Isolation of a Leucoanthocyanidin Dioxygenase (LDOX) Gene from a Spray-type Chrysanthemum (Dendranthema × grandiflorum) and Its Colored Mutants

스프레이 국화 'Argus'의 꽃잎으로부터 $DgLDOX$의 전장 cDNA와 genomic DNA를 분리하였고, 감마선 변이원으로부터 유래된 3가지 화색변이체 사이의 다양한 유전자 특성들을 밝혀냈다. cDNA 영역은 1068bp이고 356 amino acid로 변환되었다. Genomic DNA의 크기는 'Argus'에서 1346bp이었고, 3가지 화색 변이체에서는 1363부터 1374의 크기를 나타내었다. $DgLDOX$ 유전자는 두 개의 엑손 사이에 하나의 인트론을 갖고 있는 구조이고, 그 크기는 'Argus'에서 112bp 이지만 3가지 화색 변이체에서는 128 혹은 137bp였다. 이것은 감마선 조사에 의해 인트론 부분에 유전자가 삽입됐다는 것을 나타낸다. DNA 분석 결과 국화의 게놈 내에서는 하나의 $LDOX$ 유전자를 갖는 것이 확인되었다. $DgLDOX$ 유전자의 발현 정도를 분석한 결과, 연분홍의 'Argus'와 두 개의 보라색 변이체(AM1 and AM3) 에서 높게 발현되었으나 흰색 변이체(AM2)에서는 매우 약하게 발현되었다. 이러한 결과들은 $DgLDOX$ 유전자의 인트론에 삽입된 유전자 조각 혹은 엑손 부위의 일부 아미노산의 변화에 의해서 변이체의 화색이 변할 수 있다는 것을 보여주고 있다. A full-length cDNA and genomic DNA of a $leucoanthocyanidin$ $dioxygenase$ ($DgLDOX$) gene was isolated from the petals of chrysanthemum 'Argus', and comparative features of the gene among three flower color mutants derived from a gamma-ray mutagenesis were characterized. The cDNA coding region of the gene was 1068 bp and was translated into 356 amino acids accordingly. The genomic DNA size was 1346 bp for 'Argus', while three mutants revealed ranges of 1363 to 1374 bp. A single intron between two coding exons for the $DgLDOX$ gene was found, of which size was 112 bp for 'Argus', but 128 or 137 bp for three flower color mutants, indicating that a genomic insertion in the intron occurred during the gamma-ray mutagenesis. DNA blot analysis revealed the $DgLDOX$ gene presenting as a single copy in the chrysanthemum genome. The $DgLDOX$ gene was expressed in both 'Argus' of light-pink color and two purple color mutants (AM1 and AM3) but had very weak expression in only white color mutant (AM2). The results demonstrated that variations in the flower color of the mutants might be associated with changes in the amino acid moieties in the coding exons or fragment insertions in the intron of the $DgLDOX$ gene, which potentially resulted in less expression of the gene in the white colored mutant.

한국판 가해행동 대인동기 척도(TRIM-12-K)의 타당화 연구

The current study was aimed to evaluate reliability and validity of the Transgression-Related Interpersonal Motivation inventory-12-Korean. 313 adults and college students responded to TRIM-12-K, FS(Forgiveness), KFS(Forgiveness), STAXI(anger), STAI (anxiety), and CES-D (depression). Exploratory and confirmatory factor analyses results supported two-factor(avoidance and revenge) model same as original TRIM-12. Internal consistencies for subscales of the TRIM-12-K were acceptable. Significantly high negative correlation between TRIM-12-K and FS and between TRIM-12-K and KFS indicated convergent validity. TRIM-12-K was positively associated with anger, anxiety, or depression as indices of psychological maladjustment. Theses findings suggest that the TRIM-12-K is valid and reliable to assess a degree of forgiveness. Limitations and suggestions for future research were discussed.

Polymorphic bands of two fine-leaf zoysiagrass mutants (CNU 70-1, CNU 70-2) induced via a gamma-ray irradiation on seeds of Zoysia japonica were obtained by using randomly amplified polymorphic DNA (RAPD) primers. The genotype-specific fragments were then converted into PCR-based sequence characterized amplified region (SCAR) markers, which are now amenable to detecting them among other zoysiagrass species widely noticeable in Korea. The CNU 70-1-specific primer set amplified about 900 bp successfully, while the CNU 70-6 marker produced the expected 1,500 bp band, by which those markers were nominated by CNU 70-1_900 and CNU 70-6_1500 SCARs, respectively. The developed SCAR markers can be an applicable tool in sod industry where illegal appropriation hampers breeder’s right and profits due to the turfgrass plant vegetatively propagating.

