        해저열수광상 주변 압석 시료의 공극률과 전기비저항 측정 기초실험

        해양전자탐사를 이용한 해저열수광상 탐사에 기초적인 물성 지식을 제공하고자 해저열수광상 주변에서 채취한 암석시료를 원주형 암석코어로 성형한 후 밀도, 공극률 둥에 대한 불성 측정법과 그 결과를 제시하였다 암석 시료는 해저열수광상 주변에서 채취한 모암, 부석, 열수변질대, 침니(chimney) 등을 대상으로 하였으며, 이들은 부스러지기 쉽거나 물 속에서 구성물질이 용해되거나 혹은 공극이 매우 크고 거친 표면 특정을 가지고 있다. 본 연구에서는 이러한 시료들의 특정을 고려하여 각 시료마다 공극률과 전가비저항을 측정하는 방법들은 제시하였다. 측정 결과 전71전도도가 32,000uS/cm(0.5 ohm-m)인 소금물에 수포화된 흑색모암, 회색모암, 부석, 침니 암석코어들의 현지 바닷물 온도(2.5°C)에서의 전기비저항은 각각 102,39, 11,0.1 ohm-m로서 침니의 전기비저항을 l이라할때 바닷물은 5, 부석은 110, 모암은 390~1020 의 분포를 보였다. 또한, 실험 결과 물로 수포화된 암석코어는 온도가 증가할수록 전기비지향이 감소하고 특히, 20~80°C 의 구간에서는 직선적으로 감소하며, 임의의 전기전도도를 갖는 간극수로 수포화시킨 암석코어의 전기비저항이 다른 전기전도도를 갖는 간극수로 수포화시킨 암석코어의 전기비저항에 비해 갚은 용도에서 A배 증가하면 단위 온도 증가당 전기비저항 연효율도 A배 증가함을 알 수 있었다. We present the results of laboratory measurement of porosity and electrical resistivity for the samples collected near marine hydrothermal deposit to provide fundamental perspective of physical properties for future electromagnetic survey. The rock cores are sampled from the host rock, pumice, hydrothermal altered zone, and chimney. These samples are featured as easily brittle, rough surface with large pores. having components easily solvable in the water. We suggest systematic approach for measuring weights, volumes of core samples to calculate density and porosity. Measurements reveal that the resistivities of black host rock, gray host rock, pumice and chimney are 102, 39, It, 0.1 ohrn-m, respectively, when the core samples are saturated with saline water of 32,000 pS/cm (0.5 ohm-m) at temperature of 2.5°C and these correspond to the factors of 5 for sea water, 110 for pumice and 390~1020 for host rocks with respect to the resistivity of chimney. We also confirm that resistivity of rock samples saturated with water decrease with temperature linearly over the temperature range of 20~80°C.

        새로운 생명공학작물 안전성 평가를 위한 작물 성분 DB 구축

        국립농업과학원 생물안전성과는 생명공학기술로 개발되는 작물(GM작물)의 안전성 평가 중 '성분분석에 의한 비교평가'에 참고자료로 활용할 수 있도록 기존 상업화 작물성분의 분석 데이터를 제공하는 "작물성분 DB"를 구축하였다. "작물성분 DB"는 우리나라의 자연 환경에서 재배되는 주요 작물 품종들의 영양성분 함량정보를 제공함으로써 품종과 재배 지역, 연도에 따라 함량의 변화 정도를 확인할 수 있도록 데이터를 업그레이드 하고 있다. 현재 2곳 이상의 재배지역에서 2년 이상 재배한 벼와 고추 시료에 대한 주요 영양분과 항영양소, 2차 대사산물을 검증된 분석 방법을 사용하여 분석한 데이트를 제공하고 있다. 데이터는 평균값과 최저, 최대값을 제공함으로써 GM작물의 안전성 평가시 GM작물과 대응 비교작물 간에 나타나는 통계적 차이가 기존 상업작물의 생물학적 차이 또는 허용범위 내에 속하는지를 평가할 수 있도록 하였다. "작물성분 DB"는 사용자가 선택한 쿼리를 기반으로 쉽게 검색하고 활용하도록 하고 있다. 또한 유색미와 감자, 고구마, 콜리플라워와 같은 유색 작물의 기능성 성분 함량 정보를 제공하고 있어 일반 소비자들도 유용하게 정보를 활용할 수 있다. 본 논문은 상업작물 성분에 대한 중요한 정보를 제공하는 농진청 "작물성분 DB"의 구성과 사용법을 소개하였다. The Biosafety Division of the National Academy of Agricultural Science has developed a 'Crop Composition DB' that provides analytical data on commercialized crops. It can be used as a reference in the 'Comparative Evaluation by Compositional Analysis' for the safety assessment of genetically modified (GM) crops. This database provides the composition of crops cultivated in Korea, and thus upgrades the data to check the extent of changes in the compositional content depending on the cultivated area, varieties and year. The database is a compilation of data on the antioxidant, nutrient and secondary metabolite compositions of rice and capsicum grown in two or more cultivation areas for a period of more than two years. Data analysis was conducted under the guidelines of the Association of Official Analytical Chemists or methods previously reported on papers. The data was provided as average, minimum and maximum values to assess whether the statistical differences between the GM crops and comparative non-GM crops fall within the biological differences or tolerances of the existing commercial crops. The Crop Composition DB is an open-access source and is easy to access based on the query selected by the user. Moreover, functional ingredients of colored crops, such as potatoes, sweet potatoes and cauliflowers, were provided so that food information can be used and utilized by general consumers. This paper introduces the feature and usage of 'Crop Composition DB', which is a valuable tool for characterizing the composition of conventional crops.

