        돼지고기 중 알레르기 유발성분의 동정

        The pork meat been reported as one of the food occurring allergic reactions predominantly to korean. To identify the potential food allergens in pork meat, sera were collected from 25 allergic patients to the pork meat and 10 allergic patients not to pork meat as well as 5 normal subjects after skin prick test and open food challenge test. Crude extracts were prepared by blending raw pork meat in phosphate buffered saline (pH 7.0) and the heat treatment on crude extracts was carried to characterize sensibility of the allergens to heat. ELISA was performed to determine specfic IgE antibody levels of allergic patients to pork meat, and resulted in twofold higher mean value than that of tolerated patients. Extracted proteins from pork meat was separated with SDS-PAGE followed by immunoblotting using sera from pork sensitive patients and control subjects, respectively. The IgE binding response to pork meat by immunobots correlated with quentitative specfic IgE value of each person. Immunoblots showed four prominent IgE-binding bands (66, 60, 50, 44kDa) in crude extract, but two bands of those (60, 44kDa) were heat-labile. These results suggest that most prominent allergens from pork meat are four components(66, 60, 50, 44 kDa) in korean and the heat treatment on allergen is additional parameter to characterize allergen.

        선천성 간질쥐에서 청각자극에 의한 경련 유발 감수성과 경련강도의 변화

        Effects of acoustic stimuli on convulsion were determined in genetically epileptic rats. Changes of convulsive behavior were evaluated as seizure susceptibility and intensity by applying the audiogenic response score system (Jobe et al., 1973). The susceptibility and intensity of audiogenic seizure were not increased but induced earlier in life during the developmental phases of young pups by multiple acoustic stimuli applied when animals are young. The changes of intensity seemed to be a temporary phenomenon. In more mature animals the seizure susceptibility and intensity were not affected by multiple acoustic stimuli.

        쪽방 거주 장애인의 삶의 경험에 관한 연구

        본 연구에서는 쪽방촌에 거주하는 장애인들의 쪽방 유입 과정과 현재 생활에 대한 생애사 분석을 하였다. 특히 장애가 이들의 쪽방 유입과정 및 현재의 생활에 어떤 영향을 미쳤는지에 관하여 분석하였는데, 장애는 빈곤을 심화시켜 참여자들을 쪽방으로 유입시킨 주요원인이었으며, 탈빈곤할 수단인 노동능력의 감소를 야기한 조건이었고, 결국 수급자로서 만성적 빈곤상태에서 살아가도록 만든 요인이었다. 또한, 한편으로는 장애가 최저생계의 생활을 할 수 있는 수급권 보장의 도구로 기능하여 쪽방촌에서 안정적으로 생활하는데 도움이 되기도 했다. 쪽방촌 거주 현실에 대한 분석 결과, 쪽방 거주 장애인은 “빈곤과 불편의 생활화 및 장애와 갈등의 일상화”로 표현되는 힘든 생활을 하고 있었고, 이웃과 갈등 또는 협력을 하고 있었다. 본 연구는 “현실에서의 감사와 이웃에서 찾은 희망”으로 요약되는 긍정적빈곤문화를 창출해내고 있는 연구 참여자들의 삶을 통해서 사회복지 정책적 시사점을 제시한다. 쪽방거주 장애인의 탈빈곤과 자립을 돕는 정책 마련이 필요하며, 특히 지역사회 자조활동을 지원하는 지역복지 대응체계가 필요하다. 빈곤연구 및 장애인복지 연구의 영역에서 쪽방거주 장애인을 고려한 후속연구가 또한 필요하다. The purpose of this study is to understand persons with disabilities living alone in a single room occupancy community and to suggest social work strategies for them. For data analyses, the life history methods (LHM) which focuses on the personal life story was employed. A total of 6 participants were selected from the original data collected by a research team at Korea University for a research grant funded by the Korea Health Promotion Foundation. Their open coded data were analyzed for this study. As a result, disability led the participants to live in a single room occupancy of poor people villege, jokbang-chon. The persons with disabilities experience long lasting poverty, physical pain, and environmental limitations. They also experiences ‘ambivalent culture of poverty`` representing conflicts or cooperations among neighbors. Based on trusts and cooperations in this poor village, more supports for their self-help groups are necessary. The persons with disabilities living alone in a single room occupancy should be also considered in the on-going discussion of exit-poverty and disability policies.