기독교 신앙과 심리학의 조화로운 관계 탐색: 재림교회를 중심으로

Although there have been many studies on the relationship between Christianity and psychology, there is little research on how to understand psychology in the Adventist church in Korea. The purpose of this study was to summarize the overseas studies related to the perspectives of the Adventists on psychology and to suggest future research directions. First, the researcher explained the relationships between Christianity and psychology by dividing them into confrontation type, subordination type, discrimination type, and integration type and stressed the necessity of integration. Second, by summarizing the Adventist’s position on psychology, present the belief in harmony of science and the Bible and Ellen White’s perspectives, and then summarized the activities and conclusions of the Committee on Adventist Approach to Psychology. Third, psychological studies on Adventists were summarized by the theme of Adventist Youth, Health Recognition of Adventists, and Mental Health of Adventists. Finally, it suggested the necessity of carrying out various studies on psychology from the perspective of Christian worldview and Adventist viewpoint

남자 청소년의 ADHD성향과 삶의 만족 간의 관계: 대인관계 형성과 인터넷 게임중독의 매개효과를 중심으로

이 연구의 목적은 남자 청소년의 ADHD성향과 인터넷 게임중독 간의 관계를 확인하고, 그 관계를 대인관계형성과 인터넷 게임중독이 매개하는지를 검증하는 것이다. 연구대상자는 남자 중·고등학생 285명이었다. 이 연구를 위해 세계보건기구의 자기보고식 ADHD척도, 대인관계 유능성 척도, 인터넷게임중독선별검사 및 삶의 만족 척도를 사용하였다. 병렬적 이중 매개효과는 PROCESS Macro 3.5 모형 4로 분석하였다. 분석 결과, 남자 청소년의 ADHD성향은 대인관계 형성 및 삶의 만족과 각각 부적 상관이 있었으며, 인터넷 게임중독과는 정적 상관이 있었다. 남자 청소년의 대인관계 형성은 삶의 만족과는 정적 상관이 있었고, 인터넷 게임중독은 삶의 만족과 부적 상관이 있었다. 또한 남자 청소년의 ADHD성향과 삶의 만족 간의 관계를 대인관계 형성과 인터넷 게임중독이 이중으로 매개하는 것으로 나타났다. 본 연구의 결과는 ADHD성향이 강한 남자 청소년의 삶의 만족 수준을 향상하기 위해서 대인관계 형성과 인터넷 게임중독에 개입할 필요가 있음을 시사한다. This study aimed to analyze the relationship between ADHD trait and life satisfaction among male adolescents, and to examine the mediating effect of interpersonal initiation and internet game addiction on that relationship. The participants were 285 male students from middle and high schools. For this study, World Health Organization ADHD Self-Report Scale, Interpersonal Competence Questionnaire, Internet Gaming Use-Elicited Symptom Screen, and Satisfaction with Life Scale were used. PROCESS Macro 3.5 Model 4 was used to analyze a double mediating effect. Results revealed that male adolescents’ ADHD trait was negatively correlated with interpersonal initiation and life satisfaction, while it was positively correlated with their internet game addiction. And, male adolescents’ interpersonal initiation was positively correlated with life satisfaction, whereas internet game addiction was negatively correlated with life satisfaction. In addition, male adolescents’ interpersonal initiation and game addiction were mediating ADHD trait and life satisfaction. These findings suggest that the interpersonal initiation and game addiction should be treated to improve life satisfaction of male adolescents with high ADHD trait.

대학생용 인성교육 효과 측정도구 개발 : 삼육대학교를 중심으로

본 연구의 목적은 삼육대학교 재학생들의 인성교육 효과 측정도구를 개발하고 타당화하는 것이다. 주요 연구 결과는 다음과 같다. 첫째, 삼육대학교의 교육이념을 재개념화하여 보편적인 인성교육의 영역과 차원을 연결시켜 측정도구의 구조를 4영역(도덕적 인지, 도덕적 정서, 도덕적 행동, 영적 요소)과 2차원(개인적, 관계적)으로 선정하였다. 둘째, 문헌고찰을 통해 수집한 인성 요인들을 4영역 2차원에 배정하면서 통합 및 정리하여 자기이해, 양심, 타인이해, 공동체의식, 자기존중, 인내, 공감, 자비, 자기관리, 책임, 소통, 돌봄, 기독교 세계관을 하위요인으로 선정하였다. 셋째, 하위요인의 조작적 정의에 부합하는 기존의 척도 문항들과 새로 개발한 문항들로 예비문항 130개를 만들고, 삼육대학교 재학생 392명을 대상으로 조사한 결과를 분석하여 최종문항을 58문항으로 정리하였다. 넷째, 개발된 측정도구의 구성타당도는 탐색적 요인분석과 확인적 요인분석과 수렴타당도 분석을 통해 검증하였고, 내적 일치도 계수를 통해 신뢰도를 검증하였다. 마지막으로 후속연구를 위한 제언을 하였다. The purpose of this study was to develop and validate a tool for measuring the effect of character education on Sahmyook University students. The major findings are as follows. First, by reconceptualizing the educational philosophy of Sahmyook University and linking the domains and dimensions of universal character education, the structure of the measurement tool is divided into 4 domains (moral cognition, moral emotion, moral behavior, spiritual element) and 2 dimensions (personal, relational) was selected. Second, the character factors collected through literature review are allotted to the 4 domains and 2 dimensions, and integrated and organized so that self-understanding, conscience, understanding others, community consciousness, self-respect, patience, empathy, compassion, self- management, responsibility, communication, care, and the Christian worldview was selected as a sub-factor. Third, 130 preliminary questions were made from the existing scale questions and newly developed questions that meet the operational definition of sub-factors, and the final questions were organized into 58 questions by analyzing the results of a survey of 392 students at Sahmyook University. Fourth, the construct validity of the developed measurement tool was verified through exploratory factor analysis, confirmatory factor analysis, and convergent validity analysis, and reliability was verified through internal consistency coefficient. Finally, suggestions for the future studies were discussed.