        유전자변형작물 안전성평가를 위한 영양성분 비교연구 동향

        Approvals for cultivation and import of genetically modified (GM) crops have dramatically increased around the world. Comparative compositional studies are an important aspect of safety assessments of products from GM crops and are based on substantial equivalence. Compositional analyses focus on determining similarities and differences between the compositions of the GM crops and their conventional counterparts, and thereby assessing the compositional equivalence of GM crops and their conventional comparators. The analytes, such as major constituents, key nutrients, and antinutrients, are generally determined on a crop-specific basis according to the OECD consensus document. The use of standard methods throughout the processes, such as selection of comparators, field trials, analytical methods, and statistical data analysis, is crucial. In this study, we showed the general framework of compositional studies. Literature for compositional studies of GM crops conducted abroad and in Korea was reviewed to obtain information about analytes, conventional counterparts, cultivation year, location, and statistical methods. The studies conducted abroad assessed for commercial release of GM crops such as soybean, maize, and cotton, while domestic studies were mainly performed for research in rice. In addition, we suggested a guidance for conventional comparators and field trials applicable to the domestic situation. 전세계적으로 유전자변형(GM)작물의 재배와 수입에 대한 승인이 급격히 증가하고 있다. 영양성분 비교평가는 실질적 동등성에 기반하여 GM작물의 안전성 평가에서 중요한 역할을 담당하고 있다. 영양성분 비교평가는 GM작물과 대조작물 사이의 성분함량에서 차이와 유사성을, 그리고 GM작물과 상업품종간의 성분함량의 동등성을 결정하는데 중점을 두고 있다. 분석항목은 일반성분, 주요 영양성분과 항영양소 등이며 일반적으로 OECD 합의문에서 작물별로 제안하고 있는 항목들을 포함한다. 영양성분 비교평가를 위해서는 비교군의 선정, 포장 시험, 분석방법, 데이터 통계 처리 등의 전 과정에 걸쳐 표준화된 방법을 이용하는 것이 중요하다. 본 연구에서는 코덱스와 유럽식품안전청의 식품용 GM작물의 영양성분 비교평가를 위한 가이드라인을 소개하였다. 그리고 국내외의 GM작물 영양성분 비교평가 연구 논문들에 나타난 분석항목과 대조작물, 참조군 품종, 재배연도, 재배지역, 통계방법 등을 검토하였다. 해외의 경우 대두와 옥수수, 면화 GM작물에서 여러 나라 수출을 위한 규제기관의 가이드라인에 따른 연구가 많았다. 반면 국내의 경우는 GM작물 실험연구적 측면에서 비교평가 진행되었으며 주로 GM벼에서의 연구가 많았다. 마지막으로 국내에서 GM작물의 영양성분 비교평가 연구 수행에 적용할 수 있는 참조군 선정과 포장재배에 대한 가이드를 제시하였다.

