Development of species specific RAPD markers using populus cpDNA

Chloroplast DNAs(cpDNA) from eight different species of Populus(P. alba, P. glandulosa, P. alba×P. glandulosa, P. davidiana, P. maximowiczii, P. nigra×P. maximowiczii, P. nigra, and P. Koreana×P. nigra), Salix pseudo-lasiogyne and Nicotiana tabacum were compared by PCR amplification. The cpDNAs were amplified at relatively low annealing temperature so that many bands could be observed. Amplification of a spacer between 16S γRNA gene and 23S γRNA gene located in inverted repeats in cpDNA resulted in a major band in size of 2.3 Kb and several minor bands of various sizes. A band specific to Populus was found when the amplification was done at lower annealing temperature. Amplification with primers deduced from γpoC1 and γpoC2 gene produced the major target band as well as several additional (minor) bands that could be used for the differentiation of each Populus species. The minor bands appeared repeatedly even when different thermocycles were employed. With the amplification profile, it was possible to differentiate Populus species.

To investigate the causes of low seed fertility of colchitetraploid of black locust which was induced to be used for tree improvement program observations were made on the chromosome configuration in meiotic cell division and cytological characteristics and also on the growth habit of the progenies of the colchitetraploid. The results are summarized as follows. 1. In the meiotic cell division of tetraploid, the percentage of the occurrence of 20_(II) chromosomes was 21-64% and many univalent and multivalent chromosomes were observed in metaphaseⅠ, and laggard chromosomes, chromosome bridges and early separation of chromosomes were occurred with higher frequency in tetraploid than in diploid. It was considered that those phenomena cause irregular meiotic cell division in the pollen tetrads stage consequently resulting in low pollen fertility. 2. In the progenies of colchitetraploid, diploids, triploids, tetraploids and aneuploids were produced. And, the triploid, tetraploid and aneuploid progenies are lower in growth rate compared to diploids in their 2-year seedling stage, however the length of stomata and fibril are increased in those polyploid.
