Boscalid는 anilide계 살균제로서 잿빛곰팡이병, 흰가루병, 균핵병등 다양한 식물병을 방제하기 위해 널리 사용되고 있지만 후작물로 재배되고 있는 알타리무에서 잔류량의 평가에 대한 연구는 미비하다. 본 연구는 boscalid의 알타리무에서 잔류성을 평가하여 PBI 설정자료 및 안전성 평가를 위한 기초자료로 활용하고자 하였다. Boscalid의 잔류량 평가시험은 지리적으로 다른 지역의 시설 재배에서 실시하였다. Boscalid 입상수화제를 나지토양에 처리한 후30일(PBI 30)과 60일(PBI 60)에 알타리무를 파종하였다. 알타리무는 일정기간 동안 재배한 후 일시적으로 수확하여 boacalid의 잔류량 분석을 위해 사용하였다. Boscalid의 잔류량 평가는 QuEChERs 방법을 기반으로 개발된 시료 전처리 방법과 LC/MS/MS 분석법을 이용하여 수행하였다.

Green peach aphid (Myzus persicae) and Cucumber mosaic virus (CMV) are one of the most harmful insect and plant disease affecting agricultural productivity worldwide. In the present study, we isolated microorganisms exhibiting dual control activity against M. persicae and CMV. Among 103 bacterial isolates, an isolate bacteria showed significant insecticidal and antiviral activity. The isolate was named as a Pseudomonas sp. DJ15 (DJ15) based on 16S rRNA sequence analysis. The cell-free supernatant of DJ15 showed 100% of aphid mortality. The cell-free supernatant reduced the symptom severity in pepper plants infected with CMV. These results suggest that DJ15 is potential to control M. persicae and CMV for their potential to contribute toward management of viral plant diseases.

In 2003, the invasive hornet, Vespa velutina, arrived in southern Korea and spread rapidly. V. velutina has been a serious threat to honeybee colonies. Many studies have conducted to limit the impact and proliferation of V. velutina based on nest destruction, trapping, population genetics and biological control. This study investigated the attractive effects of volatile organic compounds (VOCs) produced by microorganism. Microorganisms were isolated from agricultural fermented broth, and an isolate was identified as Bacillus jeotgali BV-1 based on 16S rRNA sequence analysis. The VOCs were extracted using a Solid Phase Microextraction (SPME) technique and determined by Gas Chromatography-Mass Spectrometry (GC/MS). The total number of VOCs identified from B. jeotgali BV-1 culture were 9 and 12 at 30% and 50% sugar medium, respectively. The most abundant is 3-methyl butanol. Based on the data, B. jeotgali BV-1 was suggested as a potential agent for attracting V. velutina.
Streptomyces sp. J46의 세균성구멍병원균 Xanthomonas arboricola pv. pruni에 대한 항균 활성

BACKGROUND: Bacterial shot hole of stone fruits is a seriuos plant disease caused by Xanthomonas arboricola pv. pruni (Xap). Techniques to control the disease are required. In this study, microorganisms with antibacterial activity were isolated to develop as a microbial agent against the bacterial shot hole. METHODS AND RESULTS: An isolate with the strongest activity among the isolates was identified as Streptomyces avidinii based on 16S rRNA gene sequence analysis and designated Streptomyces sp. J46. J46 showed suppression of bacterial leaf spot with a control value of 90% at 10 times-diluted cell free supernatant. To investigate antibacterial metabolites produced by J46, the supernatant of J46 was extracted with organic solvents, and the extracts were subjected to chromatography works. Antibacterial metabolites were not extractable with organic solvents. Both reverse and normal phase techniques were not successful because the metabolites were extremely water soluble. The antibacterial metabolites were not volatiles but protein compounds based on hydrolysis enzyme treatment. CONCLUSION: Our study suggests that Streptomyces sp. J46 may be a potential as an microbial agent against bacterial shot hole. Further study to identify the metabolites is required in more detail.
유전 및 육종 : 전장 유전체 연관분석을 통한 한우 성장 연관 양적형질좌위 (QTL) 탐색

