        Succession of bacterial community structure during the early stage of biofilm development in the Antarctic marine environment

        부유 세균의 군집과 구별되는 생물막내 세균 군집은 다양한 수생태계에서 중요한 생태학적 역할을 수행한다. 자연계에서 생물막이 생태학적으로 중요함에도 불구하고, 남극 해양 환경에서 생물막 형성 과정 동안의 세균 군집 구조와 그들의 변화에 대한 연구는 수행되지 않았다. 본 연구에서, 남극 해양 환경에서 생물막 형성 초기 단계에서의 세균 군집 구조 변화를 16S rRNA 유전자의 pyrosequencing을 통해 수행하였다. 생물막내 전반적인 세균 군집은 주변의 해수의 군집과 매우 달랐다. 전체 세균 군집의 78.8%에서 88.3%를 차지한 Gammaproteobacteria와 Bacteroidetes의 상대적 풍부도는 생물막의 형성에 따라 급격하게 변하였다. Gammaproteobacteria는 생물막 형성 진행에 따라 증가하다가 (4일째에 75.7%), 7일째에 46.1%로 감소하였다. 반면, Bacteroidetes는 초기에서 중기로 갈수록 감소하다가 다시 증가하는 양상을 보이며, Gammaproteobacteria와 반대의 변화 양상을 나타내었다. 생물막 형성의 초기 과정에 우점 하는 OTU (>1%)들의 변화 양상은 시기에 따라 뚜렷한 차이를 보였다. Gammaproteobacteria에 속하는 종의 경우, 4일째까지 증가한 반면, 첫째날 가장 우점 하였던 문인 Bacteroidetes에 속하는 종은 4일째까지 감소한 후, 다시 증가하는 양상을 보였다. 흥미롭게, Pseudoalteromonas prydzensis가 67.4%를 차지하며 우점 하였는데, 이는 생물막 형성에 이 종이 중요한 역할을 수행함을 시사하는 것으로 보인다. Compared to planktonic bacterial populations, biofilms have distinct bacterial community structures and play important ecological roles in various aquatic environments. Despite their ecological importance in nature, bacterial community structure and its succession during biofilm development in the Antarctic marine environment have not been elucidated. In this study, the succession of bacterial community, particularly during the early stage of biofilm development, in the Antarctic marine environment was investigated by pyrosequencing of the 16S rRNA gene. Overall bacterial distribution in biofilms differed considerably from surrounding seawater. Relative abundance of Gammaproteobacteria and Bacteroidetes which accounted for 78.9-88.3% of bacterial community changed drastically during biofilm succession. Gammaproteobacteria became more abundant with proceeding succession (75.7% on day 4) and decreased to 46.1% on day 7. The relative abundance of Bacteroidetes showed opposite trend to Gammaproteobacteria, decreasing from the early days to the intermediate days and becoming more abundant in the later days. There were striking differences in the composition of major OTUs (${\geq}1%$) among samples during the early stages of biofilm formation. Gammaproteobacterial species increased until day 4, while members of Bacteroidetes, the most dominant group on day 1, decreased until day 4 and then increased again. Interestingly, Pseudoalteromonas prydzensis was predominant, accounting for up to 67.4% of the biofilm bacterial community and indicating its important roles in the biofilm development.

        Purpose: This study was to evaluate the effects of foot reflexology on depression and anxiety in coal workers' pneumoconiosis. Method: The research design used was a non-equivalent control group pretest-posttest design. The foot reflexology was performed for 60 minutes twice a week through five weeks in the experimental group, not in the control group. The subjects of this study consisted of both the experimental group of twenty-nine and the control group of thirty coal workers' pneumoconiosis. The data were collected from December 10, 2002 to February 15, 2003. To evaluate the effects of foot reflexology the scores of depression and anxiety were measured before and after the experiment in both groups. Data of this experiment were analyzed by Chi-square test, t-test, unpaired t-test, ANCOVA and Repeated Measures ANOVA with the SAS Program. Results: The score of depression decreased in the experimental group, not in the control group. There was a significant difference of depression between the two groups. The score of anxiety decreased in the experimental group, not in the control group. There was a significant difference of anxiety between the two groups. Conclusion: As a summary of the study results, foot reflexology was regarded that it attributed to the emotional promotion of coal workers' pneumoconiosis. Therefore the foot reflexology is considered that might be used as an effective skill of nursing intervention program for coal workers' pneumoconiosis.

        마우스복강비만세포에서 프로테아제 발현 표현형의 조직 의존적 변화

        To examine the fate of the injected peritoneal mast cells (PMCs), we injected PMCs (500 or $10^5$) derived from WBB6F1-green fluorescent protein(GFP) mice into stomach wall of $WBB6F1-W/W^v$ mice. When 500 PMCs were injected, the proportion of alcian blue $(AB)^+$ mast cells to $GFP^+$ mast cells in the muscle was 25.0% on day 1, but decreased to 0.9% on day 7. Then, it increased to 98.2% on day 35. In contrast,$GFP^+$ mast cells in the mucosa were not detectable on day 1, 3, and 7 after injection. On day 35, the proportion of $AB^+$ mast cells to $GFP^+$ mast cells in the mucosa was 97.0%. When $10^5$ PMCs were injected, the proportion of $AB^+$ mast cells to $GFP^+$ mast cells in the muscle was more than 88.2%, and that in the mucosa was more than 86.3% from day 1 through 35 after injection. These results indicated that percentage of degranulation on day 1, 3, 7, 14 after injection of 500 PMCs was significantly higher than that after injection of $10^5$ PMCs. Futhermore, when 500 PMCs were injected, protease expression phenotypes of PMCs changed from day 14 after injection. When $10^5$ PMCs were injected, protease expression phenotype of PMCs did not change after injection. Such degranulated PMCs may acquire the new phenotype and adapt the new tissue.

