      • Isolation and Purification of Protoplasts from PorPhyra tenera Thalli

        김 (Porphyra tenera)의 생물공학적인 육종법개발을 위해 그의 엽체를 효소처리하여 원형질체를 분리하고 정제하였다. 원형질체의 수율은 효소의 농도와 효소액에 의한 조직의 처리시간에 비례하였다. $12\%$의 전복 내장 건조 분말로 조제된 효소액으로 $22^{\circ}C$에서 3시간동안 처리했을때 약 $2.5\times10^6$ 개의 원형질체가 분리되었다. 원형질체들의 크기는 $9\~25\mu$m로 다양했으며 평균 14.5$\mu$m였다. Protoplasts were isolated enzymatically from thalli of Porphyra tenera in relation to its utilization in breeding. As the concentration of the enzymes and incubation period increased, so did the yield of protoplasts. When a 250 mg fresh weight of the thalli was incubated in the $12\%$ abalone digestive enzyme mixture at $22^{\circ}C$ for 3 hours, about $2.5\times10^6$ protoplasts were released. The size of protoplasts ranged between 9$\mu$m and 25$\mu$m, with an average of 14.5$\mu$m.

      • 지방산 분석을 위한 trimethylsulphonium hydroxide (TMSH) 조건 확립

        This study was carried out to develop a simple and rapid methylation method for fatty acids in fish oils which contain lots of polyunsaturated fatty acids. Methanolic rimethylsulfonium hydroxide (0.2 M in methanol, TMSH) was preprepared by trimethylsulfonium iodide (0.40 g, TMSI), 0.88g of silver oxide, and 10 mL anhydrous methanol in the test tube. The mixture allowed to react for several hours. If a positive iodide test resulted the agitation, centrifugation and testing procedure were prepeated until no iodide could be detected in the reagent solution. Generally the reaction was completed within two hours at room temperature. The transesterification of fatty acid of a squid oil was monitored with thin layer chromatography (TLC) by spot the mixture of the 250 μL of TMSH reagent and 500 μL sample oil (20 ㎎/mL). Usually the transesterification was occurred from 2 minutes and the reaction was completed in 14 minutes. The TMSH reagent compared with the commercial Prep Ⅱ esterification reagent. The time of methylation reaction was similar between the TMSH and Prep Ⅱ. For full reaction of transesterification, twenty minutes were recommended. The fatty acid content of squid oil obtained by transesterification was the highest in the 15 minutes of reaction. However, the contents was decreased at 20 minutes reaction. The methylation with TMSH showed a simple and rapid method for fish oil.

      • 동남참게 초기유생에 대한 생리 생태학적 연구

        The present experiment was designed to obtain the adequate condition of temperature, daylength and salinity on the gonadal maturation of adult female, spawning and hatching of egg, and growth of larvae of the freshwater crab Eriocheir japonicus. Gonadal maturation was influenced by water temperature, but not daylength. GSI of female crabs more increased in experimental regime of 18℃ than that of 10℃ without regard to photoperiod conditions(12L/12D and 15L/9D). However, in both photoperiod conditions of 26℃ regime, GSIs were not changed, and vitellogenic oocytes were not observed. Spawning was considerably influenced by water temperature and salinity without regard to daylength. Vitellogenic female crabs did not spawn at 10℃ in any conditions of salinity. However, at 18℃ and 26℃, over a half of rearing female crabs spawned in condition of 30% sea water(salinity 9.6‰) and 60% sea water(salinity 19.2‰), while no spawned females appeared in freshwater condition(salinity 0.0‰). In 20% sea water(salinity 6.4‰) and less conditions, ovigerous female crabs removed eggs from the abdormal egg-chamber of one s own self before hatching out. However, all ovigerous female crabs hatched out in 40% sea water(salinity 12.8‰) and more conditions. Hatching rates were 33.3% and 83.3% in 40% sea water condition and in 60% and 80% sea water conditions, respectively. In experiment of effects on temperature on hatch out of eggs, hatching rates of eggs were 66.7% and 83.3% at 18℃ and 26℃, respectively. whereas, at 10℃ and 32℃ conditions, ovigerous females died or removed eggs before hatching out. In investigation of days required to hatch out from brooding time according to the water temperature, required days to hatch out were 28.0±1.7 days in 18℃ condition and 14.0±0.3 days in 26℃. In 20% sea water and less conditions, zoea larva died before metamorphosing to larvae of megalopha stage. However, in 40% sea water and more conditions, zoea larva metamorphosed to megalopha stage. Rates of metamorphosed zoea larva to megalopha stage were 32%, 44% and 48% in conditions of 40%, 60% and 80% sea water, respectively. Under rearing condition of 80% sea water, all of the first stage of zoea larva reared at 10℃ did not metamorphose to the second zoea stage and than died within 24 days. At 15℃, 16% of zoea larva metamorphosed to the fifth zoea stage. However, larva metamorphsing to meglaopha stage were observed from 20 and 14 days rearing at 20℃ and 26℃, respectively.

