        청둥오리 위 내분비세포에 대한 면역조직화학적 연구

        The relative frequency and distribution of bovine chromogranin(BCG)-, serotonin-, somatostatin(SOM)-, and S-100 protein-immunoreactive cells in the proventriculus, gizzard and proventriculus-gizzard junction(P-G junction) of the duck(Anas platyrhynchos platyrhyncos, Linne) were studied using immunohistochemical methods. BCG-, and SOM-immunoreactive cells were detected in all ages of this study in the proventriculus and the gizzard, and BCG-immunoreactive cells were also detected in the P-G junction. SOM-immunoreactive cells were detected rarely on occasions only in 1 week, 2 weeks, 3 weeks and 9 weeks after hatching. But no S-100 protein-immunoreactive cells were detected in this study.

        췌관을 결찰한 닭 췌장 외분비부의 형태학적 변화

        To investigate morphological changes in the exocrine pancreas of chicken after pancreatic duct ligation, experimental animals were subdivided to control, 12 hours, 1 day, 2 days, 4 days, 7 days and 10 days groupes and all of three pancreatic ducts of chicken were ligated by surgical procedure and then the morphological changes were observed. In pancreatic ducts, once for a while the ducts were dilated on 12 hours after pancreatic duct ligation and then they were obstructed because of proliferated epithelial cells and connective tissues in pancreatic duct. Marginal dissociation of acini was detected in 12 hours after pancreatic duct ligation and then dissociation of acini was increased with time and finally in 4 days after pancreatic duct ligation the acini showed completely dissociation except periductular regions and around pancreatic islets. Most of dissociated acini cells showed marginal condensation of nuclear chromatin and atropy of cytoplasm, namely, apoptotic features were detected in dissociated acinar cells. Interacinar spaces of dissociated acinar regions were dilated and fulfilled with increased connective tissue and in 4 days after pancreatic duct ligation, deposition of lymphocytes and hemocytes was occurred.

        닭 태자의 십이지장에 대한 조직학적 및 면역조직화학적 연구

        With histological changes, ontogeny and relative frequencies of bovine Sp-1/chromogranin(bCG)-, serotonin-, gastrin-, cholecystokinin-8(CCK-8)-, somatostatin-, S-100 protein-, polypeptide YY(PYY)- and glucagon-immunoreactive cells were investigated in the duodenum of the chicken embryos from 10 days of incubation to hatching. Histologically, pseudostraitified columnar epithelium were observed from 10 days of incubation to 14 days of incubation, thereafter these epithelium were differentiated to simple columnar epithelium. $Liberk{\ddot{u}}hn$ glands were observed from 18 days of incubation and goblet cells were detected from hatching. In the duodenum, bCG-immunoreactive cells were detected from 14 days of incubation and increased to 18 days of incubation, thereafter decreased with ages. Serotonin-immunorecative cells were detected from 14 days of incubation and increased with ages. Somatostatin-immunoreactive cells were detected from 14 days of incubation and CCK-immunoreactive cells were detected from 19 days of incubation. No gastrin-, S-100 protein-, PYY-, glucagon-immunoreactive cells were detected in this study.

        정전가미이진탕이 HCl-aspirin으로 유발된 십이지장 궤양에 미치는 조직학적 및 면역조직화학적 연구

        In order to study the effects of Jengjengamiyjintang on the duodenal ulcer induced by HCl-aspirin in rats, the changes of histological profiles, goblet cells(PAS-positive cells), and the distribution and frequency of cholecystokinin(CCK)-8 and serotonin-producing gastro-entero-endocrine cells were observed after oral administration of Jengjengamiyjintang. Histologically, very severe injury to duodenal epithelium were observed in control groups and these injuries were increased with time intervals. But in the Jengjengamiyjintang administrated groups, no gross lesion of ulcer were demonstrated and histologically minor injury to the mucosal epithelium were observed. PAS-positive cells were increased in the Jengjengamiyjintang administrated groups compared to that of control groups. Severe degranulation of CCK-8- and serotonin-immunoreactive cells were observed in control groups but these phenomenon was seldom in the Jengjengamiyjintang administrated groups. Serotonin-immunoreactive cells were significantly decreased in control groups but increased in Jengjengamiyjintang administrated groups compared with control groups. According to these result, it is suggested that Jengjengamiyjintang would accelerat the healing of the duodenal ulcer but the functional mechanisms were unknown.

        일령에 따른 닭 태자 췌장의 해부학적 및 조직학적 연구

        Anatomical and histological changes were studied in the dorsal, ventral, third and splenic lobes of the pancreas of the chicken embryos (8 days of incubation, 10 days of incubation to hatching). From 13 days of incubation, all four pancreatic lobes, namely, dorsal, ventral, third and splenic lobes were observed. Histologically, the pancreas of 10-14 days of incubation were consisted of mesenchymal tissue, exocrine acini and pancreatic islets. But mesenchymal tissues were disappeared from 15 days of incubation. The pancreatic ducts were observed from 14 days of incubation. The dark and light typed pancreatic islets were observed in splenic lobe from 13 days of incubation, in the third lobe from 11 days of incubation, and in the dorsal lobe from 13 days of incubation. But no dark typed islets were observed in the ventral lobes.

