        無窮花의 成熟胚에서 體細胞胚 發生과 植物體 再分化

        Somatic embryogenesis and plant regeneration were obtained from mature embryos in Hibiscus syriacus L. ‘Bulsae’. Embryos were cultured on modified Murashige and Skoog(MS) or Woody Plant Medium(WPM) supplemented with 1,000mg/ℓ glutamine, 50mg/ℓ glycine and 50mg/ℓ serine, BAP(0 or 1.0mg/ℓ and 2,4-D(0.2~2.0mg/ℓ) in darkness. After 2 months, inoculated explants were transferred to MS medium without growth regulators and additives ; thereafter somatic embryos in various developmental stages were generated. When somatic embryos were transferred to half-strength MS medium containing 1.0mg/ℓ GA₃ and 1.0mg/ℓ ABA, they matured, germinated and produced plantlets with viable shoots and primary roots. Regenerated plantlets were hardened in a perlite:peatmoss(1:1, v/v) potting soil in culture room(25±1℃, 16/8 hour photoperiod) for 4 weeks, then transferred to greenhouse.

        Development of species specific RAPD markers using populus cpDNA

        Chloroplast DNAs(cpDNA) from eight different species of Populus(P. alba, P. glandulosa, P. alba×P. glandulosa, P. davidiana, P. maximowiczii, P. nigra×P. maximowiczii, P. nigra, and P. Koreana×P. nigra), Salix pseudo-lasiogyne and Nicotiana tabacum were compared by PCR amplification. The cpDNAs were amplified at relatively low annealing temperature so that many bands could be observed. Amplification of a spacer between 16S γRNA gene and 23S γRNA gene located in inverted repeats in cpDNA resulted in a major band in size of 2.3 Kb and several minor bands of various sizes. A band specific to Populus was found when the amplification was done at lower annealing temperature. Amplification with primers deduced from γpoC1 and γpoC2 gene produced the major target band as well as several additional (minor) bands that could be used for the differentiation of each Populus species. The minor bands appeared repeatedly even when different thermocycles were employed. With the amplification profile, it was possible to differentiate Populus species.

        This study was conducted to compare the physiological and biochemical responses of four Betula species in response to ozone, and to find out the relationship between ozone uptake rate and photosynthesis or antioxidant activity. One-year-old seedlings of four Betula sp, B. costata, B. davurica, B. platyphylla var. japonica, and B. ermani, exposed to 100 ppb ozone concentration for 8h day^-1 for 5 weeks in fumigation chamber. Ozone uptake rate, photosynthesis, SOD and GR activity were measured in the leaves of four species once a week. Cumulative ozone uptake rate was largest in the leaves of B. costata(53.8mmol m^-2), smallest in the leaves of B. davurica(35.5mmol m^-2). Photosynthesis of four Betula sp. exposed to ozone reduced relative to control, but the photosynthetic responses with changing stomatal conductance were different among species. Ozone exposure increased SOD activities of four species at the early exposing period, but after a critical point SOD activity decreased gradually. GR activity of B. costata was similar to the change of SOD activity, but the others showed the different patterns from B. costata. In conclusion, decreasing both SOD and GR activity at the critical point, B. costata may be sensitive species in response to ozone. In contrast, the others may be resistant species, which gradually increase GR activity following ozone exposure. GR activity was not always in accord with the change of SOD activity against ozone uptake, and the different responses between species were supposed to be affected by the cumulative ozone uptake.