        참조기 조미 건포류의 이화학적 특성

        본 연구는 참조기를 이용하여 조미액을 달리하여 4가지 건포류를 제조한 후 영양성분, 과산화물가, 대장균군수 및 관능평가를 실시한 결과 다음과 같은 결과를 얻었다. 건포류의 일반성분에서 수분은 다른 제품에 비해 녹차액을 사용한 B제품이 약간 높게 나타났으며, 조미 건포류의 단백질함량은 39~45%로 대체로 높게 나타났다. 지방함량은 녹차액이 함유된 B제품이 가장 낮았으며, 매운 조미료를 이용한 D제품에서 지방함량이 가장 높게 나타났다. 탄수화물 함량은 23~31%로 나타났으며, 매운맛을 내는 D제품의 탄수화물 함량이 가장 낮게 나타났다. 회분 함량은 특히 매운 조미료를 사용한 D제품에서 다른 제품보다 약간 높게 나타났다. 건포류의 구성 아미노산 중 함량이 가장 높은 것은 glutamic acid이었으며, 다음으로 aspartic acid, glycine, lysine 순으로 나타났다. 지방산의 구성에서 포화지방산의 비율은 50~51%범위로 거의 제품간의 차이가 나타나지 않았고, monoenes의 비율은 로즈마리액을 처리한 제품의 비율이 가장 낮았으며, polyenes의 비율은 상대적으로 로즈마리액을 처리한 제품이 높게 나타났다. 과산화물가가 가장 낮은 제품은 로즈마리액을 첨가한 C제품이었으며, 매운맛을 내는 D제품이 가장 높은 값을 나타냈다. 대장균군수는 모든 제품에서 모두 음성으로 검출되지 않았다. 관능평가결과 맛, 색의 경우 모든 제품이 쥐포와 유의적인 차이가 없었으며, 냄새의 경우 매운맛을 내는 D제품은 쥐포와 차이가 없었으나, A, B, C제품은 모두 쥐포에 비해 유의적으로 낮게 나타났다. 전반적인 좋아함에서는 모든 제품이 쥐포와 차이가 없었다. To utilize the small redlip croaker, four kinds of seasoned fillet were produced. The nutrient composition, peroxide value and coliforms count of the products were determined, and sensory characteristics were evaluated. The seasonings consisted of the formula for commercial dried file fish added with water (A, control), green tea extract (B), rosemary extract (C) and soypaste with red pepper (D). The crude protein and carbohydrate contents of the products were in the range of 39~45% and 23~31%, respectively. The lipid content of product B was the lowest, while that of product D was the highest among the tested products. All products showed similar amino acid profiles with a high content of glutamic acid, aspartic acid, glycine and lysine. The saturates in fatty acid composition were similar (50~51%) among the products. However, the polyenes were higher (17%) in product C than products A, B and D (13%). The peroxide value of product C was the lowest among the products. There were not significant differences in taste and color among the products.

        Plant regeneration via somatic embryogenesis from soybean cotyledon culture

        The experiment was attempted to regenerate whole plant from somatic embryo using immature cotyledon cultures of five soybean varieties under different media. Normal shape somatic embryos were formed with 53.3% under MS medium supplemented with 1.0mg 2,4-D and 0.2mg/ℓ kinetin, and 32.2% on MS medium added with 8.0mg/ℓ NAA. Root and shoot was developed from the embryos under supplement with 0.1mg IBA and 0.25 mg ℓ│2-iP│of│ B5 medium, and whole plants were regenerated under further culture on hormoneless MS medium. Ulsan variety showed a good producer for somatic embryo among the lines applied. From subcultured somatic embryo callus, embryo formation is possible although their germination and organ differentiation is not easy.

      • 6종 갑각류 껍질의 키틴 및 구성아미노산 함량

        To obtain basic data about crustacean shell, chitin and amino acid content of six species of crustacean shells(Antarctic krill, Euphausia superba; Red snow crab, Chinonecetes japonicus, Daelongsuyum shrimp, Solenocera prominetis, Lobster, Linuparus trigonus; Gasibal shrimp, Nephrops thomsoni; Blue crab, Portunus trituberculatus) were investigated. The chitin content of six species of crustacean shells extracted by Hackman method ranged from 11.2 to 22.7% and that of the leg shell of red snow crab was the highest of them, holding 22.7%. But the chitin contained a small amount of protein, in the range of 0.5 to 3.1%. Total amino acid content of the six species of decalcified shells ranged from 55856.1 mg/100g to 30807.6 mg/100g (55856.1 mg/100g in antarctic krill shell, 37983.4 mg/100g in red snow crab shell, 41059.3 mg/100g in daelongsuyum shrimp shell, 36275.2 mg/100g in lobster shell, 33114.9 mg/100g in gasibal shrimp shell, and 30807.6 mg/100g in blue crab shell). The great portion of amino acids in these decalcified shells was occupied by aspartic acid, glutamic acid, and histidine. In the case of decalcified shell of gasibal shrimp, histidine was responsible for 59.8% of the total amino acid.