        Effect of Gongjindan, a Polyherbal Formula on the Pharmacokinetics Profiles of Sorafenib in Male SD Rats (1) - Single Oral Combination Treatment of Sorafenib 50mg/kg with Gongjindan 100mg/kg within 5min -

        Objective : The co-administration effects of Gongjindan (GJD) on the pharmacokinetics (PK) of sorafenib were observed as a process of the comprehensive and integrative medicine. Methods : After sorafenib treatment, GJD was administered within 5 min. The plasma were collected at 30min before administration, 30min, 1, 2, 3, 4, 6, 8 and 24hrs after end of GJD treatment, and plasma concentrations of sorafenib were analyzed using LC-MS/MS methods. PK parameters of sorafenib ($T_{max}$, $C_{max}$, AUC, $t_{1/2}$ and $MRT_{inf}$) were analysis as compared with sorafenib single administered rats. Results : The absorption of sorafenib were significantly increased at 30min, 1, 6 and 6hrs after co-administration with GJD as compared with sorafenib single treated rats. Accordingly, the $AUC_{0-t}$ (47.20%) of sorafenib was significantly increased but $t_{1/2}$ (-30.63%) and $MRT_{inf}$ (-34.11%) in co-administered rats were non-significantly decreased. These findings are considered as direct evidences that GJD increased the oral bioavailability of sorafenib through increase of the absorption, when they co-administered within 5min. Conclusion : Based on the results, co-administration of GJD increased the oral bioavailability of sorafenib through increase of the gastrointestinal absorption. It is considered that the more detail pharmacokinetic studies should be tested to conclude the effects of GJD on the pharmacokinetics of sorafenib, when they were co-administered, like the effects after co-administration with reasonable intervals considering the $T_{max}$ of sorafenib (about 3.5hr-intervals) and after repeated co-administrations.Hence, concomitant uses of GJD with sorafenib may require close monitoring for potential drug interactions.

      • 흑효모유래 $\beta$-glucan의 패혈증 치료효과 및 항돌연변이 활성 평가

        Anti-mutagenic and anti-septic effects of $\beta$-1,3/1,6-glucan from Aureobasidium pullulans SM-2001 were evaluated on the on the cyclophosphamide (CPA)-cecal ligation puncture (CLP) and CPA-treated mice. To induce immunosuppression and mutagenicity, 150 and 110 mg/kg of CPA were single intraperitoneally injected at 3 or 1 day before CLP or initial $\beta$-glucan administration. In CLP animals, the cecum was mobilized and ligated below the ileocecal valve, punctured through both surfaces twice with a 22-gauge needle. 125 mg/kg of $\beta$-glucan were dissolved in saline and subcutaneously or orally administered in a volume of 10 ml/kg (of body weight), 4 times, 12 hrs intervals from 6 hrs after CLP or 1 day after second dose of CPA. After treatment of $\beta$-glucan, the mortalities were observed in CPA-CLP model, and the appearance of a micronucleus is used as an index for genotoxic potential in CPA model. As results of CPA-CLP sepsis, all animals (9/9, 100%) in CPA-CLP control were dead within 2 days after CLP. In addition, increase of the number of bone marrow MNPCEs indicated mutagenicity were also observed by treatment of CPA. However, $\beta$-glucan treatment effectively inhibited the mortalities in CPA-CLP, and it also reduced the CPA treatment-related mutagenicity, respectively. These results indicated that $\beta$-glucan has effective anti-septic and anti-mutagenic effects and can be used as an agents for treating sepsis and mutagenicity related to high-dose chemotherapy or radiotherapy. However, further studies should be conducted to observe more detail action mechanisms of it's anti-septic and anti-mutagenic effects.

        췌관을 결찰한 닭 췌장 내분비부의 형태학적 및 면역조직화학적 변화

        To investigate morphological changes in the endocrine pancreas of chicken after pancreatic duct ligation, experimental animals were subdivided to control, 12 hours, 1 day, 2 days, 4 days, 7 days and 10 days groupes and all of three pancreatic ducts of chicken were ligated by surgical procedure and then the morphological changes were observed. In pancreatic islets, the vacuolation and invasion of connective tissue were occurred in all experimental groups and dissociation of pancreatic islets was detected in 4 days after pancreatic duct ligation and hold out to 10 days. The peak of the morphological changes in pancreatic islets was detected in 4 days after pancreatic duct ligation. In the results of immunohistochemical methods against glucagon, insulin, somatostatin and bovine pancreatic polypeptide(BPP), the number of immunoreactive pancreatic islets were decreased but the size increased with time, so the number of immunoreactive cells in each pancreatic islets were increased. Glucagon-immunoreactive cells were not changed but insulin-immunoreactive cells were decreased with time(p<0.05). BPP-immunoreactive cells were increased in 2 days after pancreatic duct ligation and then decreased with time(p<0.05). Somatostatin-immunoreactive cells were increased with time(p<0.05) in dark islets.

