Recently various molecular diagnostic techniques have been used to identify rabbit hemorrhagic disease virus (RHDV), a causative agent responsible for acute hepatitis and disseminated intravascular coagulation in rabbit. But they were hard to perform and time consuming. To detect RHDV in a rapid and easy way, we developed biotinylated oligonucleotide probe within ORF 1 region encoding the polyprotein of RHDV in formalin-fixed and paraffin-embedded tissues from various tissues of 20 rabbits naturally infected with RHDV, Our in situ hybridization (ISH) was quickly carried out within two hours by MicroProbe capillary action system. The ISH produced a positive reaction in liver, kidney and lung. In conclusion, ISH with a biotintlated oligonucleotide probe provided a useful diagnostic method for detecting RHDV.

Pathological studies were performed on the five piglets experimentally infected with Aujeszky's disease virus(pseudorabies), NYJ isolate, isolated from the naturally infected pigs in Korea: two piglets were inoculated intramuscularly, two piglets intranasally, and one piglet subcutaneously at the dose of 1$m\ell$ per animal with the 105.5 $TCID_50$/0.1ml titer. Clinical signs included dyspnea, high fever(>$41^{\circ}C$), anorexia, vomiting, diarrhea or constipation, ataxia, circling movement, posterior paralysis, intermittent convulsion, and coma followed by death although some variations by age and inoculated routes were observed. Gross features included multiple necrotic foci in the liver, congestion and hemorrhage in the lymph nodes and spleen, petechial hemorrhage in the kidney, hemorrhagic pneumonia, marked meningeal congestion, severe sub meningeal hemorrhage in the spinal cord, excessive cerebrospinal fluid retention, and muscular necrosis at the inoculated area. Microscopically, non suppurative meningoencephalitis with gliosis and perivascular cuffing in CNS, ganglioneuritis, necrohemorrhagic splenitis, necrotic hepatitis, tonsillitis and rhinitis, hemorrhagic or interstitial pneumonia, and non-suppurative myositis in the injected area were observed. Eosinophilic intranuclear inclusion bodies were found in a variety of tissues the including the liver, kidney, adrenal gland, spleen, lymph nodes, tonsil, and lung. Ultrastructurally, virus particles were confirmed in nucleus and cytoplasms of pneumocytes around the necrotic areas.

Newcastle disease (ND) is a highly contagious infection of poultry, Two pathology-based techniques, in situ RT-PCR and in situ hybridization (ISH) were applied to formalin-fixed, paraffin-embedded tissues from chickens naturally infected with velogenic ND virus (VNDV). Two pairs of primers and a probe for ISH and in situ RT-PCR, respectively, were selected from highly conserved region of matrix gene of NDV. The ISH experiment was carried out using MicroProbe$^{TM}$ capillary action system within 2 hours. In situ RT-PCR was performed using MicroProbe$^{TM}$ capillary action system and GeneAmp In Situ PCR system. With ISH and in situ RT-PCR, viral nucleic acid was detected in the central nervous system of chickens from infected with neurotropic velogenic Newcastle disease virus (NVNDV), whereas viral nucleic acid was detected in various organs or tissues of chickens from infected with viscerotropic velogenic Newcastle disease virus (VVNDV). In the NVND group, positive signals were characteristically defined in the cytoplasm of neuron, vascular endothelial cells, and perivascular mononuclear macrophages in the central nervous system. One of NVND group, chicken from one farm exhibited positive signals in the bronchial epithelium. The VVND group chickens showed positive reaction in the macrophages, vascular endothelium, and bronchiolar epithelium. Markedly, viral nucleic acid was detected in the macrophages of morphologically normal tissues which were peripheral or located in distant areas from lesions. The central nervous system of chickens infected with VVND virus had positive signals in the vascular endothelial cell, perivascular mononuclear macrophages and some neuron. The number and intensity of the positive cells by in situ RT-PCR were more and stronger, respectively, in comparison with those by ISH. Particularly, positive reaction was detected in macrophages infiltrating in cardiac muscle by in situ RT-PCR, but not obtained by ISH. Therefore, these results demonstrated that ISH is a rapid diagnostic method for detection of NDV and in situ RT-PCR can be used as an efficient method for detection of low viral load infection or subclinical viral infection of NDV.
보편적 ‘참여 수준’을 전제한 공공 건축물의 ‘커뮤니티 참여 디자인 프로세스’에 관한 연구

