        야교등의 항산화 및 간보호효과

        Objectives : Polygoni Multiflori Ramulus has been widely used as a traditional medicinal herb for the treatment of insomnia, limb pain and itch. The extract of Polygoni Multiflori Ramulus (PMRE) is known to have a modulatory effect of many inflammatory responses. This study was performed to investigate the hepatoprotective effect of PMRE against arachidonic acid (AA) + iron-induced oxidative stress on HepG2 cell and carbon tetrachloride ($CCl_4$)-induced liver injury on mice. Methods : The effects of PMRE on cell viability was assessed by MTT assay. And flow cytometric analysis was performed to estimate the effects on mitochondrial function. To investigate its underlying mechanism, apoptosis-related proteins were analysed by using immunoblot analysis. In addition, ICR mouse were administrated (po) with the PMRE (30, 100 mg/kg) for 3 days and then, injected (ip) with $CCl_4$ (0.5 ml/kg body weight) to induce acute liver damage. The level of pro-caspase-3 was measured. Results : Treatment of PMRE increased relative cell viability, prevented a cleavage of poly (ADP ribose) polymerase and pro-caspase-3, and also reduced mitochondrial membrane permeability against AA + iron-induced oxidative stress. In addition, PMRE treatment decreased liver injury induced by $CCl_4$, as evidenced by increases in pro-caspase-3 level. Conclusions : These results demonstrate that PMRE has an ability to anti-oxidant and hepatoprotective effect against AA + iron-induced oxidative stress and $CCl_4$-induced liver injury.

        죽엽 열수추출물의 염증억제 효과

        Objectives : Phyllostachys Folium is leaves of Phyllostachys nigra var. henesis $S_{TAPF}$. In the East Asian traditional medicine, the herb has been used to treat nasal bleeding, dysuria, epilepsy and etc. The present study was conducted to evaluate the anti-inflammatory effects of the Phyllostachys Folium water extracts (PFE) in vitro and in vivo model. Methods : Cell viability was measured by MTT assay after the treatment of PFE and NO production was monitored by measuring the nitrite content in culture medium. iNOS, COX-2, $I{\kappa}B$, $p-I{\kappa}B{\alpha}$ amd $NF{\kappa}B$ were detected by immunoblot analysis, and levels of cytokine were analyzed by sandwich ELISA kit. Anti-edema effect of PFE was determined in the carrageenan-induced paw edema model in rats. Results : LPS increased NO and cytokines levels compared with control, these increases were attenuated by PFE. In addition, LPS-induced pro-inflammatory proteins such as iNOS, COX-2 were down regulated by PFE. These anti-inflammatory effect of PFE results from inhibition of phosphorylation of $I{\kappa}B$ and translocation of $NF-{\kappa}B$. Conclusion : These results show that PFE has some anti-inflammatory effects which might play a role in gram-negative bacterial infection inflammation and $NF{\kappa}B$ activated diseases.

        야관문(夜關門)의 항염증효과(抗炎症效果)

        Objectives : Lespedeza Cuneata has been used to treat leukorrhea, asthma, stomach pain, diarrhea, acute mastitis, in Korean traditional medicine. According to recent studies, Lespedeza Cuneata has antioxidation, hypoglycemia, cell protective, insulin secretion, whitening, corpora cavernosa smooth muscle relaxation and antimicrobial activities, but it has been rarely conducted to evaluate the immuno-biological activity. The present study was examined to evaluate the anti-inflammatory effects of the Lespedeza Cuneata MeOH extract (LCE) in vivo and in vitro. Methods : In vitro, inflammatory mediators, such as cytokines, nitric oxide and prostaglandin E2 were detected after the addition of LPS with or without LCE in Raw 264.7 macrophage cell line. In vivo, anti-edema effect of LCE was determined in the carrageenan-induced paw edema model in rats. Results : In vitro assay, LCE decreased release of nitric oxide (NO) and prostaglandin E2 (PGE2) via suppression of iNOS and COX-2 expression. LCE inhibited the phosphorylation of IĸB indicating the suppression of NF-ĸB pathway. In vivo assay, LCE significantly inhibited the formation of paw edema induced by carrageenan injection in rats. LCE effectively inhibited increases of hind paw skin thickness and inflammatory cell infilterations. Conclusion : These findings demonstrate that LCE has inhibitory effect on inflammatory mediators in LPS-activated Raw 264.7 cells and on paw edema in carrageenan-stimulated rats, showing the possibility of anti-inflammatory use of Lespedeza Cuneata.