        Immunohistochemical study of the pancreatic endocrine cells in the ICR mice

        ICR 마우스 췌장에 존재하는 내분비세포의 부위별 분포 및 상대적 빈도를 4 종류의 항혈청 즉, insulin, glucagon, somatostatin 및 human pancreatic polypeptide (PP)을 사용하여 면역조직화학적 방법으로 관찰하였다. 마우스의 췌장은 췌장섬, 외분비부 및 췌관의 3 부분으로 구별되었으며, 췌장섬은 이들 면역반응세포들의 출현 부위에 따라 다시 중심부분, mantle부분 및 가장자리부분으로 다시 세분되었다. 췌장섬의 경우, insulin 면역반응세포들은 주로 췌장섬의 중심부분과 mantle 부분에서 주로 관찰되었으나, somatostatin, glucagon 및 PP 면역반응세포들은 다양한 출현빈도를 나타내며 주로 mantle부분과 가장자리부분에 걸쳐 관찰되었다. 또한 극소수의 PP 면역반응세포들은 췌장섬의 중심부분에서도 관찰되었다. 외분비부에서도 insulin, glucagon, somatostatin 및 PP 면역반응세포들 모두 관찰되었으며, 주로 외분비 샘포 세포 사이공간에서 관찰되었다. 췌관에서는 insulin 및 glucagon 면역반응세포들이 소수 또는 극소수의 빈도로 췌관 상피세포 사이공간에서 관찰되었으며, 극소수의 PP 면역반응세포들 역시 췌관상피 아래부위에서 관찰되었으나, somatostatin 면역반응세포들은 이 부위에서 관찰되지 않았다. The regional distribution and relative frequency of the pancreatic endocrine cells in the ICR mouse were studied by immunohistochemical (PAP) method using four types of specific antisera against insulin, glucagon, somatostatin and human pancreatic polypeptide (PP). The pancreas of mice could be divided into three portions; pancreatic islets, exocrine and pancreatic ducts. Pancreatic islets, furthermore, were subdivided into three regions (central, mantle and peripheral region) according to their located types of immunoreactive cells. In the pancreatic islet portions, insulin-immunoreactive cells were located in the central and mantle regions but most of somatostatin-, glucagon- and PP-immunoreactive cells were detected in the mantle and peripheral regions with various frequencies. In addition, PP-immunoreactive cells were also found in the central regions of pancreatic islets of ICR mouse. In the exocrine portions, all four types of immunoreactive cells were demonstrated in the ICR mouse. In the pancreatic duct portions, insulin- and glucagon-immunoreactive cells were situated in the epithelial lining of ICR mouse with a few and rare frequencies, respectively. In addition, rare PP-immunoreactive cells were also demonstrated in the subepithelial regions of the pancreatic duct. However, no somatostatin-immunoreactive cells were demonstrated.

        The anti-climacterium effects of red clover dry extracts combined with pomegranate concentration powder in ovariectomized rats

        Objective : In this study, the addition of dried pomegranate concentrate powder (PCP) was affected the anti-climacterium activity of red clover dry extracts (RC) in ovariectomized (OVX) rats. Materials and methods : After bilateral OVX surgery, RC 40 mg/kg, PCP 20 mg/kg and RC:PCP 2:1 mixture (g/g) 120, 60 and 30 mg/kg (of body weight) were orally administered, once a day for 84 days, and then the changes on the serum estradiol levels, abdominal fat pad and uterus weights were observed for estrogenic effects. In addition, liver weights, serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels were also evaluated for hepatoprotective effects, and serum total cholesterol (TC), low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride (TG) levels were monitored for hypolipidemic effects. Results : As a result of OVX, the estrogen-deficient climacterium symptoms, increments of abdominal fat pad weights, serum AST, ALT, TC, LDL and TG levels with decrease of uterus and liver weights, serum estradiol levels, were demonstrated. However, these estrogen-deficient climacterium symptoms induced by bilateral OVX in rats were significantly inhibited by continuous oral treatment of RC 40 mg/kg, PCP 20 mg/kg and RC:PCP 2:1 mixture (g/g) 120, 60 and 30 mg/kg, respectively. Conclusion : The results suggested that RC:PCP 2:1 mixtures synergistically increased the anti-climacterium effects of RC in OVX rats. It, therefore, is expected that RC:PCP 2:1 mixture will be promising as a new potent protective agents for relieving the climacterium symptoms.
