Cowpea mild mottle virus (CPMMV) has a wide range of hosts, such as the pea family and tomato. CPMMV is a nonreported virus in Korea, and is domestically designated as a controlled virus associated with plant quarantine. In this study, a rapid diagnostic method for the detection of CPMMV at quarantine sites was developed. For the development of a userbased system, the PCR compositions and conditions use existing methods of quarantine for the viruses. Two sets of RTPCR and nested PCR were developed in this study that could be amplified from 579→298 dp and 638→252 bp, respectively. Furthermore, a sequence inserted positive control plasmid was developed, which is able to identify false-positives resulting from laboratory contamination. The findings of this study are important for the diagnosis of CPMMV in imported crops held in plant quarantine.

본 연구에서는 신분류군 탐색을 위하여 유입하수 첨가 배지를 제작하였다. 유입하수 첨가배지에서는 일반 배지에서 분리 또는 동정되지 않은 13속 세균들을 배양하였으며, 특히 25에 비해 37℃에서 다양한 균주들을 배양할 수 있었다. 또한 유입하수 첨가 배지에서 총 12개의 분리 균주들이 97%이하의 유사성을 나타내는 것으로 분석되었으며, 이들은 Tessaracoccus, Paracoccus 및 Candidimonas속(또는 Paralcaligenes속)과 유연관계가 있었다. In this study, we evaluated complex media with influent sewage water (ISW) to isolate novel taxa of bacteria. It was possible to cultivate 13 genera using the complex media with ISW. Additionally, more diverse genera were identified at 37°C than at 25°C, using the complex media with ISW. Total 12 strains of 179 bacterial isolates were shared less than 97% 16S rRNA gene sequence similarities with any known species. These isolates could be assigned to genera Tessaracoccus, Paracoccus, or Candidimonas (or Paralcaligenes).

최근 자동차 배출 가스 저감 등을 위한 수송 부문의 실제도로상의 CO2를 비롯한 유해 배출 가스 저감 기술 개발에 대한 많은 노력이 이루어지고 있다. 특히 도로 부문은 수송 부문의 약 96% 해당하는 온실가스 배출원(2019년 기준)으로서 절대적인 비중을 차지하며, 버스 및 택시등과 같은 공공 부문의 저탄소화 및 저공해와는 매우 중요한 배출저감 방안 중 하나이다. 그러나, 이러한 특수 목적용 차량들은 일반적인 자가용 차량 등과 주행 특성에 매우 다르며, 일반적인 연비 및 배기 인증모드 만으로는 택시 등의 주행 특성을 정확히 제시하는 것에 한계가 있을 수 있다. 이에 그간 택시와 같은 특수 목적 차량에 대한 주행 모드 개발 시도가 추진되어왔으며, 응용에 대한 필요성이 점차 확대되고 있는 상황이다. 이에 본 연구에서는 총 9대의 택시와 6대 가량의 일반차량과의 주행 특성을 통계를 바탕으로 비교 분석하였다. 택시의 주행 DB는 DTG(디지털 운행기록계)에 기록된 속도 특성을 기준으로 분석이 수행되었다. 취득 주행 DB는 주행 특성을 분석하기 위한 각종의 통계 분석이 수행되었으며, 현존하는 차량 주행 모드와의 특성 비교를 수행하여 적용 적합성에 대한 검토를 수행하였다. 한편으로 주행 DB와 통계분석 결과를 바탕으로 택시에 적합한 주행 모드의 개발을 수행하였으며, 실제 동력계에서의 모드 추종성 등의 검증을 수행하였다. 분석 결과 택시 주행은 일반적인 실험실 주행 모드와 매우 다른 특성을 제시하였으며, 가감속이 크며, 평균속도는 SC03 모드와 유사한 특성이 제시되었다.

