        The Efficiency of Deer Antler Herbal Acupuncture on Modulation and Prevention of IL-1 Mediated Activation in Rat Chondrocytes at a Receptor Level

        Objectives : Deer antler Herbal-Acupuncture (DHA) solution represents one of the most commonly used medicine to treat rheumatoid arthritis. But, mechanisms of its antiarthritic activities are still poorly understood. Identification of common DHA aqua-acupuncture capable of affording protection or modulating the onset and severity of arthritis may have important human health implications.Results : We determined if DHA could prevent the binding of IL-1β to its cellular receptors. DHA addition to rat chondrocytes treated with IL-1β or with reactive oxygen species(ROS) prevents the activation of proteoglycan synthesis. After treatment with IL-1β, DHA increased the expression of mRNA encoding the type II IL-1β receptor. These results emphasize the potential role of two regulating proteins of the IL-1β signaling pathway that could account for the beneficial effect of DHA in osteRArthritis. The present study also identifies a novel mechanism of DHA-mediated anti-inflammatory activity. Conclusion : It is shown that DHA inhibits both IL-1β- and TNF-α-induced NO production in normal human articular chondrocytes. The observed suppression of IL-1-induced NO production is associated with inhibition of inducible NO synthase(iNOS) mRNA and protein expression. In addition, DHA also suppresses the production of IL-1-induced cyclooxygenase-2 and IL-6. The constitutively expressed cyclooxygenase-1, however, was not affected by the sugar. These results demonstrate that DHA expresses a unique range of activities and identifies a novel mechanism for the inhibition of inflammatory processes.

        Effects of deer antler Water Extract (pilose antler of Cervus Korean TEMMINCK var. mantchuricus Sinhoe) on chondrocytes

        전통적인 면역억제제와 면역활동제인 녹용수용추출액(DAA)은 뼈의 재생에 있어서 중요한 역할을 한다고 여겨진다. 녹용약침이 비활성 연골세포의 분화를 야기시킬수 있는 지를 결정하기 위해, 융합된 세포 배양균을 녹용약침과 함께 24, 36, 48, 72, 120 시간동안 먼저 처리하였다. 이러한 처리후에 배양액을 10-10 ~ 10-8M 1,25-(OH)2D3를 포함하는 새로운 배양액으로 바꾸어 세포들을 추가로 24시간동안 배양했다. 앞선 연구에서 더 성숙된 활성연골세포가 이러한 Vit D3 대사산물에 반응한다는 것을 보여주었기 때문에 이러한 두 번째 처리를 선택하였다. 세포성숙에 있어서 녹용약침의 전처리 효과는 ALPase의 특이활동을 측정하는 것으로 확인하였다. 기질 단백질 합성의 변화는 35SO4 결합이 proteoglycan으로의 변화되는것과 collagen 합성을 측정함으로써 증명되었다. 비활성 연골세포가 녹용약침과 함께 120시간동안 전처리되고 다시 1,25-(OH)2D3를 추가한 처치에서 ALPase 특이활동과 collagen 합성이 농도 의존적으로 증가를 일으켰다. 그러나 proteoglycan의 생성에는 영향을 미치지 않았다. 1,25-(OH)2D3와 함께 전처리 된 비활성 연골 세포는 어떠한 전처리도 받지 않았던 비활성 연골세포처럼 반응했다. 이러한 결과는 녹용약침이 직접적으로 비활성 연골 세포를 활성 연골세포로 성숙시키는데 관여한다는 것을 알려준다. 그러므로 녹용약침은 연골내 골화과정에서 연골세포의 성숙을 조절하는데 중요한 역할을 하는 것으로 나타났다.

        Object : In this thesis we researched the transformation of five symptoms of final life stage from ${\ll}$Sumun(素問)${\gg}$, ${\ll}$Yingshu(靈樞)${\gg}$ and ${\ll}$Nanjing(難經)${\gg}$ and pulse diagnosis method. Method : We studied medical books from Mawangdui Han dynasty tomb Results and Conclusion : 1. The symptoms of final life stage according to Yinyanmaisihou(陰陽脈死候) is suggested by observation of symptoms introducting Yinyan five-elements theory. 2. The symptoms of final life stage according to Yinyanmaisihou(陰陽脈死候) and "Qijue(氣絶)" from ${\ll}$Lingshu(靈樞) Jingmai(經脈)${\gg}$ made use of twelve merdian bian(遍) diagnosis method and ${\ll}$Lingshu(靈樞) Zongshi(終始)${\gg}$, ${\ll}$Sumun(素問) Zhenyaojingzhonglun(診要經終論)${\gg}$ made use of twelve merdian biaoben(標本) diagnosis method. Afterward oriental medical biology which suggests agreement between outside and inside symptoms needs to be examined by use of science and medical theory to apply medical treatment .

