Cytochrome P450 3A4 (CYP3A4), is the dominant human liver hemoprotein enzyme localized in the endoplasmic reticulum (ER), and is responsible for the metabolism of more than 50% of clinically relevant drugs. While we were studying CYP3A4 expression and activity in human liver, we found that anti-CYP3A4 antibody cross-reacted with a lower band in liver cytoplasmic fraction. We assessed the activities of CYP3A4 and its truncated form in the microsomal and cytoplasmic fraction, respectively. In the cytoplasmic fraction, truncated CYP3A4 showed catalytic activity when reconstituted with NADPH-cytochrome P-450 reductase and cytochrome b5. In order to determine which site was deleted in the truncated form in vitro, we transfected cells with N-terminal tagged or C-terminal tagged human CYP3A4 cDNA. The truncated CYP3A4 is the N-terminal deleted form and was present in the soluble cytoplasmic fraction. Our result shows, for the first time, that N-terminal truncated, catalytically active CYP3A4 is present principally in the cytoplasm of human liver cells.

6M KOH 수계 전해액에 potassium poly(acrylate) (PAAK)를 3 wt% 포함시켜 제조한 하이드로겔을 poly(acrylonitrile)부직포 분리막에 코팅하고, 이를 활성탄 수퍼커패시터의 분리막 및 전해질로 사용하여 수퍼커패시터의 고율특성 향상을 시도하였다. 이 분리막 및 전해질은전자현미경 관찰 결과 PAAK 하이드로겔이 부직포의 표면기공에 균일하게 코팅되어 있으며, 24일 동안 하이드로겔의 합습도가 230% 이상으로 균일하게 유지되었고, 6 M KOH 전해액을 사용한 경우(3.6×10-2 S cm-1)보다 약간 낮은 2.9×10-2 S cm-1의 이온전도도를 나타내었다. 활성탄을 활물질로 사용한 대칭형 수퍼커패시터에 이 분리막 및 전해질을 채택한 경우 사이클릭볼타메트리 시험에서 1000 mV s-1의 고속스캔 조건에서도 27 F g-1 이상의 높은 비축전용량과 1000 사이클 경과후에도 97% 이상의 유지율을 나타내는데, 이는 부직포 상에 코팅된 PAAK 하이드로겔 전해질이 활성탄 전극과 부직포 분리막 사이에서 강력한 계면밀착을 유지할 수 있기 때문이다. A hydrogel electrolyte consisting of potassium poly(acrylate) (PAAK) (3 wt%) in 6 M KOH aqueous solution is coated on poly(acrylonitrile) nonwoven separator to examine high-rate characteristics of activated carbon supercapacitor adopting the separator. The hydrogel is homogeneously coated on the surface pores of the nonwoven separator. The electrolyte uptake of the PAAK hydrogel maintains for 24 days higher than 230% and the coated separator shows slightly lower ionic conductivity (2.9×10-2 S cm-1) than that (3.6×10-2 S cm-1) of using 6 M KOH only. The activated carbon supercapacitor adopting the coated separator shows a specific capacitance higher than 27 F g-1 at 1000 mV s-1 and a retention ratio higher than 97% after the 1000th cycle. This is due to strong interfacial contact of coated hydrogel electrolyte between the activated carbon electrode and the nonwoven separator.

Purpose: The increased incidence of asthma due to rising allergic diseases requires the prevention of worsening asthma. It is necessary to develop a patient-tailored asthma prediction model. Methods: We developed causative factors for the asthma forecast system: infant and young children (0-2 years), preschool children (3-6 years), school children and adolescents (7-18 years), adults (19-64 years), old aged adult (>64 years). We used the Emergency Department code data which charged the short-acting bronchodilator (Salbutamol sulfate) from Health Insurance Review and Assessment Service for the development of asthma prediction models. Three kinds of statistical models (multiple regression models, logistic regression models, and decision tree models) were applied to 40 study groups (4 seasons, 2 sex, and 5 age groups) separately. Results: The 3 kinds of models were compared based on model assessment measures. Estimated logistic regression models or decision tree models were recommended as binary forecast models. To improve the predictability, a threshold was used to generate binary forecasts. Conclusion: We suggest the binary forecast models as a patient-tailored asthma prediction system for this category. It may be needed the extended study duration and long-term data analysis for asthmatic patients for the further improvement of asthma prediction models. (Allergy Asthma Respir Dis 2016:4:328-339)

