        상추 (Lactuca sativa L.) callus로부터 ACAT 억제 활성물질, phytol의 생산

        상추의 기내 callus 배양으로부터 ACAT (Acyl-CoA: Cholesterol acyltransferase) 억제 활성물질로 알려진 phytol 의 생산가능성이 연구되었다. Callus는 0.5mg.L$^{-1}$ NAA가 첨가된 MS 배지에 4주간 치상된 자엽 절편체로부터 유도되었으며, 다양한 수준의 chlorophyll을 함유하고 있었다. Phytol이 chlorophyll분자를 구성하는 물질로 알려져 있음에 도 불구하고, phytol의 추출량은 녹색의 자엽에서 보다 callus 에서 높게 나타났다. GC 분석결과, phytol의 추출량은 callus의 chlorophyll 함량과 비례하지 않는 것으로 나타났으며, callus의 chlorophyll 함량수준은 callus의 형성능과 관련이 있는 것으로 사료되었다. 본 연구의 결과는 phytol의 기내생산에 있어서 상추의 callus 배양의 잠재성을 제시한다 하겠다. The possibility for mass production of phytol, inhibitory diterpene against ACAT (Acyl-CoA: Cholesterol acyltransferase) was investigated by using callus culture of lettuce. The callus were induced from lettuce cotyledon explants on MS medium containing 0.5 mg.L$^{-1}$ NAA after 4 week's culture. Adventitious roots were formed from the explants on MS medium containing 0.5 mg.L$^{-1}$ IBA or 1.0 mg.L$^{-1}$ NAA. Adventitious shoots and roots were emerged from the callus when the callus was transferred to MS medium containing auxin alone, or with cytokinin. The plant growth regulators and their concentrations for the organogenesis were 1.0 mg.L$^{-1}$ NAA, 0.1 mg.L$^{-1}$ BA, 0.5 mg.L$^{-1}$ NAA with 0.1 mg.L$^{-1}$ kinetin, or 0.5 g.L$^{-1}$ 2.4-D with 1.0 mg.L$^{-1}$ kinetin. Analyses of chlorophyll contents showed that chlorophyll contents were higher in morphogenic calli than in non-morphogenic calli. However, the chemical analyses of gas chromatography indicated that phytol contents were not proportionate to the chlorophyll contents of callus. The content of phytol was higher in callus than in lettuce cotyledon.ledon.

      • 계량설비용 디지탈 출력 로드셀의 개발에 관한 연구

        본 연구에서는 계량 설비에 사용되는 스마트형 디지털 로드셀을 개발하였다. 로드셀 센서는 중량의 변화에 대해 매우 민감하여햐 하므로 정밀한 A/D변환을 위하여 온도 안정성, 낮은 드리프트 특성 그리고 분해능이 우수하여야 한다. 단일칩 마이크로프로세서에 의해 제어되는 고안된 아날로그 회로로써 OP엠프 오프셋과 드리프트 특성을 저감시키며 소프트웨어 알고리즘에 의해 안정되고 정밀한 A/D 변환이 가능하도록 디지털 로드셀을 설계하였다. 또한 RS-485통신 방식으로 로드셀을 제어하고 보정용 데이터와 제어 데이터를 기억시키는 기능들도 포함하였다. 시뮬레이션과 실측 평가를 통하여 개발된 로드셀의 우수성을 입증하였으며, 본 연구의 결과는 원격계량 센서로서 계량 설비 분야에 유용한 활용이 기대된다. This paper describes the design and development of a smart digital load cell used forweighing installations. Sice the load cell sensor to be used is very sensitive for weight cariation, the load cell must have the temperature stability, low-drift and the high-resolution of the A/D conversion for accuracy. A new analog circuit which is controlled by one chip micro-processer has been developed to reduce the offset voltage and the drift characteristics of operational amplifiers, and has been adapted into the digital load cell. Also, a software algorithm has been developed to obtain the stable and accurate A/D conversion. This software includes a RS-485 communication program to control the digital load cell, which gives a capability of backing-up the calibration data and transferring control data. The simulation and evaluation of the designed digital load cell has been shown as having the good performance. which will give useful application to the weighing installations as a remote weighing sensor.

      • Beam形 load cell의 FEM simulation과 그 電氣的特性

        In this paper, we simulate and calculate the stress and output voltage of the beam structure load cells by using FEM as varing physical structure parameters and loading positions. It is proved that stress enhance as the increase of the notch pitch and radius of the load cell, but decrease as the increase of the notch thickness and beam width. The results are good matched for basic formulas of the single fixed beam, and are verified our simulation is correct. Also, it is found that the stress characteristics of the load cell is varied according to loading positions with structure parameters, and caculated output voltage of the load cell approximate to those of the real manufactured ones. As a result, this study will offer efficient design and analysis technique for making special and variety capacity of load cells.

      • Truck Scale 용 Load Cell의 Smart interface Board 개발

        This paper presents the design of a smart A/D conversion interface used for measuring the load of a truck. Since the load-cell sensor to be used is very sensitive for weight variation, the interface board must have the low-drift and the A/D conversion for accuracy. A new integrator and comparator has been developed to reduce the offset voltage and the drift current of operational amplifiers and has been adapted into the interface board. Also, a software algorithm has been developed to obtain the stable and accrurate A/D conversion. This software includes a RS-485 communicationprogram to control the interface, which gives a capability of backing-up the calibration data and transferring control data. The test and evaluation of the designed interface has been shown as having the better performance compared to the other types of existing weighing systems and sensor instruments.

      • 상추(Lactuca sativa L.) 형성능 Callus의 특성

        Calli of Lactuca sativa L. were induced from cotyledons on MS media containg 0.1∼2.0mg·L^-1NAA. or 0.1∼2.0mg·L^-1 2,4-D. Plantlet regeneration could be obtained through organogenesis when calli were transferred to MS media containg 0.1∼1.0mg·L^-1NAA with 0.1∼1.0mg·L^-1BA, or 0.1∼1.0mg·L^-12,4-D with 0.1∼1.0mg·L^-1 kinetin. Morphogenic callus was characterized with that a callus consister with smaller, denser cytoplasm, and colored green. It seemed that chlorophyll content of the cell was closely related to the morphogenic ability of the callus.
