Objectives: Hyaluronic acid, high molecular glycosaminoglycan, exists in extracellular matrix of tissue, especially, in skin and has been known to be deeply involved in skin hydration. In this study, we investigated the effect of methanol extract of Hwang-gi, Astragalus membranaceus root, on hyaluronic acid production in human keratinocyte HaCaT cells. Methods: We determined hyaluronic acid synthase 2 gene expression and hyaluronic acid production in HaCaT cells by using RT-PCR and ELISA, respectively. Results: Hwang-gi extract didn't show the toxicity to HaCaT cells within the treated concentration and increased the hyaluronic acid synthase 2 gene expression and hyaluronic acid production. Conclusions: Hyaluronic acid production increased by Hwang-gi could be, partially, contribute to the moisturing effect in skin by it.

Objectives: This study was designed to investigate the effects of herbal composition and fermented Cervi Pantotrichum Cornu on the growth of longitudinal bone in the adolescent male rats. Methods: Longitudinal bone growth was measured by fluorescence microscopy. To examine the effects on the growth plate metabolism, the total height of growth plate and the induction of local insulin-like growth factor-1 (IGF-1) and bone morphogenetic protein-2 (BMP-2) were measured. Results: The herbal composition treatment enhanced longitudinal bone growth and total heght of growth plate and promoted the induction of local IGF-1 and BMP-2 of growth plate. Conclusions: This study showed that the herbal composition have the effect of bone growth in adolescent rats and might be used for the growth delayed adolescent and inherent growth failure patient.

배양산삼 추출액의 Doxorubicin 독성 완화 효과

Objectives: This study was aimed to define the protective effect of Tissue Cultured Root of Wild Ginseng (CWG) against doxorubicin (Doxo) toxicity, and investigate the anti-tumor synergic effect of CWG in combination with Doxo in tumor-bearing C57BL/6 mice. Methods: Tumor-bearing mice were established by single inoculation with B16/F10 melanoma cells (2×10(6)/㎖) subcutaneously. Tumor-bearing mice (tumor volume between 50-100 ㎣) were selected and divided them into control, Doxo, and Doxo+CWG group. Mice of Doxo group were received with Doxo (4 ㎎/㎏ of B.W.) intraperitoneally at 0, 4, 8 days after starting the experiment. Mice of Doxo+CWG group were received CWG water extract during 12 days in combination with Doxo treatment. The body weight, tumor volume, tumor weight, and organ weight (heart, liver, kidney, and testis) were measured. And serum SPK, GOT and creatinine values were analysed. Results: The volume and weights of tumor masses in Doxo group were decreased significantly compared with the those of control group. And the those of Doxo+CWG group were not significantly different from the those of Doxo group. Whereas the weight of body, liver, kidney and testis in Doxo+CWG group were increased significantly compared with the those of Doxo group. The level of serum CPK and GOT in Doxo group were increased compared with the those of control group. But the value of Doxo+CWG group were decreased significantly compared with the values of Doxo group. Conclusions: These results suggest that CWG has protective effect against doxorubicin toxicity. And these effect is guessed that is caused in augmentation of vital energy.
Diabetic db/db Mice에 산약추출물(DA-9802) 투여에 따른 소장내의 Disaccharidases 활성에 미치는 영향

The present study was designed to clarify the antidiabetic activity and mechanism of Dioscorea rhizome in diabetic db/db mice. Mice were administered Dioscorea rhizome and rosiglitazone orally for 7 weeks and the effects of these compounds on fasting blood glucose, glucose tolerance and intestinal disaccharidase activity in db/db mice were evaluated. The fasting serum glucose of the D. rhizome treated group was reduced when compared with that of the db/db control group. In addition, the disaccharidase activities in homogenates of the proximal, middle and distal segment of the small intestine were significantly decreased response to D. rhizome treatment, especially in the middle segment. These results suggest that D. rhizome decreases blood glucose via a decrease in the activity of disaccharidase in the mucosa of the middle region of the small intestine in db/db mice.
무당벌레(Harmonia axyridis) 추출물에 의한 BV-2 세포주의 Nitric Oxide 생성 저해 활성

퇴행성 뇌질환은 뇌에 존재하는 면역세포인 소교세포의 염증반응이 발병 요인 중의 하나로 알려져 있다. 이에 본 연구는 초고속자동화시스템(high throughput screening: HTS)을 이용하여 약용곤충추출물로부터 항산화와 항염증 기능이 있다고 알려진 무당벌레 추출물로부터 염증발생인자인 nitric oxide의 생성에 어떠한 영향을 주는지 다음과 같은 결과를 얻었다. 소교세포인 BV-2세포에 대한 무당벌레 추출물의 세포독성은 물과 메탄올, DMSO 추출물에서는 100 ng/ml 까지는 거의 없었으나 에탄올 추출물은 1 ng/ml에서도 세포독성이 있었다. 물과 메탄을 추출물(50 ng/ml)은 LPS로 활성화된 BV-2세포에서 $TNF-{\alpha}$와 $IL-1{\beta}$의 발생을 35-60% 가량 억제 하였다. LPS로 유도된 NO의 생성은 물과 메탄올 추출물을 처리했을 때 각각 55%, 76% 억제되었다. 또한, MeOH 추출물을 처리했을 경우 LPS에 의한 iNOS 발현 정도를 단백질 수준과 mRNA 수준에서 현저하게 억제시킴을 확인하였다. Inflammation in the brain has known to be associated with the development of a various neurologiacal diseases. The hallmark of neuro-inflammation is the activation of microglia, brain macrophage. Pro-inflammatory compounds including nitric oxide(NO) are the main cause of neuro-degenerative disease such as Alzheimer's disease. In the study, we examined whether Harmonia axyridis extracts inhibit the NO production by a direct method using Griess reagent, western blotting and by RT-PCR(Reverse Transcription-Polymerase Chain Reactionin) the gene expression of inducible nitric oxide synthase(iNOS). Distilled water$(H_2O)$ and methanol(MeOH) extracts of H. axyridis inhibited the protein expression of TNF-a(Tumor Necrosis Factor) and IL-6(Interleukin) in LPS (Lipopolysaccharide) stimulated BV-2 cells at the concentration of 100 ng/ml. Incubation of BV-2 cells with the extracts of $H_2O$ of MeOH inhibited the LPS induced NO and iNOS protein. And this inhibition of iNOS protein is concordant with the inhibition of iNOS mRNA expression. These data suggested that H. axyridis extracts may play a crucial role in inhibiting the NO production.