본 연구는 지역 커뮤니티의 일관된 의견 수렴 및 의사 결정 모형 수립을 위해 보편적 ‘참여 수준’을 전제한 공공 건축물의 체계적인 ‘커뮤니티 참여 디자인 프로세스’ 기준을 제안하는 것에 목적을 두었다. 연구의 방법으로는 ‘건축적 공공성’과 ‘커뮤니티 참여 디자인’의 이론 고찰을 시작으로 ‘참여 수준’과 관련한 주요 연구자들의 논거(論據)를 종합하여 ‘참여 디자인프로세스’의 근간이 되는 보편적 ‘참여 수준’ 체계를 선행 수립하고자 하였다. 이후, 국내 ‘참여 디자인’ 적용 연구 사례와의 비교 및 분석을 통해 현행 국내 ‘참여 디자인 프로세스’의 문제점을 파악하고 이를 해결하기 위한 방안을 제시하고자 하였다. 본 연구의 범위는 이론 연구 사례와 적용 연구 사례 범위로 구분되며, 이론 연구 사례에 대한 범위는 ‘참여 수준’과 관련하여 다수의 연구 문헌에 인용되는 주요 연구자 8인의 논거로 한정하였다. 적용 연구 사례에서 공간적 수집 범위는 국내로 한정하고 시간적 범위로는 2000년 이후의 관련 연구 문헌(25개)으로 한정하였다. 본 연구를 통해, 총 10단계의 보편적 ’참여 수준‘을 도출해 낼 수 있었으며, 국내 ‘참여 디자인’ 연구 문헌 분석으로 공공 건축물 중 지역 커뮤니티와 깊은 관계성을 가지고 있는 문화복지시설에 관한 연구가 다른 용도에 비해 ‘참여 수준’ 설정 및 이를 바탕으로 한 체계적인 프로세스 수립이 상대적으로 미진함을 파악할 수 있었다. 또한, 분석 대상 다수의 ‘참여 디자인 프로세스’가 ‘참여 수준’에 대한 근본적인 고려가 전무한 상태에서 연구자별 주관적 견해에 의해 수립되어 연구 문헌간의 보편성과 일관성이 부족한 것으로 분석되었다. 이에 본 연구에서는 상기의 연구 결과를 종합하여 보편적 ‘참여 수준’을 전제한 일관화된 ‘커뮤니티 참여디자인 원칙’과 국내 현행 설계 업무 체계가 고려된 체계적인 ‘커뮤니티 참여 디자인 프로세스’의 기준을 제시하였다. This study aims to suggest a standard of systematic “community participation design process” of public buildings that was premised on universal “participation level” in order to establish a consistent opinion convergence and decision-making model of local community. As the method of study, It would like to precede and establish a universal ‘participation level’ that is based on ‘participation design process’ and to collect basis of arguments that is collected arguments by main researchers that is related from ‘architectural publicness’ and a theory consideration of ‘community participation design’ to a level of participation. A scope of study is divided into a range of applied study cases and theoretical study case. and a scope of the theoretical study case is limited to basis of arguments of 8 people who are main researcher that used to appear in numbers of study literatures with a ‘participation level’. In applied study cases, A scope of spatial collection is restricted to domestic and a scope of temporal collection is restricted to related study literature(25 literatures) that is written since 2000. Through this study, We were able to begin to derive the universal “participation level” of all 10 steps, and through a analysis of domestic “participation design” study literatures, we were able to catch the fact a study of culture welfare facilities that is deeply related on regional community among the public building isn’t entirely satisfactory in setting of “participation level” and in the systematic establishment of process compared with other uses. In addition, It is showed universality and consistency between study literatures are not entirely satisfactory because of subjective opinions by researchers in a state of almost nothing about fundamental consideration of ‘participation level’ in a ‘participation design process’. Therefore, We did piece study result together in this study and this study did suggest some standards of systematic ‘community participation design process’ considered domestic present condition of design system and standardized ‘community participation design principle’ that is premised on universal ‘participation level’.

