Soybean is known to have a variety of biological activities including anti-tumor activity against hormone dependent tumors such as breast end prostate cancers and other hormone independent ones such as head and neck and colon cancers. Several peptides including trypsin inhibitor peptide and small molecule compounds, genistein and isoflavones, from soybean are supposed to contribute the anti-tumor activity of soybean. However, some of these compaunds do not show cytotoxicity on tumor cells. The proliferation of new capillary blood vessels, called angiogenesis or neovascularization, is one of crucical steps in transition from a small, harmless cluster of mutated cells to a large, benign growth, capable of spreading to other organs throughout the body. It is highly possible that some of peptides or isoflavones from soybean may be able to work out cooperatively for the inhibition of angiogenesis in the body resulted in tumor regression without showing cytotoxicity on tumor cells.
Cyclomaltodextrinase를 생산하는 Alkalophilic Bacillus sp. KJ-133의 분리와 효소생산 조건

산업적으로 유용한 미생물 유래 CDase를 생산, 이용하기 위해 전국 20여곳에서 채취한 토양 샘플로부터 균주를 분리하였다. 분리균은 호기성, Gram 양성이고, 간균(0.2~0.4 × 1.4~4.4 ㎛)의 형태를 하고 있으며, 편모를 가지고 있어 운동성을 보이고, 구형의 포자를 형성하였다. 또한 DNA 염기조성(G+C)함량은 35.8 mol%이었으며, 생장을 위한 pH와 온도 범위는 각각 7.5~11.0 및 30~40℃였다. 이러한 형태학적, 생리·생화학적 특성의 결과에 따라 본 균주는 alkalophilic Bacillus sp. KJ-133으로 동정되었다. KJ-133의 배양시간동안 CDase 활성을 검토하고자 21 배양용량의 삼각플라스크에 균주를 접종하여 37℃, 250 rpm에서 배양을 하였다. 그 결과 배양 48시간 후의 효소활성이 가장 높았다. 또한 효소는 Hg^2+, Ag^2+, Cu^2+ 및 p-chloromercuribenzoate(PCMB)와 같은 thiol reagents에 의해 저해되었다. To produce and utilize mecrobial cyclomaltodextrinase being industrially useful, we isolated an alkalophilic Bacillus strain from soil which was capable of degrading cyclodextrins. The newly isolated strain was aerobic, gram-positive, spore-forming, motile, rod shape(0.2~0.4 × 1.4~4.4 ㎛), and 35.8 mol% of DNA base composition. Based on its morphological, physiological, and biochemical properties, it was identified as alkalophilic Bacillus sp. KJ-133 and cultivated well in the ranges of 30~40℃ and pH8.0~9.0. The cyclomaltodextrinase of the strain showed maximal production after 48 h of cultivation at 37℃. and the activity was inhibited by Ag^2+, Hg^2+, Cu^2+, and p-chloromercuribenzoate.