        Sodium arsenite가 GM-CSF로 자극한 골수세포에 미치는 영향

        The effects of arsenic on the proliferation and differentiation of bone marrow cells were studied by treatment of bone marrow cells with sodium arsenite and/or granulocyte/macrophage-colony stimulating factor (GM-CSF). GM-CSF stimulated the bone marrow cells, and the maximum growth rate of bone marrow cells following exposure to GM-CSF for 1, 3, 5 and 7 days was 133, 275, 400 and 208% of the control respectively. When bone marrow cells were treated with sodium arsenite ranging from 0.01 to 50 μM for 1, 3, 5 or 7 days, the growth of bone marrow cells was inhibited by dose- and time-dependent manner at the concentration higher than 1 μM, and the mean IC50 value was 6.2, 7.5, 9.0 and 7.7 μM, respectively. Apoptosis was also observed in all groups treated with 10 μM sodium arsenite regardless of duration of treatment. By exposure of bone marrow cells to sodium arsenite concomitant with 10 μM GM-CSF for 1, 3, 5 and 7 days, IC50 value in each group was shifted to the higher value. These results indicate that fast proliferating bone marrow cells by stimulation with GM-CSF are likely to be less vulnerable to inhibiting effects of arsenic on the cell growth, and apoptosis induced by high concentration of sodium arsenite could be suggested as one of the mechanism for arsenic in treatment of leukemia.

        선천성 고혈압 쥐의 연령별 뇌조직 주의 모노아민 함량 및 MAO 활성 변화

        The age-related changes of monoamine contents and monoamine oxidase (MAO) activities in the whole brain and blood pressures were measured and compared every week during the ages of 5 weeks to 12 weeks in two types of animals; (1) spontaneously hypertensive rats (SHR) and (2) normotensive Sprague-Dawley rats (NR). Blood pressures in SHR began to rise at the ages of 7 or 8 weeks and were significantly higher thereafter than in NR. Abnormally low MAO activities in the brain of SHR were detected from the ages of 8 weeks. Comparisons were also made between brain monoamine-norepinephrine, dopamine and 5-hydroxytrytamine contents of SHR and those of NR at the same ages throughout the test period. In SHR, none of those monoamine values was consistently higher or lower than in NR during the test period. Protein contents of the brain between SHR and NR were not significantly different at any ages. These observations suggest that the abnormally low MAO activities in SHR brain may be one of the underlying neurological factors for the susceptibilities to hypertension and the deficit of MAO activities may be due to the changes in properties rather than in amount of this enzyme.

        피록시캄이 니페디핀의 약력학 및 약동학에 미치는 영향

        Because nonsteroidal anti-inflammatory drugs are reported to cause fluid retention and hypertension by inhibition of prostaglandin synthesis, the effects of piroxicam on pharmacodynamics and pharmacokinetics of nifedipine were studied in male spontaneously hypertensive rats. They received nifedipine (0.5mg/kg) alone or combined with piroxicam (5mg/kg) intravenously Plasma levels norepinephrine, an index of sympathetic stimulation, were measured prior to each treatment and 5 min after drug administration. Changes in blood pressure were examined serially and blood samples for analysis of nifedipine were also taken for 6hr following drug administration. Plasma nifedipine concentrations were assayed by HPLC and pharmacokinetic parameters were calculated. Blood pressure was reduced (p<0.01), but plasma norepinephrine level was increased (p<0.05) by nifedipine administration. Anti-hypertensive effect of nifedipine was potentiated (p<0.05) by piroxicam coadministration, but effect of nifedipine on plasma norepinephrine level was not affected. In case of rats received nifedipine and piroxicam, plasma nifedipine concentrations were higher (p<0.05) than those from rats received nifedipine alone at 2, 3, 4, 5 and 6 hours following drug administration. The area under the plasma concentration vs. time curve was increased (p<0.05), while the elimination rate constant was decreased (p<0.01) by piroxicam coadministration. No significant differences were observed in the plasma clearance, apparent volume of distribution and elimination half-life. Thus, piroxicam not only potentiated antihypertensive effect of nifedipine, but also altered nifedipine pharmacokinetics in the rats. It is concluded that the potentiation of nifedipine antihypertensive effect might correlate with the increment of its plasma concentration by piroxicam coadministration.