The concentrations of some components of the serum in a liver function test were determined in samples of 74 apparently healthy dairy cattle, imported from foreign countries in Jeonnam district, during the periods of June to September of 1975 and July to August of 1976. The ranges, mean concentrations and activities of the SGOT, SGPT, SALP, thymol turbidity, total protein, serum albumin, serum globulin, A/G ratio, total bilirubin and total cholesterol were investigated in this work. This results obtained in the survey were summarized as follows: 1. The SGOT activities obtained from Holstein cows ranged from 57 to 129 Sigma Frankel units/ml, with a mean of $96.5{\pm}19.38$ S.F. units/ml. 2. The SGPT activities obtained from Holstein cows ranged from 5 to 49 Sigma Frankel units/ml, with a mean of $21.27{\pm}9.52$ S.F units/ml. 3. The alkaline phosphatase activities of serum obtained from Holstein cows ranged from 0.3 to 3.8 Sigma Frankel units/ml, with a mean of $1.88{\pm}0.94$ S.F. units/ml. 4. The thymol turbidity of serum obtained from Holstein cows ranged from 0.2 to 4.4 Shank Hoagland units/ml, with a mean of $1.69{\pm}0.30$ S.H units/ml. 5. The total serum protein values of Holstein cows ranged from 5.9 to 8.6g/100ml with a mean of $7.17{\pm}0.65g/100ml$. 6. The serum albumin values of Holstein cows ranged from 2.5 to 4.3g/100ml with a mean of $3.24{\pm}0.28g/100ml$. 7. The serum globulin values of Holstein cows ranged from 2.9 to 5.8g/100ml with a mean of $4.02{\pm}0.72g/100ml$. 8. The A/G ratio of serum obtained from Holstein cows ranged from 0.5 to 1.0 with a mean of $0.78{\pm}0.12$. 9. The total bilirubin of serum obtained from Holstein cows ranged from 7.2 to 0.8mg/100ml, with a mean of $0.32{\pm}0.11mg/100ml$. 10. The total cholesterol of serum obtained from Holstein cows ranged from 50.5 to 240.6mg/100ml with a mean of $135.70{\pm}57.44mg/100ml$. 11. There was little difference in the concentrations of the various serum components between cow groups by birth countries and total cow group, except for SGOT activities, serum alkaline phosphatase activities, thymol turbidity of the Holstein cows from New Zealand.
국내에서 발생한 돼지 뇌심근염 바이러스 감염증의 병리학적 관찰

Pathological studies by light and electron microscope were carried out on the twenty piglets naturally affected by encephalomyocarditis virus infection. Gross findings included pale or yellow, small necrotic foci on myocardium, together with pulmonary edema and liver congestion in some cases. On light microscopy, nonsuppurative interstitial endocarcitis, epicarditis and myocarditis, myocardial infarction, and dystrophic calcification or fibtosis were observed in heart. Perivascular cuffings, gliosis and nonsuppurative meningitis were appeared in brain. Focal or diffuse necrosis with mononuclear cell infiltration in lacrimal gland and multifocal necrosis in liver were observed in some cases. Congestion and edema of lung, hyperemia, hemorrhage and deletion of lymphocytes of lymph nodes and spleen were recognized. On electron microscopy, severe swelling and vacuolization of mitochondria and sarcoplasmic reticulum, large intracellular vacuolation and edema, separation and fragmentation of myofibrils were observed. Virus particles were seen in the sarcoplasm of degenerated cardiac muscle cell.
에틸렌이 유도한 강낭콩 잎 키틴분해효소의 정제 및 특성

Chitinase (EC 3.2.1.14), a potential pathogensis-related protein, was induced in leaves of 30-day-old bean (Phaseolus vulgaris) plants with treatment of 10 nl/ml ethylene for 30 hr. The enzyme was extracted in sodium citrate buffer (pH 5.0), and then purified by heat treatment, (NH₄)₂SO₄ precipitation, chitin-affinity and Sephadex G-75 chromatographies. Chitinase was enriched 22.5-fold with a yield of 29%. Its apparent molecular weight was estimated on SDS-PAGE to be 30 kD. Chitinase showed optimum pH 5.0 and broad pH stability, and showed thermal stability below 50℃. K_m and V_(max) values of the purified chitinase were 0.36 ㎎/㎖ and 0.7 nmole G1cNAc/min, respectively, for swollen chitin as a substrate. Chitinase was significantly inhibited by 10 mM Cu^(++), Hg^(++), S₂O₃^(--), S₂O₄^(--) or SO₃^(--) in reaction mixtures. Antibody was raised in rabbits against the chitinase and used for specific immunodetection of the enzyme.
