPE cell and in situ temperature recordings were made over time. Thermal profiles are given for representative cells experiencing damage resulting from exposures of 0.2 to 2 s. The measured maximum temperature rise for each cell ranges from 39 to 73C; the RPE cells showed a signature of death for all the cases reported herein. In order to check the cell viability, real-time fluorescence microscopy was used to identify the transition of pigmented RPE cells between viable and damaged states due to laser exposure.</P>

        <P>Pre-eclampsia involves a maternal inflammatory response that differs from both normal pregnancy and normotensive intrauterine growth restriction (IUGR). Our objective was to examine neutrophil Toll-like receptor (TLR), cryopyrin, nuclear factor-kappaB (NF-kappaB) subunit and interleukin-1beta (IL-1beta), and inflammatory cytokine profiles in women with pre-eclampsia or normotensive IUGR, as well as in normal pregnancy and non-pregnancy controls.</P>

      • Evaluation of imputation-based association in and around the integrin- -M (ITGAM) gene and replication of robust association between a non-synonymous functional variant within ITGAM and systemic lupus erythematosus (SLE)

        <P>We recently identified a novel non-synonymous variant, rs1143679, at exon 3 of the ITGAM gene associated with systemic lupus erythematosus (SLE) susceptibility in European-Americans (EAs) and African-Americans. Using genome-wide association approach, three other studies also independently reported an association between SLE susceptibility and ITGAM or ITGAM-ITGAX region. The primary objectives of this study are to assess whether single or multiple causal variants from the same gene or any nearby gene(s) are involved in SLE susceptibility and to confirm a robust ITGAM association across nine independent data sets (n = 8211). First, we confirmed our previously reported association of rs1143679 (risk allele 'A') with SLE in EAs (P = 1.0 x 10(-8)) and Hispanic-Americans (P = 2.9 x 10(-5)). Secondly, using a comprehensive imputation-based association test, we found that ITGAM is one of the major non-human leukocyte antigen susceptibility genes for SLE, and the strongest association for EA is the same coding variant rs1143679 (log(10)Bayes factor=20, P = 6.17 x 10(-24)). Thirdly, we determined the robustness of rs1143679 association with SLE across three additional case-control samples, including UK (P = 6.2 x 10(-8)), Colombian (P = 3.6 x 10(-7)), Mexican (P = 0.002), as well as two independent sets of trios from UK (P(TDT) = 1.4 x 10(-5)) and Mexico (P(TDT) = 0.015). A meta-analysis combing all independent data sets greatly reinforces the association (P(meta) = 7.1 x 10(-50), odds ratio = 1.83, 95% confidence interval = 1.69-1.98, n = 10 046). However, this ITGAM association was not observed in the Korean or Japanese samples, in which rs1143679 is monomorphic for the non-risk allele (G). Taken together along with our earlier findings, these results demonstrate that the coding variant, rs1143679, best explains the ITGAM-SLE association, especially in European- and African-derived populations, but not in Asian populations.</P>

        부품공급업체의 신제품 개발 프로젝트 참여에 대한 실증 연구

        이 논문에서는 제조 기업이 부품업체가 보유하고 있는 지식자원을 제품개발에 활용하기 위해서, 부품업체를 신제품 개발 과정에 참여시킨다고 가정한다. 이러한 지식기반관점에서 부품업체 참여를 유발하는 요인으로 제조기업의 신제품 개발 시간경쟁, 모듈 디자인 전략, 부품업체 파트너십 전략을 제안하였다. 즉, 제조 기업이 신속한 제품개발을 통한 시장선점을 노리는 경우, 필요한 지식자원을 직접 획득, 학습하기 보다는 부품업체와의 제휴를 선호한다고 예상하였다. 그리고 모듈화의 표준화 된 인터페이스는 제조 기업과 부품업체와의 통합, 조정을 원활하게 하고, 제조기업의 부품업체 파트너십 전략은 두 기업 간의 지식교류를 촉진시키고 상호 공유되는 지식의 범위를 넓힘으로써 효과적으로 신제품을 공동 개발할 수 있는 토양을 만들어 준다고 예상하였다. 그리하여 제조 기업은 신제품개발 과정에서 필요한 지식획득 시간을 단축하며, 오류 및 실수를 조기에 발견하고, 비용절감을 설계 단계에서 실현하게 되어, 신제품의 시장경쟁력(품질, 원가, 출시/납기)을 향상 시킬 수 있다고 기대하였다. 본 연구에서는 한국, 미국, 일본 및 유럽 5개국에서 수집된 총 159개의 신제품개발 프로젝트 데이터를 이용하여 제안된 모형을 검증하였다. From a Knowledge Based View, we conceptualize supplier involvement in a buyer’s new product development (NPD) project as the buying firm’s efforts to gain access to the supplier’s knowledge resources. This study proposes three antecedents to supplier involvement: the buying firm’s time-based new product development competition strategy, modular design strategy, and partnership with suppliers. All three antecedents were expected to result in greater supplier involvement in NPD projects. Data were analyzed from the High Performance Manufacturing Round III, which comprises the joint data collection efforts of Korea, Japan, the U.S., Austria, Germany, Italy, Sweden, and Finland.
