섬유아세포에서 민들레 추출물 함유제(AF-343)에 의한 제 1형 교원질 발현 증가

We previously reported that the extract of Taraxacum platycarpum (AF-343) had several biological properties such as skin hydration and anti-inflammatory effects, thereby AF-343 be a promising anti-atopic dermatitis agent. However, few studies have been conducted to evaluate its effect on modulation of extracellular matrix proteins in human skin fibroblasts. The purpose of this study was to investigate the expressions of type I collagen, MMP-1, Smad2/3, and TIMP-1 proteins in AF-343-treated human skin fibroblasts. Human skin fibroblasts were treated by various concentrations of AF-343 (0~2 mg/ml). The expressions of type I collagen, matrix metalloproteinase-1 (MMP-1), Smad2/3, and TIMP-1 proteins were analyzed by Western blot analysis. In addition, level of type I collagen mRNA was analyzed by CAT assay. Expression of type I collagen protein was increased in AF-343-treated human skin fibroblasts by dose and time-dependent manners. Consistent with this result, the expressions of phospho-Smad2/3 in skin fibroblasts were increased and MMP-1 expression was decreased by AF-343 treatment. TIMP-1 expression was not significantly changed in AF-343 treated skin fibroblasts. Extract of Taraxacum platycarpum (AF-343)-induced up-regulation of type I collagen expression was through increased expression of phospho-Smad2/3. These results were occurred combined with down-regulation of MMP-1 in skin fibroblasts. Taken together, this study indicated that AF-343 has property of the modulation of ECM in tissue as well as skin hydration and anti-inflammation.
소엽의 Ethyl Acetate 분획이 인체구강유상 피암종세포와 인체 피부흑색종세포에 끼치는 성장억제효과

The cytotoxicity of the ethyl acetate soluble fraction of Perilla frutescens on human oral epithelioid carcinoma cell and human skin melanoma cell lines was evaluated by 3-(4,5-dimethylthiazol2-yl)-2,5-diphenyl-2H-tetrazalium bromide (MTT) colorimetric method. The light microscopic study was carried out to observe morphological changes of cultured human oral epithelioid carcinoma cell lines (KB) and human skin melanoma cell lines (SK-MEL-3 Cells). These results were obtained as follows ; 1. In MTT assay, subfraction 1 and 2 were shown significant cytotoxicity at 10^-2㎎/㎖ concentration. 2. The comparison of IC_50 values of various fractions in human oral epithelioid carcinoma cell and human skin melanoma cell fines was exhibited that their susceptibility to theses fractions decreased in the following order: human oral epithelioid carcinoma cell lines: subfraction 5 < subfraction 4 < subfraction 3 < subfraction 2 < subfraction 1, human skin melanoma cell lines: subfraction 5 < subfraction 4 < subfraction 3 < subfraction 1 < subfraction 2 by MTT assay. In light microscopy, the subfraction 1 and 2 of the ethyl acetate soluble fracion of Perilla frutescens showed the highest cytotoxicity. These findings suggest that fracion 1 and 2 possess the most antitumorous agent.

The production and intensive use of organochlorine chemicals, both in agriculture and industry, have led to wide spread contamination of the environment. There may be biological effects of organochlorine contaminated breast milk even when levels are below those concentrations that are currently regarded as tolerable. To evaluate the exposure of to fetus PCBs, we measured the sum of 13 congeners of PCBs in maternal blood, cord blood, breast milk and placenta of 30 Korean women by gas chromatograpy/electron capture detector. The concentration of total PCBs of maternal blood, cord blood and breast milk were 7.16±4.29 ㎍/ℓ, 5.16±3.12 ㎍/ℓ, 1.77±2.87 ㎍/ℓ respectively and they have no significant correlation. PCBs in placenta was not defected. However there were no significant differences of PCBs according to age, education, body mass index, parity, breastfeeding, smoking and drinking.
