This experiment was designed to observe the early pathologic changes induced by local Co-60 γ-ray irradiation in the lung and to investigate the mechanism of that changes. Co-60 γ-ray(500, 750 and 1,000 rad) was locally irradiated to the right lung of Sprague Dawley rats. After irradiation, the rat lungs were isolated at intervals of 12 hours, 1 day, 3 days, 1 week, 2 weeks, and 3 weeks, respectively. Histological(hematoxylin-eosin, Masson's trichrome, van-Gieson, and reticulin stain) and enzyme histochemical(acid phosphatase, alkaline phosphatase, and nonspecific estease stain) findings were observed and the foolowing results were obtained: 1. After Co-60 γ-ray irradiation, alveolar hemorrhage, exudation and perivascular infiltration of inflammatory cells were developed more rapidly in proportion to the dose of irradiated Co-60 γ-ray at early stage. 2. After Co-60 γ-ray irradiation, the number of alveolar macrophasges in the alveolar wall and histiocytes in the alveolar septum were decreased at early stage, but it showed some evidences of restoration after 1 week. 3. One week after irradiation, denudation of the cells of the alveolar wall was appeared. The majority of which were alveolar macrophages. And these cells were appeared as abnormal giant cells in the large dose irradiated droup. 4. In the large dose irradiated group, some of the bronchial epithelial cells were changed to squamous epithelium from 1 week after irradiation. 5. In the enzyme histochemical findings, acid phosphatase, alkaline phosphatase and nonspecific esterase showed little difference in the irradiated dose or the duration of irradiation. These obervations permit the conclusion that the mechanism of early pathologic changes in the lung induced by irradiation is an inflammatory changes due to increased vascular permeability, and the degree of inflammatory changes is proportional to the irradiated doses. And the cell damage induced by irradiation is directly proportional to the degree of differentiation.