본 연구는 한우 거세우 266두에서 유전자형 결정이 완료된 4,522개의 SNP를 이용하여 한우 성장형질 (6, 12, 18 및 24개월 령 체중)에 대한 양적형질좌위 (QTL)을 탐색 하였다. 각 SNP와 성장형질과의 연관성 분석은 회귀분석 (single marker regression) 을 이용하여 수행하였으며, 통계적 유의성은 P-value (P<0.001)로 설정하였다. 그 결과, 6개월체중에서 3개 좌위, 12개월 체중에서는 5개 좌위, 18개월체중에서 5개좌위 그리고 24개월체중에서 4개 좌 위가 통계적 유의차를 보였다. 통계적 유의차를 보인 SNP의 상가 적 유전분산을 분석한 결과, 몇몇 SNP에서는 6~11% 정도의 상가 적 유전효과를 보였으며, 대부분의 SNP들은 2~5%로 매우 작은 효과를 보였다. Genome-wide association study was performed on data from 266 Hanwoo steers derived from 66 sires using bovine 10K mapping chip in Hanwoo (Korean cattle). SNPs were excluded from the analysis if they failed in over 5% of the genotypes, had median GC scores below 0.6, had GC scores under 0.6 in less than 90% of the samples, deviated in heterozygosity more than 3 standard deviations from the other SNPs and were out of Hardy-Weinberg equilibrium for a cut-off p-value of 1-15. Unmapped and SNPs on sex chromosomes were also excluded. A total of 4,522 SNPs were included in the analysis. To test an association between SNP and QTL, a single marker regression analysis was implemented in this study. SNP was assumed to be in LD with QTL in close proximity and the effect evaluated was additive effect (QTL allele substitution effect). The number of significant SNP at a threshold of P<0.001 was 3, 5, 5 and 4 loci for live weight at 6, 12, 18 and 24 months, respectively. For live weight at different ages, significant SNP were spread out across chromosome but some of significant SNP (rs29012453 and rs29012456 on BTA24) had shown highly significant effects. As for the distribution of size of SNP effects, few loci for live weight at different age had moderate effects (6~11%) but most of significant loci had small effects (2 to 5% of additive genetic variance) against total additive genetic variance. In conclusion, live weight at different age might be affected by few loci with moderate effect and many loci with small effects across genome in Hanwoo.

자돈의 생시 체중은 자돈의 이유 체중 및 생존율에도 크게 영향을 미친다. 또한 출하 시까지 돼지의 성장률 뿐만아니라 출하 일령 단축 및 출하 체중과도 밀접한 연관성이 있는 것으로 알려져 있다. 본 연구는 돼지 7번 염색체의 PGK 2(phosphoglycerate kinase 2) 유전자의 promoter 영역 및 transcription 영역에 해당하는 DNA 염기서열을 유전체 구조분석을 통해 20개의 SNP(Single Nucleotide Polymorpism)를 발굴하였고, 발굴된 SNP 중 PGK 2 유전자의 전사 조절 영역 내 g.122 T>G 다형을 재래돼지와 Landrace를 이용한 F2 집단 268두 자돈의 성장 형질과의 연관성 분석을 실시한 결과, 생시 체중(p<0.01) 및 3주령 체중(p<0.001)에서 통계적으로 고도의 유의적 연관성이 있음을 확인 할 수 있었다. 따라서 본 연구를 통하여 확인된 PGK 2 유전자의 promoter 영역 내 성장 형질 관련 SNP는 건강한 자돈 및 종돈을 조기 선발 하기 위한 유전자 marker로 활용 가능성을 제시하였다. The purpose of this study was to analyse of association between growth traits and single nucleotide polymorphisms(SNPs) polymorphism of phosphoglycerate kinase 2(PGK 2) gene in pigs. The birth weight of piglet influences on weaning weight and survival rate that are import economic traits in pig industry. Also, these growth traits are representative factor to decrease a period getting to marketing weight as well as growth rate in pig. The PGK 2 is an isozyme that catalyzes the first ATP-generating step in the glycolytic pathwayand important enzyme related with energy metabolism. Twenty of SNPs were discoveredby genome structure analysis that compares the sequence on promoter and transcription region of PGK 2 gene in porcine chromosome 7. An association between PGK 2 SNPs and growth traits was analyzed in F2 reciprocal-crossbred population between korean native pig(KNP) and Landrace. Association analysis indicated that polymorphism of the PGK 2 gene promoter region has significant effects on weight at birth(p<0.01) and weight at 3 weeks of age(p<0.0001). These results suggest that PGK 2 gene polymorphism was associated with energy metabolism and physiological function of growth in pig.
