        Pleurotus ostreatus에서 분리된 Glyoxalase I의 특성

        Glyoxalase I was purified 2,294-fold from Pleurotus ostreatus by S-hexylglutathione affinity chromatography, Sephadex G-150 gel filtration chromatography and DEAE-sepharose A-50 CL-6B ion exchange chromatography with an overall yield of 21.7%. The molecular mass determined by gel filtration was found to be approx. 34 kDa. SDS-PAGE revealed that the enzyme consists of two identical subunits with a molecular mass of approx. 17 kDa. The K sub(m) values of this enzyme for methylglyoxal and phenylglyoxal were 0.39 mM and 0.22 mM, respectively. And this enzyme had a strong affinity for L-xylosone and hydroxypyruvaldehyde. The enzyme showed its optimal activity at pH 6.5-7.5 and at $40^{\circ}C$. $^1H$-NMR spectroscopic analysis of enzymic reaction showed that this enzyme catalyzes intramolecular proton transfer. Pleurotus ostreatus로부터 glyoxalase I(S-lactoyl-glutathione methylglyoxal lyase, EC 4.4.1.5)이 S-hexylglutathione affinity chromatography, Sephadex G-150 gel permeation chromatography, DEA-sepharose A-50 CL-6B ion exchange chromatography를 통해 순수 분리되었다. 이 결과, 전체 활성도의 21.7% fmf 수확하였으며, 분리 배수는 2,294 배 이었다. Gel filtration chromatography로 측정한 효소의 분자량은 34 kDa이며, SDS-PAGE 결과 본 효소는 분자량 17 kDa인 동일한 소단위체 두 개로 구성된 이합체라고 생각된다. Methylglyoxal과 phenylglyoxal에 대한 $K_m$ 값은 각각 0.39 mM 과 0.22 mM 이며 L-xylosone과 hydroxypyruvaldehyde에 대해서도 강한 친화력을 보여주었고, pH 6.5~7.5, $35~45^{\circ}C$에서 활성도가 가장 높았다. 이 효소의 반응 과정을 핵자기공 명분광법으로 분석한 결과, 분자내의 양성자 전달과정이 뚜렷이 관찰되었다.

        This study compared and analyzed the core theory on educational autonomy, the local educational autonomy system in foreign country, and a revised bill of the law on the local educational autonomy which is pending in the National Assembly. The study sought for a direction that needs to be pointed by the decentralized educational autonomy system in line with a real situation of Korea, based on the general opinions and views in members of educational board and main bodies related to education nationwide in terms of pending problems and issues of educational autonomy. The local educational autonomy system may be consistent with the constitutional spirit and vitalizes the essence in education when being separated and becoming independent of general administration. However, it needs to seek for a plan that maintains the mutually connective and cooperative system with general autonomy aiming at responsibility and right, efficiency in administration, and stable security in educational finances. According to the results of the study, it needs to be also sought a plan of blocking the dual deliberation and inspection system through supplementing the educational board system of local assembly, which is operated, by way of showing an example by Jeju Special Self-Governing Province. And, the election system a member of educational board and a superintendent needs to be changed into a semi-direct election system or resident direct-election system, and the standard for qualification of educational career needs to be maintained. In conclusion, the local educational autonomy needs to be revised a statute so that the interest of education beneficiary group can be reflected rather than a education related group, on the basis of harmony in autonomy and professionalism of education in addition to ideology of democracy called citizen autonomy.

        초등교육의 위상

        Elementary education has great influence on the life of the people. But elementary teachers are not properly treated in terms of social recognition and economic reward, taking the problem of teacher quality into consideration. Therefore some drastic change in educational policy is required. The following are the proposals to straighten out the present status of elementary education. 1. Through the enactment of laws, teachers' social status as professionals should be enhanced, the climate where teachers are respected should be restored, and parents' educational orientation valuing competition and egoism should be changed. 2. Teachers should receive appropriate amount of economic reward as professionals, and various welfare pokicies should be enforced. 3. Centralized bureaucratic dysfunction should be wiped out in educational administration and the entrance examination as well as the content and implementation of curriculum in the teachers college should be improved. 4. Educational environment should be improved. The positions of educational professionals and administrators should be distributed equally to elementary and secondary level. 5. Principals should get rid of authoritarian attitude and try to enhance the morale and welfare of teachers with technical, human relations, and conceptual skills. 6. The amount of non-professional duties should be reduced and the apportunities for self-directed study and inservice education should be increased. 7. Teachers should build up their character and teach with loving, fare, and firm attitude with pride for their work. These proposals are related to nation, society, administrators, and teachers themselves. With the combined effort of all these concerned, the morale of teachers will be enhanced, elementary education will be renovated, and "education for creativity" will be realized.
