        방사면역학적 측적방법을 이용한 운동 선수들의 뇨중 T / E , T / LH 비율 측정 검사

        The studies reported that the urinary ratios of testosterone/epitestosterone (T/E) and testosterone/leutenizing hormone(T/LH) were increased significantly by the administration of anabolic steroids to normal subjects. Based on the report above, the T/E and T/LH ratios were investigated by a RIA method for the 194 urine specimens of the Asian Games atheletes. The normal distribution of the testosterone(T), epitestosterone(E), and leutenizing hormone(LH) values were established and the T/E and T/LH ratios were investigated for steroid doping control. The correlation study between the T/E and T/LH ratios indicates that the high T/E ratio is not necessarily associated with the high T/LH ratio within the population who have the normal values of the T/E and T/LH ratios. It was also found that the RIA performances of T and E assays were very sensitive and could quantitatively detect the T and E respectively without cross-reaction. The standard curve range covers a broad range of steroid values.

      • The Evaluation of T/E and T/LH Ratios in Athelete's Urine by the Radioimmunoassay Methods

        Anabolic steroid를 정상인에게 투약하면 testosterone/epitestosterone(T/E) 비율과 testosterone/leutenizing hormone(T/LH) 비율은 상승된다고 보고된 바 있다. 위와같은 보고를 근거로, 1986년도 제 10회 아시아 경기에 입상한 선수 및 그 후보선수의 뇨를 검체로 하여 RIA를 이용한 testosterone(T)과 epitestosterone(E) leutenizing hormone(LH)의 정량분석을 수행했으며, T/E, T/LH의 정상치 및 그 분포도를 확립하였다. 또한 T/E와 T/LH의 상관관계를 조사한 결과 T/E와 T/LH가 정상범위에 속할 경우에는 T/E가 높은 값을 갖더라도 T/LH가 반드시 높은 것으로 나타나지는 않았다. 이번에 사용한 RIA방법은 강도가 매우 높고 광범위한 농도에 걸쳐서 교차 반응없이 T와 E를 각각 정량적으로 측정할 수 있음을 확인하였다.         4년제 대학 취업준비생의 다차원적 완벽주의와 사회비교경향성에 따른 군집유형별 취업불안과 구직효능감의 차이

        본 연구의 목적은 취업준비생의 다차원적 완벽주의의 하위변인인 자기지향 완벽주의, 사회부과 완벽주의와 사회비교경향성에 따라 군집을 분류한 후 군집별로 취업불안과 구직효능감에 차이가 있는지 확인하는 것이었다. 이를 위해 온라인 설문을 실시하였다. 수집된 자료 중 389부를 SPSS 22.0을 사용하여 기술통계분석, 2단계 군집분석, 일원배치분산분석을 실시하였고 이를 통해 형성된 군집을 확인하고, 군집 간 차이를 분석하였다. 연구결과는 다음과 같다. 첫째, 4개의 자연발생적 집단이 형성되었다. 군집 1은 ‘비완벽주의-비사회비교 집단’, 군집 2는 ‘사회부과 완벽주의 집단’, 군집 3은 ‘완벽주의-사회비교 집단’, 군집 4는 ‘자기지향 완벽주의-사회비교 집단’으로 나타났다. 이러한 군집유형에 따라 취업불안과 구직효능감에 차이가 있는지 살펴본 결과는 다음과 같다. 취업불안은 군집 1, 군집 2, 군집 4에 비해 군집 3 완벽주의-사회비교 집단이 유의미하게 높게 나타났다. 구직효능감은 군집 1, 군집 2, 군집 3에 비해 군집 4 자기지향 완벽주의-사회비교 집단이 유의미하게 높게 나타났다. 본 연구의 결과는 4년제 대학 취업준비생의 완벽주의와 사회비교경향성에 따라 취업불안과 구직효능감이 다르므로, 상담장면에서 내담자가 어느 하위집단에 속하는지에 대한 평가와 이에 따른 차별적인 개입이 필요함을 시사한다. The purpose of this study was to determine whether there are differences in job-seeking anxiety and job-seeking efficacy between groups of job seekers according to self-directed perfectionism, socially prescribed perfectionism, and social comparison orientation. For this purpose, an online survey was conducted. Using SPSS 22.0, 389 of the collected data were used to conduct descriptive statistics, two-step cluster analysis, and one-way ANOVA. The results of the study are as follows. First, four naturally occurring clusters were formed. Cluster 1 was the Non-perfectionist-social comparison group; Cluster 2 was the Socially-prescribed perfectionist group; Cluster 3 was the Perfectionist-social comparison group; and Cluster 4 was the Self-directed perfectionist-social comparison group. Job-seeking anxiety was significantly higher in cluster 3 to clusters 1, 2, and 4. Job-seeking efficacy was significantly higher in cluster 4 to clusters 1, 2, and 3. The results of this study suggest that it is necessary to understand the differences in job-seeking anxiety and job-seeking efficacy among job seekers based on self-oriented perfectionism, socially prescribed perfectionism, and social comparison tendencies, and to provide differentiated interventions to clients.

        인터루킨 - 2 정량을 위한 효소면역 측정법

        To establish an enzyme-linked immunosorbent assay technique to quantitate interleukin-2 (IL-2), five different monoclonal antibodies against human recombinant IL-2 were prepared. A pair of monoclonal antibodies recognizing different epitopes on the IL-2 molecule were selected for sandwich ELISA. The procedure involved utilization of one specific antibody immobilized on a microtiter plate and the other labelled with alkaline phosphatase. A linear relationship was achieved, using concentrations of IL-2 up to 100 ng/ml, when the absorbance of the reaction mixture was measured on a spectrophotometer. The whole assay procedure can be completed within five h. This assay technique for IL-2 would provide a simple and rapid alternative to other assays such as bioassay using IL-2 dependent CTLL cells.