        Diversity and Physiological Characteristics of Culturable Bacteria from Marine Sediments of Ross Sea, Antarctica

        남극 로스해의 퇴적물로부터 배양을 통해 분리한 균주의 분류 및 생리학적 특성 분석을 수행하였다. 분리 세균 63균주의 16S rRNA 유전자 염기서열을 이용한 계통분류학적 분석 결과, 이들은 Actinobacteria, Bacteroidetes, Alphaproteobacteria 및 Gammaproteobacteria 내의 21개의 파일로타입(phylotypes)에 속하였다. 98.65% 염기서열 유사도를 기준으로, 약 49%의 균주가 잠재적으로 신종 또는 신속 후보인 것으로 나타났다. 분리된 균주 중, 각각 46%, 25% 및 32%의 균주가 세포외 단백질분해효소, 지질분해효소 및 외부다당체 생성에 대한 활성을 나타냈다. 43개의 균주는 최소 1개의 세포외 분비 물질을 생산하였고, 이들 중 21개 균주는 최소 2개의 세포외 단백질분해효소, 지질분해효소 또는/및 세포외다당체를 생성하였다. 이러한 결과는 남극 로스해 퇴적물 내의 배양된 세균 군집이 해당 환경에서 탄소와 질소와 관련된 유기물질의 가수분해에 영향을 미치고 있다는 것을 시사한다. The affiliations and physiological characteristics of culturable bacteria isolated from the sediments of Ross Sea, Antarctica were investigated. Sixty-three isolates obtained by cultivation were grouped into 21 phylotypes affiliated with the phyla Actinobacteria and Bacteroidetes and with the classes Alphaproteobacteria and Gammaproteobacteria by phylogenetic analysis of 16S rRNA gene sequences. Based on phylogenetic analysis (<98.65% sequence similarity), approximately 49% of total isolates represented potentially novel species or genus. Among them, extracellular protease, lipase, and exopolysaccharide activities at $10^{\circ}C$ or $20^{\circ}C$ were detected in approximately 46%, 25%, and 32% of the strains, respectively. Forty-three isolates produced at least one type of extracellular material and 21 of them produced at least two extracellular protease, lipase, and/or exopolysaccharides. Our findings indicate that culturable bacterial diversity present within the marine sediments of Ross Sea, Antarctica may contribute to the hydrolysis of the major organic constituents which is closely related with carbon and nitrogen cycling in this environment.

        High-Resolution X-Ray Photoelectron Spectroscopy Study of a Sb<sub>2</sub>Te<sub>3</sub> Thin Film with the Polycrystalline Phase

        스퍼터를 이용하여 실리콘 기판위에 제작된 안티몬-테레니움 다결정 박막을 방사광을 이용한 고해상도 엑스선 광전자 분광법 실험을 수행하여 화학적 상태를 분석하였다. 엑스선 회절 실험을 통해 제작된 안티몬-테레니움 박막은 롬보헤드럴 구조를 가지는 다결정임을 확인하였다. 엑스선 광전자분광법을 수행하기 위하여 표면의 산화막 제거를 위해 저에너지 네온 이온 스퍼터링을 빔에너지 1 kV로 1 시간동안 수행하였고, 이를 통해 표면 산화막이 완벽히 제거됨을 확인하였다. 또한, 스퍼처링에 의하여 표면 비정질화된 상태를 결정화 상태로 만들기 위해 상변화온도인 $100^{\circ}C$에서 5 분간 초고진공상태에서 열처리를 수행하였다. 이후 획득되어진 테레니움 4d와 안티몬 4d 속전자레벨 분석에서 각각의 묶음에너지가 40.4 그리고 33.0 전자볼트임을 확인할 수 있었으며, 각각은 단일한 화학적 상태를 나타내고 얻어진 피크의 밀도분석을 통해 화학적조성비가 2 : 3임을 확인하였다. We investigated chemical states of a $Sb_2Te_3$ thin film with the polycrystalline phase by using high-resolution x-ray photoelectron spectroscopy with synchrotron radiation. The $Sb_2Te_3$ thin film was formed by sputtering. The rhombohedral phase was confirmed by x-ray diffraction. To remove the surface oxide, we performed $Ne^+$ ion sputtering for 1 hour with the beam energy of 1 kV and post-annealing at $100^{\circ}C$ for 5 min in ultra-high vacuum. We obtained the Te and Sb 4d core-levels spectra with the peaks at the binding energies of 40.4 and 33.0 eV, respectively. The full-width of half maximum of both the Te and Sb $4d_{5/2}$ core-levels is 0.9 eV. The Te and Sb core-levels only show a single chemical state, and we also confirmed the stoichiometry of approximately 2 : 3.
