분별탐색을 이용한 온주밀감 성숙 과실의 과발현 유전자 분리 및 동정

과실의 성숙은 식물의 다른 조직에서 볼 수 없는 특이적인 과정이다. 이 과정에서 과실의 품질에 관여하는 색깔, 당함량, 향기 등의 변화가 특징적으로 나타난다. 이와 같은 과실의 성숙을 이해하고 육종에 이용하기 위해서는 기관 특이적으로 발현되는 유전자들에 관한 연구가 필요하다. 감귤의 성숙과정에서 과실 특이적인 발현이 예상되는 유전자들을 분리하기 위하여 잎과 과실의 mRNA를 이용하여 분별탐색을 수행한 결과 과실의 cDNA 유전자 은행으로부터 Lea, 피루브산 키나아제, 탈수소효소, 신호인지입자 등과 상동성을 보이는 유전자 단편과 상동성이 보고되지 않은 다른 과발현 유전자들을 분리하였다. 온주밀감의 과실에서 분리된 Lea 유전자를 비롯한 3종류의 유전자들을 이용하여 잎과 과실에서의 발현량을 비교한 결과 cDNA 유전자은행으로부터 잎보다 과실의 탐침에 의해 우세한 신호를 보인 것처럼 각 유전자는 과실에서 과발현 되었다. 과실에서 우세하게 발현되는 분리 유전자들은 과실에서 발생하는 스트레스 저항성과 당대사 등의 과실 내의 생리적 기작과 매우 밀접한 관련성이 있음을 제시해준다. Fruit ripening is a specialized process that can not be observed in other plant organ. Research about genes relating to major characteristic of fruit such as color, sugar content, and aroma is required to understand the ripening process and to breed new cultivar by molecular engineering. Through differential screening between leaf and fruit mRNA, we isolated six genes including Lea (late-embryogenesis-abundant) protein, pyruvate kinase, signal recognition particle protein, dehydrogenase and two unknown genes, whose transcripts were dominantly detected in the fruit. The expression analysis showed stronger signal in fruit than in leaf, which is matched to the results of differential screening. The results suggest that fruit-dominant genes are related to the physiological functions including stress-responsive and sugar metabolism in fruits.
