[산ㆍ학ㆍ연 논단] 버섯균사체 및 청국균 발효대두의 항산화능과 암세포 증식억제능

콩의 배당체 isoflavone류를 생리 활성형 aglycone으로 전환율을 증진시켜 콩 발효식품의 기능성을 증진시키고자, 식용버섯 균사체와 청국장 발효균 B. megaterium SMY-212를 이용하여 단독과 2단계 배양한 발효대두의 메탄을 추출물에 대한 항산화능과 인체 전립선암과 자궁암 세포주의 성장억제 효과를 조사하였다. 2단 발효대두 추출물의 수소공여능 효과는 1,000 μg/mL 농도에서 17.8~26.9%로서 단독 발효대두보다 30~40% 높았으며, 그 중에 동충하초와 SMY-212를 배양한 것이 가장 높았다. Linoleic acid에 대한 항산화 효과는 단독 발효대두는 동충하초가 가장 높았으며, 2단계 배양한 발효대두는 팽이균사체와 SMY-212가 오히려 높게 나타났다. 인체 전립선암 세포주(DU145)와 자궁암 세포주(Hela)에 대한 성장억제 효과는 단독배양에서 모두 팽이버섯 균사체를 이용한 발효대두가 가장 높았으며, 2단계 배양에서 DU145에서는 팽이버섯과 아가리쿠스 균사체 배양물에 SMY-212를 이용한 발효대두가 높았고, HeLa에서는 팽이버섯, 동충하초, 새 송이버섯 균사체의 추출물 순으로 높았다. 결론적으로 삶은 콩에 버섯 균사체의 단독배양보다는 버섯균사체와 SMY-212를 2단 배양한 발효대두 추출물이 암세포주 성장억제 효과가 높게 나타났다.

Fresh Omija (Schisandra chinensis) has good marketability, but its quality is difficult to maintain during storage and distribution. Freezing and freeze-thawing treatments can be utilized for the quality maintenance and processing of cold press juice. In this study, the color, antioxidant properties, and the major components of soaked liquor from Omija with freeze-thawing treatment were analyzed during the extraction periods. Each of the frozen and freeze-thawed Omija samples was soaked in 35% ethanol, extracted for 15 days, and used for analysis. The frozen and freeze-thawed samples showed a tendency toward better color and higher antioxidant activity and major component levels than the controls, and freeze-thawing was the best. The results of this study showed that freeze-thawing treatment improved the color, antioxidant properties, and level of the major components of Omija soaked liquor, and freeze storage is suitable for making soaked liquor.

Rehmannia glutinosa root (R. root) has been used as an traditional medicine, and is important resource for natural medicines and functional foods. However, R. root contains catalpol which is bitter, and undigested sugars, including stachyose and raffinose, which can cause diarrhea. Therefore, this study was performed to identify the changes in reducing sugar from undigested sugars and in catalpol contents in R. root slurry induced by aging treatments. R. root slurry was treated at $10{\sim}70^{\circ}C$ for up to 72 hr; and extracted with a 50% ethanol solution. The catalpol content was analyzed using HPLC-UVD. Reducing sugar content generated from undigested sugars was measured by the Nelson-Somogyi methods, and the reaction rates were calculated from their variation according to aging time and temperature. During the aging treatment, reducing sugar increased and catalpol decreased. Their formation and degradation rates were highest at $50^{\circ}C$ and $30{\sim}40^{\circ}C$, and their rates were $2.05mg/g{\cdot}hr$ and 23.09 to 23.33%/hr, respectively. These results indicated that aging treatment can positively affect the sweetness and digestibility of R. root slurry. Therefore, an aging treatment could be considered for improving the taste and digestibility of R. root.