      • Production, Purification and Characterization of Monoclonal Antibodies Specific to Progesterone

        Progesterone-$11{\alpha}$-hemisuccinate-BSA를 제조하여 Balb/c 생쥐에 주사하여 얻은 비장세포를 myeloma 세포와 융합하여 여러 가지 subtype 형태($IgG_1$, $IgG_{2a}$, $IgG_{2b}$, IgM)를 가진 항체를 분비하는 7개의 하이브리도마 세포주를 얻었다. 이들이 분비하는 항체들을 간단히 ammonium sulfate 침전법, Protein A-Sepharose 4B 등으로 분리정제하여 그 특성을 조사하였다. 그 결과 단일클론 항체가 18-44% 정도의 수률로 SDS-PAGE상에서 순수분리 되었음을 확인하였다. 이들 항체들의 affinity constant는 $5{\times}10^7-4{\times}10^8M^{-1}$ 정도였으며, 이들 항체들은 Progesterone-3-(O-carboxymethyloxime)-BSA를 면역원으로 하여 얻은 항체와 비교했을 때, 특이성과 다른 steroid 호르몬과 교차반응성 등이 클론에 따라 특이하게 나타났다. 특히 이들 항체 중 051-01 clone은 다른 clone에 비해 매우 빠른 성장을 보였으며, 항체의 양이 복수액의 전체 단백질의 38% 정도나 되어 titer level이 매우 높았으며, 방사면역측정법이나 효소면역측정법으로 반응성을 측정했을 때 그 감도가 매우 좋았다. Progesterone $11{\alpha}$-hemisuccinate-bovine serum albumin conjugate was prepared using carbodiimide and then used as an immunogen. Spleen cells of Balb/c mice immunized with the immunogen were fused with mouse myeloma (P3-X63-Ag8.653) cells. Hybridoma cells were screened by radioimmunoassay using $^3H$-progesterone. The clones 051-01, 051-03, and 051-05 were purified using protein A-Sepharose 4B affinity chromatography utilizing their subtype characteristics. The purity of the isolated IgG was identified by SDS-PAGE. The progesterone binding capacity of the 5 monoclonal antibodies ranged from 7 to 72 pmole/mg IgG and the affinity constant ranged from $5.0{\times}10^7\;M^{-1}$ to $4{\times}10^8\;M^{-1}$ by the Scatchard analysis method. Cross-reactivities of the monoclonal antibodies, 051-01 and 051-02 with other steroid hormones were compared. These antibody reactions were very specific to progesterone and sensitive in a competitive immunoassay.

        인체내 혈청 중의 보체 C3 의 정제 및 특성연구

        The third component of complements (C3) has been purified from plasma (blood type: B) by sequential employment of QAE-Sephadex A-50 chromatography, 20% (w/v) PEG-3,350 precipitation, heparin-Sepharose CL-6B, and hydroxylapatite chromatographies. Final recovery of C3 was 57% as quantitated by single radial immunodiffusion (SRID), and purification fold was 78. The SDS-PAGE indicated that the purified C3 was homogeneous. Mercury ions or low concentration of magnesium ion showed stimulatory effects on the hemolytic activity of C3, which were inhibited by EDTA or high concentration of reducing agent. Degradation of C3 appeared to be selectively enhanced by several factors, while lysate of sheep red blood cells (SRBC) inhibited it.

        아세토니트릴 용매 중에서 Copper-1,5,9,13-Tetrathiacyclohexadecane착물의 전기화학적 거동과 그 분석적 응용

        비수용매인 아세토니트릴 중에서 Cu(Ⅱ)와 1,5,9,13-tetrathiacyclohexadecane[16-ane-$S_4$]의 착물에 대한 전기화학적 거동으로서 직류폴라로그램과 미분펄스폴라로그램으로부터 환원전류의 유형과 가역성을 조사하고, 이들 화합물의 분석적 응용으로서 수용액에 있는 Cu(Ⅱ) 이온을 염석추출법으로 정량하였다. 또한 아세토니트릴 용매 중에서 착물의 안정도 상수를 구하고, 분석적 응용으로 수용액 중의 Cu(Ⅱ)를 염석추출법으로 정량하기 위하여 추출조건, 곧 킬레이트와 염석제의 효과, pH 범위를 구하고 또한 Cu(Ⅱ) 이온을 정량하는데 있어서의 검량성과 공존이온 효과를 조사하였다. 실험결과로부터 환원 과정은 비가역적이었으며 환원전류는 확산지배적임을 알았다. 또한 아세토닐트릴 용매 중에서 착물의 log $K_f$의 값은 3.51이었으며 Cu(Ⅱ) 이온을 정량하는데 있어 공존하는 이온의 영향을 별로 받지 않아 선택성이 좋았으며 본 실험방법에 의한 정량한계는 60ppb 이었다. The electrochemical behaviors and analytical application of copper-1,5,9,13-tetrathiacyclohexadecane[16-ane-$S_4$] complex in acetonitrile(AN) solution have been investigated by the use of DC polarography and differential pulse polarography. Thus the formation constant of copper complex was $10^{3.51}$. Copper (Ⅱ) ion was found to form complex of 1-to-1 composition with [16-ane-$S_4$]. In addition, reduction step was irreversible and the reduction current was diffusion controlled. And the effect of concentration of the salting-out reagent and chelating agent and pH of aqueous phase on the determination of copper (Ⅱ) was investigated and diverse ion effect was discussed. By salting-out extraction technique, we can be determined until the concentration of copper (Ⅱ) of 60 ppb.
