Aralia elata, a common medicinal and culinary herb, has been consumed for centuries without any reported adverse effects. However, due to its limited safety information, we decided to investigate the repeated-dose toxicity of ethanolic extract of Aralia elata. The test was administered once daily by the gavage to male and female rats at doses of 0, 250, 500 and 1,000 ㎎/㎏/day for four weeks. Throughout the study, no treatment-related deaths or clinical signs were observed. Also, no apparent changes were detected in ophthalmoscopy, urinalysis, serum biochemistry, hematology and gross necropsy. The test result showed a significant decrease in body and heart weight of males treated with 250 ㎎/㎏ of extract of Aralia elata compared to normal control, a significant increase in relative brain weight and adrenal weight in females treated with 250 ㎎/㎏ of extract compared to normal control. However, all these changes were not considered toxicologically important due to irrelevant dose-response relationship to gross and microscopic findings. Histopathologically, abnormal changes were not observed in any target organs. On the basis of these results, the NOAEL of extract of Aralia elata was estimated to be more than 1,000 ㎎/㎏/day under the tested conditions.

Antihypertensive effects of ethanol extracts of Aralia elata (Miq.) Seem. (AE) were investigated in spontaneously hypertensive rats (SHR). SHR aged 14 weeks were treated for 8 weeks with AE (10 or 50 mg/kg/day) or amlodipine besylate (Am; 10 mg/kg/day) orally. Hypertension results in injury to several organs and can produce a significant increase in malondialdehyde (MDA) content as a result of lipid peroxidation and endothelial dysfunction. In this study, oral administration of AE and Am significantly reduced systolic blood pressure, organ weight index, and MDA content in tissues but increased significantly the plasma nitrite and nitrate concentrations. The endothelium-dependent relaxant activities of acetylcholine ($10^{-10}-10^{-3}M$) in norepinephrine (NE)-precontracted aorta were increased in AE- and Am-treated rats. Particularly strong endothelium-dependent relaxant activities were observed in AE-treated (50 mg/kg) rats. The endothelium-independent relaxant activities of sodium nitroprusside ($10^{-10}-10^{-3}M$) in NE-precontracted aorta were not changed. The results of this study suggest that AE has both antihypertensive and end-organ protective effects in SHR.

This study investigated the effects of onion juice on the serum lipid components and blood pressure in obese rats fed high fat diets. Thirty-nine of Sprague-Dawley rats were divided into six groups, and were treated for 8 weeks: (1) normal diet (ND); (2) high fat diet (HFD); (3) HFD for first 4 weeks and high fat diet with 40% onion juice for the last 4weeks (H-H+O); (4) HFD with 40% onion juice for 8 weeks (H+O); (5) HFD for first 4 weeks and ND for the last 4 weeks (H-N); (6) HFD for first 4 weeks and ND with 40% onion juice for the last 4 weeks (H-N+O). The rates of increasing body weight were reduced in H+O and H-H+O groups compared with HFD group. The levels of triglyceride, low density lipoprotein-cholesterol and total cholesterol in blood serum were significantly decreased in the H+O and H-H+O groups compared with the HFD group. Administration of onion reduced the size of adipocyte, steatosis, and serum hyperlipidemia in obese rats fed HFD. Moreover, the antihypertensive effects of onion were observed in obesity rat fed HFD. Overall results suggest that onion reduces the serum lipid components and improves hypertension in obese rat fed HFD.

This study was conducted to investigate the potential acute toxicity of Aralia elata by a single oral dose in ICR mice. Thirty mice of each sex were randomly assigned to three groups of 10 mice each. The test articles were administered once by the gavage to mice at dose levels of 0, 2,500 and 5,000 ㎎/㎏ body weight. The mortality and changes on body weight and clinical signs of gross observation were monitored for 14 days after dosing. At the end of 14-day observation period, all animals were sacrificed and complete gross postmortem examinations were performed. There were no dead animal and adverse effects on clinical signs, the body weight and the gross finding. As the results, we could not find any toxic effect at the dose levels of 2,500 or 5,000 ㎎/㎏ in mice and the minimal lethal dose was considered to be over 5,000 ㎎/㎏ body weight in mice.

A rapid and simple HPLC method has been developed for simultaneous determination of medimin E. a new a -tocopherol derivate and atocopherol in swine serum. The analytes were extracted with ethanol from 0.2 ml of swine serum and separated by HPLC on a C_(18) reversed-phase column using methanol as a mobile phase and UV detection a t 285 nm. The method was validated over a concentration range of 2-100 μg/ml with a correlation coefficient of 0.998 for medimin E and 0.2-10 μg/ml with 0.996 for a -tocopherol. The lower limit of quantification (LOQ) in serum were 2μg/ml for medimin and 0.2 μg/ml for a-tocopherol: intra- and interday coefficients of variation were ≤13.0% except for LOQ. Advantages of this validated assay include small samples volume. simplified serum extraction, excellent extraction recovery and fast run time. This method can be applied to analysis of medimin E and a-tocopherol in clinical pharma-cokinetics and biological activities studies.
