Mahwangsan is used in the treatment of asthma due to wind and cold(風寒喘). This study was carried out to investigate the effect of Mahwangsan extract and each drug composing it on the contractile force of the isolated guinea pig trachea muscle and elucidate its mechanism. The results were obtained as follows; 1. The isolated trachea smooth muscle of guinea pig was signifi-cantly relaxed by the administration of Mahwangsan extract. 2. The contractile response of the isolated trachea smooth muscle of guinea pig to histamine was significantly inhibited by the administration of Mahwangsan extract. 3. The contractile response of the isolated trachea smooth muscle of guinea pig to histamine was significantly inhibited by the administraction of Herba Ephedrae, Cortex Cinnamoni, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract. 4. The contractile response of the isolated trachea smooth muscle of guinea pig to acetylcholine was significantly inhibited by the administration of Mahwangsan extract. 5. The contractile response of the isolated trachea smooth muscle of guinea pig to acetylcholine was significantly inhibited by the administration of Herba Ephedrae, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract. 6. The contractile response of the isolated trachea smooth muscle of guinea pig to 5-hydroxytryptamine was significantly inhibited by the administration of Mahwangsan extract. 7. The contractile response of the isolated trachea smooth muscle of guinea pig to 5-hydroxytryptamine was significantly inhibited by the administration of Herba Ephedrae, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract.
삼소음(蔘蘇飮)이 백서(白鼠)의 국소뇌혈류량(局所腦血流量)과 혈압(血壓)에 미치는 영향(影響)

Objective : The objective of this study is to measure the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats with the following injection. Methods : The measurement was continually monitored by laser-doppler flowmeter-Transonic Instrument, USA- and pressure tranducer(Grass, USA) in anesthetized adult Sprague-Dawley rats about for two hour to two hour and a half hours through the data acquisition system composed of Maclab and Macintosh computer. Result : This experiment with Samso-eum increased the changes of rCBF in rats, significantly, but did not change the blood pressure. The rCBF of Samso-eum decreased by pretreated propranolol, and was not changed by pretreated L-NNA and ODQ. Conclusion : It is considered that the dosage-dependent increase on rCBF is mediated by adrenegic ${\beta}$-receptor.
폐암세포주(肺癌細胞株) H460에 대(對)한 보중익기탕(補中益氣湯)의 세포고사효과(細胞枯死效果) 및 기전연구(機轉硏究)

Objectives : This study was designed to evaluate the effect on cytotoxicity of Bojungikki-tang(BIT) in human lung cancer H460 cells. Methods : BIT-induced cell death was confirmed as apoptosis characterized by chromatin condensation and increase of the $sub-G_1$, DNA content. It was tested whether the water extract of BIT affects the cell cycle regulators such as, p2l/Cipl, p27/Kipl, cyclin $B_1$. Results : The data showed that treatment of BIT decreased the viability of H460 cells in a dose-dependent manner. p2l/Cip1 is gradually decreased by the addition of the cells with BIT extract. Interestingly, p27/Kip1 is not detected for 24 hr after the addition of BIT extract, however, after 24 hr, p27/Kipl markedly increased. In addition, cyclin $B_1$, decreased in a time dependent manner after the addition of the water extract. The activation of caspase -3 protease was further confirmed by degradation of procaspase-8 protease andpoly(ADP-ribose) polymerase(P ARP) by BIT in H460 cells. Moreover, BIT induced the increase of Bak expression. Conclusion : These results suggest that the extract of BIT exerts anticancer effects to induce the death of human lung cancer H460 cells via down regulation of cell cycle regulators such as p2l/Cip1, and cyclin B1 or up regulation of cell cycle regulators such as p27/Kip1. Moerover results suggest that BIT induces an apoptosis in H460 cells via activation of intrinsic caspase cascades.
瀉白散의 肥滿細胞 嫫介性 炎症性 細胞活性物質 分泌 抑制 效果

Ocjective : Sabaeksan has been used for the purpose of prevention and treatment of bronchial asthma and allergic asthma in korea. Methods : To investigate the biological effect of Sabaeksan, the author examined cytotoxicity and inflammatory cytokines secretion with human leukemic mast cell line, HMC-1. HMC-1 was stimulated with phorbol 12-myristate 13-acetate (PMA) and calcium ionophore A23187. Sabaeksan by itself had no effect on viability of HMC-1. Result : The effects of Sabaeksan on the secretion of tumor necrosis factor-alpha (TNF-α) and interleukin (IL)-6 from HMC-1 were evaluated with enzyme-linked immunosorbent assay. Sabaeksan (1 mg/㎖) inhibited PMA plus A23187-stimulated TNF-α and IL-6 secretion, by 43.86 ± 5.26%, 56.39 ± 3.65%, respectively. Sabaeksan also inhibited the NF-κB (p50/p65) expression. Conclusion : Taken together, these results suggest that Sabaeksan inhibit the production of inflammatory cytokines in HMC-1 cells through blockade of NF-κB activation.