        Suppressive effects of a water extract of Ulmus davidiana Planch (Ulmaceae) on collagen-induced arthritis in mice

        참느릅나무(UD)의 껍질(유근피) 추출액이 생쥐에서 유발될 수 있는 염증성 사이토카인의 발현을 조절할 수 있는지를 관찰하였으며 또한 생쥐의 Collagen 유발 관절염(collagen-induced arthritis:CIA)에서 유근피 추출액의 치료효과를 평가하기 위하여 족척과 무릎관절을 형태학적, 조직학적으로 관찰하였다. DBA/1 생쥐를 bovine type Ⅱ collagen(소의 제2형 collagen)에 감작되게 하였다. 제2형 collagen에 감작되게 한 이후, 유근피 추출액 100mg/kg를 3주 동안 하루에 한번 경구투여 하였다. 족척은 발적, 부종, 변형을 형태학적으로 관찰하였으며, 발목에서 TNF-α, IL-1β의 수준을 조사하였다. 무릎관절의 관절염에 의한 손상정도는 연골파괴와 pannus 형성의 조직학적 관찰에 의해 평가되었다. 유근피 추출액의 투여는 CIA의 진행을 현저하게 억제시켰고, 족척에서의 TNF-α, IL-1β의 생성을 억제하였다. 연골의 파괴는 유근피 추출액 투여 이후 생쥐의 무릎에서 현저하게 감소하였다. 유근피 추출액을 투여한 생쥐의 혈청에서는 IL-4, IL-10, 항염증성 사이토카인의 수준이 증가하였다. 이러한 결과로부터 유근피 추출액의 투여는, 연골의 보호를 포함하는, collagen 유발 관절염에 대해서 치료효과가 있고, 류마티스 관절염에 대하여 잠정적인 치료효과를 가진다는 결론을 내릴 수 있다.

        Inhibitory Action of Ulmus Davidiana Planch Extract Solution to Osteoclast Cell Proliferation and Prostaglandin E2 Synthesis in Mice

        Objectives : Ulmus davidiana Planch (UD) has long been known to have anti-inflammatory and protective effects on damaged tissue, inflammation and bone among other functions.Methods : This study was undertaken to address whether the water extract of the bark of UD could modulate proliferation of mouse osteoclasts in vitro and to investigate its effect on cyclooxygenase-2 (COX-2), which converts arachidonic acid to prostaglandin E2 (PGE2) and is highly expressed in osteoclasts. Mouse osteoclasts were tested in vitro for growth inhibition, proliferation cell nuclear antigen expression, and COX-2 activity and expression after treatment with UD extract. Results : Its effects were compared with those of indomethacin (a nonselective COX inhibitor) and celecoxib (a selective COX-2 inhibitor) by Cell viability assay, Cell cycle analysis, Immunohistochemical analysis of PCNA expression, Western blot analysis and PGE2 Enzyme immunoassay (EIA). UD demonstrated a strong growth inhibitory action in both tested osteoclasts cells. The IC50s were 10 μg/ml for UD, 6 μM for celecoxib and 42 μM for indomethacin. UD, as well as celecoxib and indomethacin, suppressed proliferation cell nuclear antigen expression and PGE2 synthesis in osteoclasts. UD inhibited COX-2 expression, whereas celecoxib inhibited COX-2 activity directly. Conclusion : UD selectively and effectively inhibits osteoclasts cell growth in vitro. Inhibitory action of PGE2 synthesis via suppression of COX-2 expression may be responsible for its anti-inflammatory activity.

