독활기생탕가미방(獨活寄生湯加味方)이 배양 척수운동신경세포에 미치는 영향

Objectives & Methods : To evaluate the mechanism of oxidative damage by Xanthine oxidase(XO) and hypoxanthine(HX)-induced oxygen radicals, XTT assay was carried out after the cultured mouse spinal motor neurons were preincubated for 5 hours with various concentrations of XO/HX. And DNA synthesis, PKC activity and c-fos immunohistochemistry were measured to evaluate the protective effect of Dokhwalgisaeng-tang(Duhuojisheng-tang: DGT), Dokhwalgisaeng-tanggami(Duhuojisheng-tangjiawei: DGTG), Herbar Chelidoni(HC) water extract on cultured spinal motor neurons damaged by XO/HX. Results : The results were XO/HX showed the quantitative decrease of DNA synthesis, increase of PKC activity and c-fos positive cell, in proportion to concentration and exposed time on cultured spinal motor neurons and DGT, DGTG and HC showed the protective effects against decrease of DNA synthesis and increase of PKC activity and c-fos positive cell by XO/HX. Conclusions : From the above results, it is suggested that XO/HX have a neurotoxic effect on cultured spinal motor neurons and the herbs extract, such as DGT, DGTG and HC, prevent the toxicity of XO/HX effectively.
부자반하탕(附子半夏湯) 추출액(抽出液)이 산소자유기(酸素自由基)에 의해 손상(損傷)된 배양척수운동신경세포(培養脊髓運動神經細胞)에 미치는 영향(影響)

Objectives : To evaluate the effect of Bujabanha-tang(Fuzibanxia-tang) water extract on cultured mouse spinal motor neuron which was inhibited by xanthine oxidase/hypoxanthine(XO/HX) Methods and Matrials: induced cytotoxicity, cell viability by the MTT and NR assay and PKC activity, cAMP assay examined after the cultured mouse spinal motor neurons were pre-incubated with various concentrations of Bujabanha-tang(Fuzibanxia-tang) water extract for 3 hours prior to exposure of XO/HX Result : 1. XO/HX, a oxygen radical, decreased the survival rate of the cultured mouse spinal motor neuron cells on NR assay and MTT assay. 2. Bujabanha-tang(Fuzibanxia-tang) have decreasing efficacy of PKC activity increased by XO/HX in cultured mouse spinal motor neuron. 3. Bujabanha-tang(Fuzibanxia-tang) have decreasing efficacy of cAMP amout increased by XO/HX in cultured mouse spinal motor neurons. Conclusions : Bujabanha-tang(Fuzibanxiatang) has marked efficacy as a neuroprotection for the damages caused by XO/HX-mediated oxidative process.