Background: The Kimura disease, a chronic inflammatory disorder of unknown etiology, is characterized by subcutaneous mass, regional lymphadenopathy, peripheral blood eosinophilia or increased serum immunoglobulin E level. So far, there has been limited information concerning this disease. Objective: To investigate clinical features of Kimura disease in Korea. Method: We retrospectively reviewed 33 patients who had a histologically proven Kimura disease from January 1995 to March 2010 at Hospital. Then clinical features and treatment outcomes were evaluated. Result: Of all patients, 28 were male (84.8%), and mean age was 25.2±15.1 years. Involved sites were as follows; preauricular (n=8), upper arm (n=8), retroauricular (n=6), submandibular (n=4), eyelid (n=4), inguinal (n=4), and cervical area (n=4). Peripheral blood eosinophilia was observed in 87.1% of patients. Twenty-seven patients were treated by operation, corticosteroids, or radiation, and the other 6 were just followed without any intervention. Among the 27 patients treated, only 12 have achieved a state of control. Treatment outcomes in terms of size and mass numbers were variable. Conclusion: Kimura disease should be ruled out in young male who shows painless soft tissue swellings with peripheral blood eosinophilia in Korea. The optimal management for Kimura disease needs to be investigated further. (Korean J Asthma Allergy Clin Immunol 2010;30:285-292)
병적인 난관폐쇄 치료를 위한 미세난관문합술 35 예에 관한 고찰

1984년 1월 1일부터 1986년 12월 31일까지 서울대학교병원 산부인과에 입원하여 순수하게 난관문합술만을 시행한 환자 중 12개월 이상 추적조사가 가능했던 35예를 대상으로 분석을 하여 다음과 같은 결론을 얻었다. 1. 총 임신율은 34.1%(12/35)이었으며 자궁외임신율은 8.6%(3/35)이었다. 2. 수술로부터 임신까지의 기간은 평균 12.3 7.1개월이었으며 1년이내에 임신의 58.4%가 이루어졌고 2년 내에 모든 임신이 이루어졌다. 3. 35세 이상 군에서 임신율의 급격한 하락을 보였으며 38세이상에서는 임신이 된 경우가 없었다. 4. 양측 모두 문합술이 시행된 경우의 임신율은 50%이었으나 한쪽만 시행된 경우에는 17.6%에 불과하였다. 5. 병리소견상 빈도는 특별한 병변이 발견되지 않은 경우가 14예로 가장 많았고 만성난관염, S.I.N., 자궁내막증, 결핵성난관염의 순서였다. 6. 병리소견상 특별한 병변이 없었던 경우나 자궁내막증의 경우에는 비교적 임신성적이 좋았으나, 만성난관염이나 S.I.N. 또는 결핵인 경우에는 성적이 저조하였다. 7. 수술부위에 따른 임신율은 원위부로 갈수록 감소하는 경향을 보였다. 이상의 결과로 미루어 볼 때 병적인 원인의 난관근위부 및 중위부폐쇄에 있어서 난관문합술이 체외수정보다는 우선적으로 시도되어야 할 것으로 보인다. 그러나 임신결과에 영향을 미칠 수 있는 요인들을 고려하여 치료방침을 결정한다면 더 좋은 결과를 기대할 수 있을 것으로 사료된다. Between the period from January 1, 1984 to December 31, 1986, 49 pure cases of tubal resection and anastomosis were performed in the department of Obstetrics and Gynecology, Seoul National University Hospital. Of these only the cases followed up for more than 12 months(35 cases) were included and analyzed according to age, pathology and procedure. The pregnancy rate at the end of 2 years of observation was 34.1%. The intrauterine pregnancy rate was 25.7% and the tubal pregnancy rate was 8.6%. All pregnancies were occurred during the first 24 months and the average interval from operation to conception was 12.3 months. Microsurgical tubal resection and anastomosis should be the primary approach over IVF-ET in the management of pathologic proximal and midtubal occlusion until now.