        Prevention of Collagen-induced Arthritis in Mice by Deer Antler Extract(DAE)

        Objectives : The effect of water extract of the pilose antler of Cervus korean TEMMINCK var. mantchuricus Swinhoe (Nokyong), a traditional immuno-suppressive and immuno-activating Korean oriental medicine, on collagen-induced arthritis (CIA) mice model was studied. Identification of common Nokyong capable of affording protection or modulating the onset and severity of arthritis may have important human health implications. Methods : Nokyong has shown to possess anti-inflammatory and anticarcinogenic properties in experimental animals. In this study we determined the effect of DAE on collagen-induced arthritis in mice. Results : In three independent experiments mice given DAE in water exhibited significantly reduced incidence of arthritis (33% to 50%) as compared with mice given no DAE in water (84% to 100%). The arthritis index also was significantly lower in DAE-fed animals. Western blot analysis showed a marked reduction in the expression of inflammatory mediators such as cyclooxygenase 2 (Cox-2), Interferon-γ (INF-γ), and tumor necrosis factor α (TNF-α) in arthritic joints of DAE-fed mice. The neutral endopeptidase (NEP) activity was approximately 6-fold higher in arthritic joints of non-DAE-fed mice in comparison to nonarthritic joints of nonimmunized mice whereas it was only 2-fold higher in the arthritic joints of DAE-fed mice. Additionally, total IgG and type II collagen-specific IgG levels were lower in the arthritic joints of DAE-fed mice. Conclusion : Taken together our studies suggest that DAE may be useful in the prevention of onset and severity of arthritis.

        침자 술기 실습 모델에서 침감의 정량적 특성에 관한 연구

        Objectives : In this study, we quantitatively evaluated the needle forces using needle insertion-measurement system and compared the needle sensation of each acupuncture practical model. Methods : After inserting acupuncture with a sensor to six models, a lifting-thrusting motion was implemented using the needle insertion-measurement system. The needle force was measured repeatedly, and the measurement was analyzed based on the modified Karnopp friction model for a comparison of friction coefficients. After the insertion, practitioners did lifting-thrusting manipulations. They quantified the similarity of needle sensation with VAS (Visual Analogue Scale). Results : When friction force and coefficients of friction in five different models were compared with a porcine shank model, all five models were significantly different from a porcine shank model, cotton and apple showing the closest frictional values to that of a porcine shank model. In the Cp and Cn values of cotton and in the Cp values of IM injection pad, there was no statistically significant difference. The similarity of the needle sensation between the porcine shank and five models was the highest in the apple, and overlapping papers was the lowest. Conclusions : This study quantitatively compared the physical forces in the practical model when implementing lifting-thrusting manipulations, using a needle insertion-measurement system. We suggest that a reproducible exercise model that reflects the characteristics of various human tissues, such as viscoelasticity or strength, needs to be further developed. This will contribute to establishing standardized acupuncture practice training.

        Effect of Ulmus davidiana Planch herbal acupuncture Solution on proinflammatory cytokine IL-1β and TNF-α production in collagen-induced arthritis of rats

        여러 기능들 중에서 손상된 조직, 염증과 뼈에 대한 항염증 및 보호 효과로 오랫동안 알려진 유근피의 효과는 쥐에서 관절염을 야기하는 type Ⅱ 콜라겐(CIA)의 발달을 통해 연구되었다. 수컷 쥐들은 CⅡ와 완전한 CFA 200 ㎍ 유화액에 면역되었다. 그 뒤 실험하는 동안 쥐들에게 유근피 약침 용액(UDHA) 또는 생리식염수를 복강내에 주입하였다. 림프절 세포들(LN cells)은 면역 후 14일 째에 쥐로부터 얻어졌고, CⅡ와 함께 시험관 안에서 배양되었다. 대조군인 생리식염수를 주입한 쥐들과 비교했을 때, 14일 동안 이틀마다 한번씩 20 ㎍/100 g rat 보다 더 많은 양의 UDHA는 T 세포 사이토카인 IL-1β와 TNF-α를 만드는 서혜부 림프절 세포의 능력을 억제한다. 쥐에게 양의 적혈구(SRBC)를 복강내에 주입했을 때, UDHA 처치된 쥐와 대조군 쥐들에서 혈구응집반응 수치는 적은 양의 UDHA를 주었을 때와 유의한 차이가 없었다. 그러나 7일동안 매일 10 ㎍/100 g보다 많은 양의 복강내 투여는 점차 항체 생성을 억제하였다. 결론적으로 치료적 UDHA의 복강내 주입은 CⅡ에 대한 질병과 면역 반응의 임상적 과정에 영향을 준다.