        LPS로 유도한 RAW 264.7 세포의 염증반응에서 흰민들레의 항염증 효과

        Objectives : Taraxacum coreanum (TC) have been used as a traditional medicine to treat inflammatory diseases and anti-oxidant effect in Korea. However, the anti-inflammatory effect of TC water extract on lipopolysaccharide (LPS)-induced inflammation is not well-known. Therefore, this study was performed to identify the anti-inflammatory effect of TC on LPS induced inflammatory. Methods : RAW 264.7 cells were treated with 500 ng/mL of LPS. Water extracts of TC (0.1, 0.25, 0.5 mg/ml) was treated 1 h prior to LPS. Cell viability was measured by MTT assay. Levels of nitric oxide (NO) were measured with Griess reagent and pro-inflammatory cytokines were measured by enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (real-time PCR). We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B (NF-κB) activation by western blot. Results : Water Extract from TC itself did not have any cytotoxic effect in RAW 264.7 cells. TC treatment inhibited the production of NO production, and pro-inflamamtory cytokines such as interleukin (IL)-6 and IL-1β on protein and mRNA levels. In addition, TC treatment inhibited the LPS-induced activation of MAPKs such as extracellular signal-regulated kinase1/2 (ERK1/2), p38 kinases (p38), c-Jun NH2-terminal kinase (JNK) and NF-κB. Conclusions : In summary, our result suggest that treatment of TC could reduce the LPS-induced inflammation. Thereby, TC could be used as a protective agent against inflammation. Also, this study could give a clinical basis that TC could be a drug or agent to prevent inflammation.

        몰약(沒藥) 물 추출물의 급성 췌장염 보호 효과

        Objectives : Commiphora myrrha (CM) has been used in traditional medicine for treating disease such as obesity, hyperlipidemia, atherosclerosis, diabetes and osteoarthritis. However, the protective effects of CM on acute pancreatitis (AP) has not been reported. Thus, the aim of this study was to evaluate the protective effects of CM water extract on cerulein-induced AP. Methods : AP was induced in mice via intraperitoneal injection of supramaximal concentrations of the stable cholecystokinin analogue cerulein (50 μg/kg) every hour for 6 times. Water extract of CM (0.1, 0.2, or 0.5 g/kg) was administrated intraperitoneally 1 h prior to the first injection of cerulein. The mice were killed at 6 h after the final cerulein injection. Pancreas was rapidly removed for morphologic and histochemical examination, myeloperoxidase (MPO) assay. Blood samples were taken to determine serum amylase and lipase activities. Results : Administration of CM significantly inhibited pancreatic weight/body weight ratio, pancreas histological injury. And CM administration inhibited the serum digestive enzyme elevation such as amylase and lipase on cerulein-induced pancreatitis. In addition, Pancreas MPO activity which indicates neutrophil infiltration was inhibited by CM extract on cerulein-induced pancreatitis. Conclusions : In conclusion, our results could suggest that pre-treatment of CM reduces the severity of cerulein-induced AP. Therefore, CM could be used as a protective agent against AP. Also, this study could give a clinical basis that CM could be a drug or agent to prevent AP.

        마치현(馬齒□)의 급성 췌장염 보호 효과

        Objective : Portulaca oleracea (PO) has been used as an important traditional medicine for inflammatory and bacterial diseases in East Asia. However, the protective effects of PO on acute pancreatitis (AP) is not well-known. Therefore, this study was performed to identify the anti-inflammatory and prophylactic effects of PO on cerulein-induced AP. Methods : AP was induced in mice via intraperitoneal injection of supramaximal concentrations of the stable cholecystokinin analogue cerulein (50 ㎍/㎏) given every hour for 6 times. Water extracts of PO (100, 300, or 500 ㎎/㎏) was administrated intra-peritoneally 1 h prior to the first injection of cerulein. The mice were killed at 6 h after the final cerulein injection. Pancreas and lung were rapidly removed for morphologic and histochemical examination, myeloperoxidase (MPO) assay. Blood samples were taken to determine serum amylase and lipase activities. Results : Administration of PO significantly inhibited pancreatic weight/body weight ratio, pancreas andlung histological injury. And MPO activity which indicates neutrophil infiltration was inhibited by PO extracts on cerulein-induced pancreatitis. In addition, PO administration inhibited digestive enzymes such as serum amylase and lipase activity on cerulein-induced pancreatitis.         Microarray 를 이용한 유의한 유전자 탐색

        Microarrays are novel biotechnologies which are being used increasingly in medical research. By allowing the monitoring of expression levels for thousands of genes simultaneously, such a microarray technique may lead to a more complete understanding of the molecular variations and may detect differentially expressed genes. However, except for some basic classification and clustering analyses, and testing procedures, not many sophisticated statistical analysis methods have been developed. In this paper, we consider a regression model approach to detect a differentially expressed genes in a single microarray and compare it with other approaches.