        이부프로펜이 씨프로플록사신의 생체이용률에 미치는 영향

        The effects of ibuprofen on bioavailability of ciprofloxacin were studied in rabbits. Animals were divided into three groups ; group A received 60mg/kg of ciprofloxacin; group B and C received 60mg/kg of ciprofloxacin with 60 and 240mg/kg of ibuprofen, respectively. Ciprofloxacin and ibuprofen were given by single oral administration. Serum concentrations of ciprofloxacin were measured by high performance liquid chromatography with UV detector and pharmacokinetic parameters were calculated. Area under the serum concentrations versus time curve (+/-S.E.) of ciprofloxacin were decreased in group B and C compared with group A (12.26+/-0.94 and 12.57+/-0.94 vs. 15.71+/-1.06mcg.hr/ml, p<0.05), whereas total clearances were increased (1.81+/-0.13 and 1.76+/-0.12 vs. 1.40+/-0.09 l/hr/kg, p<0.05). No significant differences in these parameters were observed between group B and C. Relative bioavailability of group B and C to group A were 78 and 80%, respectively. These results suggest that the coadministration of ibuprofen with ciprofloxacin may reduce the bioavailability of ciprofloxacin.

        적출관류 간에서 대황, 마황 및 황금이 7-에톡시쿠마린의 대사에 미치는 영향

        In order to study the effects of Rhei rhizoma, Ephedrae herba and Scutellariae radix on hepatic metabolism, we examined the pretreatment effect of those on the metabolism of 7-ethoxycoumarin (EC). Water extracts (1g/kg) of Rhei rhizoma, Ephedrae herba and Scutellariae radix were administered orally to rats for 7 days, respectively. Livers were then isolated and perfused with 100mcM EC for 2 hours. The metabolites of EC, 7-hydroxycoumarin, sulfate conjugate and glucuronide conjugate were measured in the perfusates. The amount of glucuronide conjugates was decreased in Rhei rhizoma pretreated rats (p<0.01), however. 7-hydroxycoumarin was increased in Ephedrae herba pretxeated rats (p<O.OD. We examined whether the change of enzyme activity is related to the change of cytochrome P4501A1 a n d P4502B1 mRNA level in the perfused rat liver, which are responsible for EC metabolism. CYPlAl a n d CYP2B1 mRNA level was increased, which was not statistically significant with rhei rhizoma nor ephedrae herba pretreatment. We also assessed the hepatotoxicity of Rhei rhizoma. Ephedrae herba a n d Scutellariae radix. The activities of ALT and AST were assayed at 24 hours after 7 days administration. Only the ratio of ALT over AST was increased in ephedrae herba pre treated rats (p<0.05). Lipid peroxidation was increased in Rhei rhizoma treatment (p<0.05). while histopathological examination performed after liver perfusion did not show a n y difference compared with vehicle treatment. These results suggest that Ephedrae herba pretreatment increases the o-deethy-lation of 7-ethoxycoumarin in rats, which may be mediated b y CYPlAl mRNA induction.

        The effects of five lupane-triterpenoids from leaves of two Acanthopanax spp., chiisanogenin, chisanoside and 22alpha-hydroxychiisanogenin, acakoreoside A and acantrifoside A on the mitogen-induced proliferation were investigated in vitro. T cell proliferation (TCP) to concanavalin A (Con A) and the B cell proliferation (BCP) to lipopolysaccharide (LPS) were increased by chiisanogenin. TCP to Con A was significantly increased by chiisanoside and acankoreoside A, but not affected by chiisanogenin, 22 alpha-hydroxychiisanogenin and acantrifoside A, BCP to LPS was significantly increased by acankoreoside A and acantrifoside A, and slightly increased by chiisanoside, chiisanogenin and 22 alpha-hydroxychiisanogenin.