        배추김치의 자동화 제조 공정 중 미생물 분석

        본 연구는 자동화된 상업용 배추김치 제조공정 중에서 원료와 공정별로 미생물의 변화를 조사하고자 수행하였다. 김치제조에 사용되는 여러가지 재료별로 미생물 분포를 조사한 결과, 마늘, 생강, 고추가루에서 많은 수의 호기성 박테리아, 젖산균, Leuconostoc, 그리고 효모가 검출되었으며, 이로써 위 부원료들이 김치에 존재하는 미생물들의 주요 공급원임을 알 수 있었다. 김치를 제조하는 공정 별로 미생물 변화를 모니터링 한 결과, 배추절임 공정에서 총 균수가 증가하였고 그 이후 세척공정에서는 단지 미생물수를 1 log CFU/g 줄이는데 그쳐 미생물 수 감소효과는 적은 것으로 판명되었다. 효모 또한 다양한 부재료와 최종 김치에서 검출 되었다. 따라서, 김치 제품에 최종적으로 존재하는 미생물 수를 줄이고자 한다면 위의 원료 중에서는 위의 부재료에 대해, 그리고 제조공정에서는 절임에 사용되는 천일염과 절임탱크에 대해 적절한 살균대책이 강구되어야 하겠다. 또한 차염소산 살균 공정은 절임과 세척공정 사이에 위치해야만 가장 큰 효과를 기대할 수 있을 것으로 판단되었다. 또한 검출된 효모에 대한 저해 수단으로 적절한 저해제의 첨가도 고려되어야 하겠다. 본 연구의 결과는 공장김치의 HACCP 구축과 개선에 활용될 것으로 기대된다. The objective of this study was to analyze the microbial populations in the raw ingredients of kimchi and their changes during an automated commercial manufacturing process. High population numbers of total aerobic bacteria, lactic acid bacteria, Leuconostoc sp., and yeast were detected in garlic, ginger, red pepper powder and this result revealed that these ingredients were the major source of microbials in kimchi. Additionally, during the salting process of Chinese cabbage, rapid microbial growth was observed and the consecutive washing process was determined to be ineffective, lowering the microbial count by only one log reduction. Yeast was also detected in various ingredients. These results strongly suggest that, in order to lower the microbial population numbers in kimchi, the side-ingredients and salting process should be subjected to the appropriate sanitization or sterilization processes at the HACCP level. Beside, treatment of salted Chinese cabbage with sodium hypochlorite solutions after the salting step is recommended. To inhibit yeast growth, appropriate chemical treatment and approval of additive uses to control microbials should be considered. These experimental results and suggestions will be used to improve the kimchi manufacturing process in factories.

        신욕초기 어머니 됨과 돌봄 자신감

        Being a mother is motherhood, which means maternal role attainment and has to meet maternal identity and role confidence of caring infant. This study was designed to examine maternal identity as a motherhood and role confidence in the immediate postpartum period, and then explain the correlations of them. Data were collected for 2 months by questionnaires from 64 mothers in the hospital. The scales were to measure motherhood questionnaire which consisted of 11 items and role confidence questionnaire which consisted of 13 items. The analysis of data was done with SPSSWIN program for descriptive statistics and t-test, ANOVA, Pearson Correlation coefficients. The results were as follows: 1. Mean score of maternal identity of primipara was 55.41 and multipara was 53.81. In this outcome primipara's score was higher than multipara's. 2. Mean score of role confidence of primipara was 44.0 and multipara was 46.81. Multipara's score was higher than primipara's. 3. Correlation between maternal identity and role confidence was r=0.29 (P=0.03). This was an interesting result. Because many studies reported that primipara had a lower score of maternal identity and role confidence than multipara's. But this study showed that primipara's score of maternal identity was higher than multipara's. So nurse has to support primipara's identity as a mother, taking advantage of nursing intervention from prenatal to postnatal periods. Also nurse has to offer many opportunities to experience premiparas and multiparas how to care their infants.