본 연구는 언어 학습이 실행공동 체 (Communities of practice) 참여를 통한 사회적 학습과정이라는 전제하에, Norton (2001)의 상상 공동체 (Imagined communities) 개념을 바탕으로 언어학습자들의 다면적 동기를 이해하기 위한 목적으로 수행되었다. 특별히 기존 제2언어습득 문헌에서 상대적으로 연구되지 않은 미국 내 소수민족언어 (less commonly taught languages) 중 하나인 한국어 학습자의 다면적 동기를 조명하고자 하였다. 이를 위해 미국 동부 소재 대학에서 한국어 강의를 수강하는 11명의 학생들을 대상으로 심층적 인터뷰를 실시하였다. 연구 분석 결과 학습자 그룹 내에 관찰되는 상상공동체의 유형으로 K-pop 문화 공동체, 직업 공동체, 한국 친인척 공동체, 민족 정체성 공동체의 네가지 유형이 관찰되었다. 그리고 학습자 개개인의 상상 공동체는 언어 배경, 학습 기간, 매체 사용 등의 다양한 경험을 통해 다양하게 발전, 변화하는 것으로 나타났다. 특히 계승어 학습자와 비계승어 학습자 간에 지향하는 상상 공동체의 유형을 비교해 보았을 때 명확한 차이가 나타났다. 또한 기존 문헌에서 계승어 관련 동기 (heritage motivation or motivation related to heritage)라 명명되는 개념 내에도 학습자의 경험에 따라 실제로는 상이한 공동체 유형이 존재함이 발견되었다. (펜실배니아 대학교) The current study seeks to understand the multi-faceted desires of language learners through the theoretical lens of imagined communities (Norton, 2001). Particularly, the study focuses on the learners of Korean language―one of the less commonly taught languages in the U.S. that has received relatively less attention in previous literature on second language motivation. The study analyzed and compared the narratives told by eleven Korean language learners in a post-secondary language program, and identified four types of imagined communities: Communities of K-pop Culture, Communities of Professionals, Communities of Korean Family and Relatives, and Communities of ethnic Koreans. The study found that these imagined communities were not restricted to a specific region or an ethnic group but encompassed various populations connected through the use of Korean language. The study also found variability within what has been readily labelled as heritage motivation (or motivation related to heritage), as well as striking differences between heritage language learners and non-heritage language learners in terms of their scope of imagination. Peach rosette mosaic virus (PRMV)는 1933년 복숭아에서 처음 보고되었으며, 복숭아, 자두, 블루베리, 민들레, 벚나무 등에 감염되는 식물바이러스이다. PRMV는 한국에서 보고된 적이 없으나, 식물검역에서 관리병(control viruses)으로 지정되어 있다. 이번 연구에서는 PRMV를 더욱 신속하고 특이적으로 진단하기 위하여 Loop-mediated isothermal amplification 분석법을 적용한 진단법을 개발하였다. LAMP방법은 기존의 PCR 방법(RT-PCR 및 nested PCR)과 같은 검출 강도를 가지고 있다. 또한 LAMP 반응을 확인하기 위해 PRMV cDNA을 outer primer sets (Product size 264 bp)로 PCR 한 뒤, Pvu II (CAG/CTG) 제한효소를 처리하였다. 제한효소 처리 결과 2개의 digestion fragments (207 + 57 bp)가 확인되었다. PRMV의 LAMP 진단 방법은 관련 식물로부터 더욱 신속한 모니터링이 가능할 것으로 기대된다. Peach rosette mosaic virus (PRMV) is a plant virus that was first reported in 1933 by Peach. It can infect hosts including peach, grape, blueberry, dandelion, plum, cherry tree, and weeds. PRMV is non-reportable in Korea, but it is designated as a controlled virus requiring plant quarantine. In this study, for the rapid and specific detection of PRMV, we developed an assay using loop-mediated isothermal amplification (LAMP). Comparison between conventional polymerase chain reaction (PCR) methods (real time-PCR and nested PCR) and LAMP for the detection of PRMV revealed an equivalent level of sensitivity by all the tested methods. For the LAMP assay, outer primer sets were used to amplify a 264-bp PCR product, which was then digested using the restriction enzyme Pvu II (CAG/CTG), and the visualization of two digestion fragments (207 + 57 bp) indicated a positive reaction. The developed LAMP assay for PRMV is expected to enable the rapid monitoring of PRMV in plants.
